RESUMO

Infectious bursal disease virus (IBDV) is a member of the Avibirnavirus genus of the Birnaviridae family which genome consists of two segments (A and B) of double stranded RNA. Segment A gene of KNU08010 isolate, which was isolated from a 15-day-old chicken flock in 2008, was sequenced and compared with other IBDV isolates including SH/92 strain, the first Korean very virulent (vv) IBDV isolate. The amino acid sequences of segment A gene showed that KNU08010 had 99.2% homology with SH92 strain. KNU08010 isolate had specific amino acids A222, I242, I256, I294 and S299 which are highly conserved among vvIBDV strains. Phylogenetic analysis based on the nucleotide sequences of variable region of the VP2 gene of 18 IBDV strains revealed that KNU08010 was grouped with vvIBDVs and was closely related to Korean vvIBDVs isolated from wild birds.

RESUMO

The cDNA nucleotide sequence of genome segment B encoding the VP1 protein was determined for the aquatic birnavirus GC1 isolated from the rockfish Sebastes schlegeli in Korea. The VP1 protein of GC1 contains a 2,538 bp open reading frame, which encodes a protein comprising 846 amino acid residues that has a predicted MW of 94 kDa. The sequence contains 6 potential Asn-X-Ser/Thr motifs. Eight potential Ser phosphorylation sites and 1 potential Tyr phophorylation site were also identified. GC1 contains the Leu-Lys-Asn (LKN) motif instead of the typical Gly-Asp- Asp (GDD) motif found in other aquatic birnaviruses. We also identified the GLPYIGKT motif, the putative GTPbinding site at amino acid position 248. In total, the VP1 regions of 22 birnavirus strains were compared for analyzing the genetic relationship among the family Birnaviridae. Based on the deduced amino acid sequences, GC1 was observed to be more closely related to the infectious pancreatic necrosis virus (IPNV) from the USA, Japan, and Korea than the IPNV from Europe. Further, aquatic birnaviruses containing GC1 and IPNV have genogroups that are distinct from those in the genus Avibirnaviruses and Entomo-birnaviruses. The birnavirusstrains were clustered into 5 genogroups based on their amino acid sequences. The marine aquatic birnaviruses (MABVs) containing GC1 were included in the MABV genogroup; the IPNV strains isolated from Korea, Japan, and the USA were included in genogroup 1 and the IPNV strains isolated primarily from Europe were included in genogroup 2. Avibirnaviruses and entomobirnaviruses were included in genogroup 3 and 4, respectively.

RESUMO

Se aislaron tres virus de la infección de la bolsa de Fabricio (VIBF) a partir de muestras obtenidas de muestras en granjas avícolas en México, aquéllos se denominaron H, P y G. Se determinaron sus propiedades antigénicas mediante la técnica de ELISA por captura de antígenos, utilizando los anticuerpos monoclonales (Mab) B-29, R-63, BK-9, B-69 y 57. Se evaluó su virulencia en aves inoculadas mediante la observación de signos clínicos, mortalidad, proporción del peso bursal con el peso corporal, lesiones microscópicas, persistencia de los virus en tejidos linfoides, serología contra VIBF y medición de la respuesta inmune hacia Brucella abortus y eritrocitos de ovino. Los virus P y G presentaron los epítopes reconocidos por lo B-29, R-63 y B-69, que están presentes en las cepas clásicas de VIBF, el virus H no pudo ser reconocido por ningún Mab. Los tres aislamientos indujeron la presentación subclínica de la IBF en las aves inoculadas, se observó marcada atrofia bursal con deplesión linfoide severa, persistencia del VIBF en bolsa hasta los catorce días posinoculación y seroconversión de las aves a partir de la segunda semana después de la inoculación. A pesar de que no se detectó plenamente un estado de inmunodepresión mediante la respuesta a los dos antígenos utilizados, no se puede descartar un eventual efecto inmunodepresor por el aislamiento "H". Tampoco se detectaron diferencias en la virulencia de los tres VIBF aislados
