RESUMO

Fatal anaphylactic shock is common in forensic practice. However, it is difficult to diagnose for lacking specific pathological and morphologic changes in forensic autopsy. The application of some biochemical indicators is of great significance. This paper reviews the biological characteristics of some biochemical indicators and detection methods. The forensic application, problems and prospects of these indicators are also introduced in details. The stable biochemical indicators, IgE, tryptase and chymase, show great potential and advantages in the identification of fatal anaphylactic shock in forensic medicine.

RESUMO

OBJECTIVE@#To explore the value of flow cytometry in anaphylactic shock diagnosis by CD63 expression being detected using flow cytometry to conform the activation of basophils.@*METHODS@#Sixteen rats were randomly divided into two groups: control group and anaphylactic shock group. The model of anaphylactic shock rat with ovalbumin injection was established. CD63, CD45 and CD203c antibody combination, flow cytometry was employed to detected blood basophil CD63 expression. Immunofluorescence method was employed to observe the CD63 immunofluorescence staining in the rat lung tissue.@*RESULTS@#(1) Pure basophils were obtained by CD45 and CD203c gating. (2) The percentages of basophils CD63 were (17.34 +/- 2.04)% and (1.52 +/- 0.35)% in the experimental and control group, respectively. The differences between two groups were statistically significant (P < 0.01). (3) Compared with the control group, the expression of CD63 in basophils increased in anaphylactic shock lung tissue.@*CONCLUSION@#The detection of CD63 by flow cytometry could be the supplement of vivo allergic reactions and have good clinical value.

RESUMO

OBJECTIVE@#To seek the pathomorphological targets for forensic expertise in anaphylactic shock.@*METHODS@#The expression of IgE in hearts, lungs, livers, spleens, kidneys, gastrics, intestinals, tracheas and tonsils of anaphylactic shock guinea-pigs was observed at 0, 6, 12 h and 24 h respectively by tissue chip S-P immuno-histochemical method.@*RESULTS@#Positive expression of IgE presented in lungs and tracheas in the test group with the peak at 0 hour and it declined as time advanced, and also there were significant differences at different times (P<0.05).@*CONCLUSION@#The immuno-histochemical method of detecting the expression of IgE in lungs, tracheas and spleens can be supposed to be the pathomorphological targets for forensic expertise in anaphylactic shock. The weakening of the positive expression of IgE in lungs and tracheas as the time advanced suggested that in this kind of case the autopsy should be arried out as early as possible.

RESUMO

Relative amount of histamine and SRS-A released during Schultz Dale reaction were indirectly estimated. Percent block produced by specific antagonists, mepyramine and FPL 55712, on antigen induced response in various tissues was compared with controls. The relative bioactivity of histamine and SRS-A released was 93% and 7%, respectively, in rat lungs: 37% and 63% in guinea pig lungs; 82% and 18% in guinea pig intestine, 59% and 41% in rat intestine and 100% and 0% in skin of guinea pig and rats. Expectedly, individual experiments showed gross variations because anaphylaxis is rarely dose related.
