ABSTRACT Pro-inflammatory cytokines and glial cells, especially microglial cells, have been implicated in persistent pain sensitization. Less is known about the role of astrocytes in pain regulation. This study aimed to observe the expression of the astrocytic biomarker glial fibrillary acidic protein (GFAP) and the serum levels of interleukin 1 beta (IL-1ß) and tumor necrosis factor alpha (TNF-α) after short-term administration of central pain relievers in rats not submitted to noxious stimuli. Male Wistar rats were divided into five groups, receiving for nine days- (1) amitriptyline (Amt-10 mg/kg/day, by gavage); (2) gabapentin (Gb-60 mg/kg/day, by gavage; (3) methadone (Me-4.5 mg/kg/day, intraperitoneal route [IP]); (4) morphine (Mo-10 mg/kg/day, IP); or (5) 0.9% saline solution, IP. Brain samples were collected for immunohistochemical study of GFAP expression in the mesencephalon and nucleus accumbens (NAc). The area of GFAP-positive cells was calculated using MetaMorph software and serum levels of IL-1ß and TNF-α were measured by enzyme-linked immunosorbent assay. Serum TNF-α levels were decreased in the groups treated with Mo, Me and Gb, but not in the Amt-treated group. IL-1ß decreased only in rats treated with Me. The astrocytic expression of GFAP was decreased in the brainstem with all drugs, while it was increased in the NAc with Amt, Me and Mo

INTRODUÇÃO: O absenteísmo-doença, enquanto falta ao trabalho justificada por licença médica, é um importante indicador das condições de saúde dos trabalhadores. Em geral, características sociodemográficas e ocupacionais situam-se entre os principais fatores associados ao absenteísmo-doença. A administração pública é responsável por 21,8% dos empregos formais no Brasil. Esta população permite o estudo de uma grande variedade de categorias profissionais. OBJETIVO: Analisar o perfil e os indicadores de absenteísmo-doença entre servidores municipais de Goiânia, no Estado de Goiás, Brasil. Métodos: Estudo transversal das licenças certificadas para tratamento de saúde superiores a três dias, de todos os servidores, desde janeiro de 2005 a dezembro de 2010. Foram calculadas as prevalências, utilizando como critérios o número de indivíduos, os episódios e os dias de afastamento. RESULTADOS: Foram concedidas 40.578 licenças certificadas para tratamento de saúde a 13.408 servidores numa população média anual de 17.270 pessoas, o que resultou em 944.722 dias de absenteísmo. A prevalência acumulada de licença no período foi de 143,7%, com média anual de 39,2% e duração de 23 dias por episódio. A prevalência acumulada de absenteísmo-doença foi maior entre mulheres (52,0%) com idade superior a 40 anos (55,9%), com companheiro (49,9%), de baixa escolaridade (54,4%), profissionais de educação (54,7%), > 10 anos de serviço (61,9%) e múltiplos vínculos profissionais (53,7%). Os grupos de diagnósticos (CID-10) com as maiores prevalências acumuladas de licenças foram os do capítulo de transtornos mentais (26,5%), doenças osteomusculares (25,1%) e lesões (23,6%). CONCLUSÕES: Os indicadores de absenteísmo-doença expressam a magnitude desse fenômeno no serviço público e podem auxiliar no planejamento das ações de saúde do trabalhador, priorizando os grupos ocupacionais mais vulneráveis. .


