ABSTRACT High toxicity of the preservatives most frequently used in wood treatment and the resulting risks of handling pose a threat to small producers and to the environment. In an attempt to mitigate these problems, the present study was conducted with the objective of evaluating the preservative effect of tannic extract on biodeterioration of Acacia mearnsii wood. For this purpose, untreated and preserved specimens, some with tannin extract and some with a preservative mixture based on CCB (Chromated Copper Borate), were submitted to accelerated rotting trials with the fungus that causes white rot (Pycnoporus sanguineus) for 16 weeks. The evaluations were made with a basis on weight loss and chemical components analysis, and they showed that the natural resistance of Acacia wood is moderate when exposed to the white rot fungus. The tannin concentrations showed similar effects to those of the CBB mixture in all evaluations, i.e., they significantly increased the biological resistance of the material, which started to be classified as very resistant to the fungus. Overall, the results suggest that tannin can be considered as a potential natural preservative product.

The heavy metal resistant bacterium isolated from field soil and identified as Enterobacter sp. RZS5 tolerates a high concentration (100-2000 mM) of various heavy metal ions such as Mn2+, Ni2+, Zn2+, Cu2+, CO2+ and Fe2+ when grown in such environment and produces exopolysaccharides (EPS). Here, we have demonstrated EPS production by Enterobacter sp. RZS5 during 60 h of growth in yeast extract mannitol broth (YEMB). The yield increased by two fold after the addition of 60 M of Ca2+; 50 M of Fe2+ and 60 M of Mg2+ ions in YEMB, and the optimization of physico-chemical parameters. EPS was extracted with 30% (v/v) of isopropanol as against the commonly used 50% (v/v) isopropanol method. EPS-rich broth promoted seed germination, shoot height, root length, number of leaves and chlorophyll content of wheat (Triticum aestivum) and peanut (Arachis hypogaea) seeds. The higher colony-forming unit of Enterobacter sp. in soil inoculated with EPS rich broth of Enterobacter sp. indicated the root colonizing potential and rhizosphere competence of the isolate. The FTIR spectra of the EPS extract confirmed the presence of the functional group characteristics of EPS known to exhibit a high binding affinity towards certain metal ions. This overall growth and vigour in plants along with the effective root colonization, reflected the potential of the isolate as an efficient bio-inoculant in bioremediation.

The Madagascar Periwinkle, Catharanthus roseus (L.) G. Don (a valued medicinal plant) was exposed to different concentrations ofheavymetals like, CdCl, and PbCl, with a view to observe their bioaccumulation efficiency. Germination was inhibited by both the heavy metals in the seeds previously imbibed in GA, and KNO, for 24 hr. EC50 (the effective concentration which inhibits root length by 50%) was recorded as 180 microM for CdCl2, and 50 microM for PbCl2. Both alpha-amylase and protease activity were reduced substantially on treatment of seeds with increasing concentrations of CdCl2, and PbCl2. Malondialdehyde (MDA) a product of lipoxigenase (LOX) activity also increased due to the treatment of both CdCl, and PbCl2. When two-months-old plants grown in normal soil were transferred to soils containing increasing amounts of these two heavy metals, senescence of lower leaves and extensive chlorosis were noticed after four days of transfer However, plants gradually acclimatized and after 20 days the chlorophyll content was almost comparable to normal. Plants receiving CdCl2 treatment (250 microg g(-1) and less) became acclimatized after two weeks and started normal growth. But PbCl2 of 432 microg g(-1) and less could not affect the plant growth throughout, after a preliminary shock was erased. In case of CdCl2 treatment, a stunted growth with reduced leaf area, reduced biomass and sterility were recorded after six months, while plants show normal growth and flowering in case of PbCl2 treatment. Total alkaloid was also found to be decreased in the roots of CdCl2 treated plants. No change was observed in case of PbCl2. GA3 treatments to the CdCl2 treated plants show internode elongation and increase in leaf area with relatively elongated leaves and thinning of stem diameter AAS analyses of leaves of treated plants exhibited 5-10% accumulation of cadmium, but there was no accumulation of lead at all.

A biorremediaçäo natural, ou seja, o uso de microorganismos nativos para degradar substâncias periosas sem nenhuma estimulaçäo tecnológica, mostra-se como uma técnica promissora e de baixo custo para se aplicada no manejo de plumas de hidrocarbonetos de petróleo. Esta técnica exige que se faça uma total caracterizaçäo e monitoramento do local contaminado para se verificar se os processos de atenuaçäo natural continuam proporcionando uma adequada proteçäo de riscos. Um grande progresso tem sido feito na Europa e nos Estados Unidos para entender os fatores hidrogelógicos e microbiológicos que influenciam a biorremediaçäo natural de aquíferos contaminados por combustíveis. No entanto, esta experiência deve ser extrapolada com cuidado em locais contaminados no Brasil, onde a gasolina contém 22 por cento de etanol. Estudos preliminares de laboratório mostram que o etanol pode aumentar a solubilizaçäo dos compostos BTX. É necessário que se conheça os efeitos bioquímicos, físicos e ecológicos do etanol para que se desenvolva no Brasil uma base racional para seleçäo, modelagem matemática e monitoramento de sistemas apropriados de biorremediaçäo natural