BACKGROUND: Sickness absence, as work absenteeism justified by medical certificate, is an important health status indicator of the employees and, overall, sociodemographic and occupational characteristics are among the main factors associated with sickness absence. Public administration accounts for 21.8% of the formal job positions in Brazil. This population allows the study of a wide range of professional categories. OBJECTIVE: To assess the profile and indicators of sickness absence among public workers from the municipality of Goiania, in the State of Goiás, Brazil. METHODS: A cross-sectional study on certified sick leaves, lasting longer than three days, of all civil servants from January 2005 to December 2010. Prevalence rates were calculated using as main criteria the number of individuals, episodes and sick days. RESULTS: 40,578 certified sick leaves were granted for health treatment among 13,408 public workers, in an annual average population of 17,270 people, which resulted in 944,722 days of absenteeism. The cumulative prevalence of sick leave for the period was of 143.7%, with annual average of 39.2% and duration of 23 days per episode. The cumulative prevalence of sickness absence was higher among women (52.0%), older than 40 years old (55.9%), with a partner (49.9%), low schooling (54.4%), education professionals (54.7%), > 10 years of service (61.9%), and with multiple work contracts (53.7%). Diagnoses groups (ICD-10) with higher cumulative prevalence of sick leaves were those with mental disorders (26.5%), musculoskeletal diseases (25.1%), and injuries (23.6%). CONCLUSIONS: Indicators of sickness absence express the magnitude of this phenomenon in the public sector and can assist in planning health actions for the worker, prioritizing the most vulnerable occupational groups. .

Polyinosinic:polycytidylic acid (Poly I:C; 5 mg/kg body weight, ip) and lipopolysaccharide (LPS; 0.3 mg/kg body weight, ip) induced microglial and astrocytic activation in Sprague Dawley rats. Higher microglial and astrocytic activities were noticed in Poly I:C infused rats throughout the hippocampus till postnatal day 21 with a comparatively weaker response in LPS group. However, LPS induced inflammation persisted even after postnatal day 21, indicating thereby, that the Poly I:C (viral mimic) produces an acute inflammation, while LPS (bacterial endotoxin) produces chronic inflammation when exposed during early neonatal life.

In this study TSCA10 has been demonstrated as a testis-specific human gene that encodes a protein localized in sperm-tail and conserved in ciliary structure. Further investigations showed TSCA10 signalling and expression during embryogenesis, brain development and some malignancies including brain tumors. Given the role of this protein in neuronal development and in certain tumors, it could potentially serve as a diagnostic marker and therapeutic target in brain tumors. Therefore, using immunohistochemistry, we evaluated the localization of TSCA10 in different regions of brain, and its pattern/level of expression in tissue microarray [Cybrdi] containing human brain tumors and normal brain. In rat specimens, TSCA10 was mainly expressed in subventricular zone, hippocampus and granular layer of cerebellum of the brain. The antibody also stained the diverse and different types of human brain cancers. The TSCA10 was strongly over-expressed in glioblastoma and astrocytoma when compared to normal human brain. The expression of TSCA10 was also confirmed in astrocyte derived from a human astroctyoma cell line by immunocytochemistry. This study indicates that TSGAl0cxm be used as an immunohistochemical marker for human neuroglia and astrocyte cells and is over-expressed in brain tumors

Since astrogliosis is a histological marker usually observed in HIV-associated dementia (HIV-D),we decided to investigate the potential relationship between the expression of glial fibrillary acidicprotein (GFAP) and the regional distribution of cells positive (+) for this specific marker of astrocyte activation.Histological sections of brain tissues obtained at necropsy from 5 HIV-D patients and 5 age-matched controlswithout history of neuropsychiatric illness were immunostained with peroxidase. Mean numbers of GFAP(+)astrocytes were significantly increased in entorhinal cortex, hippocampus and subcortical white matter of patients,but values in frontal cortex and basal ganglia were similar to those of controls. In contrast, surface density ofimmunoreactive GFAP was significantly increased in all tested brain areas from all patients, including unusuallyaffected regions such as entorhinal cortex and hippocampus. Therefore, such consistent finding of hypertrophicastrocytes, ranging from highest cell percentajes in subcortical white matter to lowest in basal ganglia indicatesthat quantification of surface density in GFAP (+) cells appears to be a more reliable approach to score gliosisthan the counting of their cell nuclei. Because astrocyte activation involves both protective and detrimental effectson adjacent neuronal subsets, the evidence of regional differences in this reactive potential highlights theimportance of accurately defining their contribution to the neuropathogenesis not only of HIV-D, but of a widerange of neurodegenerative disorders.


Diferencias regionales en la activación astrocitaria en demencia asociada a HIV. Siendo laastrogliosis un signo histológico habitualmente presente en demencia asociada a HIV, se investigóla eventual relación entre expresión de proteína gliofibrilar ácida (GFAP) y localización regional de células positivaspara ese marcador específico de la activación astrocitaria. Por inmunoperoxidasa, se procesaron cortes histológicosde tejidos cerebrales obtenidos por necropsia de 5 pacientes y 5 controles de edades similares pero sin antecedentesneuropsiquiátricos. Según los valores de las medias registrados por conteo de astrocitos GFAP(+) en pacientes,el número fue significativamente mayor en corteza entorrinal, hipocampo y sustancia blanca subcortical, mientrasque en corteza frontal y ganglios basales no se encontraron diferencias con controles. En cambio, la densidad desuperficie del material GFAP inmunorreactivo en pacientes estuvo significativamente aumentada en todas las áreascerebrales analizadas, incluso en regiones inusualmente afectadas, como corteza entorrinal e hipocampo. Entreesos astrocitos hipertróficos, el mayor porcentaje correspondió a sustancia blanca subcortical, y el menor a gangliosbasales. Cabe concluir que el constante hallazgo de agrandamiento astrocitario señala a la medida de la superficieinmuno-reactiva como mejor índice de activación celular que el conteo de núcleos de las células marcadas. Dadoslos reconocidos efectos de la astrogliosis sobre las subpoblaciones neuronales vecinas, la comprobadaregionalización de ese potencial reactivo destaca el interés de precisar su contribución en la neuropatogenia, tantode demencia asociada a HIV como de otras enfermedades neurodegenerativas.

Viruses share antigenic sites with normal host cell components, a phenomenon known as molecular mimicry. It has long been suggested that viral infections might trigger an autoimmune response by several mechanisms including molecular mimicry. More than 600 antiviral monoclonal antibodies generated against 11 different viruses have been reported to react with 3.5 percent of cells specific for uninfected mouse organs. The main pathological feature of tropical spastic paraparesis/human T-lymphotropic virus type I (HTLV-I)-associated myelopathy (TSP/HAM) is a chronic inflammation of the spinal cord characterized by perivascular cuffing of mononuclear cells accompanied by parenchymal lymphocytic infiltration. We detected the presence of autoantibodies against a 98- to 100-kDa protein of in vitro cultured human astrocytes and a 33- to 35-kDa protein from normal human brain in the serum of HTLV-I-seropositive individuals. The two cell proteins exhibited molecular mimicry with HTLV-I gag and tax proteins in TSP/HAM patients, respectively. Furthermore, the location of 33- to 35-kDa protein cross-reaction correlated with the anatomical spinal cord areas (in the rat model) in which axonal damage has been reported in several cases of TSP/HAM patients. Our experimental evidence strongly suggests that the demyelinating process occurring in TSP/HAM may be mediated by molecular mimicry between domains of some viral proteins and normal cellular targets of the spinal cord sections involved in the neurodegeneration.

Cytoplasmic accumulation of granules of hemosiderin was observed in neoplastic astrocytes related to gliomas in two patients. The first patient was a 61-year-old man who had mixed oligodendroglioma and astrocytoma of the right frontal lobe of the brain. The second patient was a 17-year-old girl who had hypothalamic astrocytoma. Granules of hemosiderin had accumulated within the cytoplasm of astrocytes in both cases. In case 2, there was also an accumulation of hemosiderin in other phagocytes. The astrocytic nature of the hemosiderin-laden cells was confirmed by immunohistochemical localization of glial fibrillary acidic protein within the cytoplasm. The term "phagocytosis" was employed because of accumulation within astrocytic perikaryon of particulate material. Review of the literature discloses that various neuroepithelial derivatives such as choroidal epithelial cells, astrocytes, neurons, and ependymal cells can perform phagocytic function as revealed by accumulation of granules of hemosiderin within their cytoplasm but this finding has seldom been described.
