RESUMO

Cross-sectional study that used the Social Network Index and the genogram to assess the social network of 110 family caregivers of dependent patients attended by a Home Care Service in São Paulo, Brazil. Data were analyzed using the test U of Mann-Whitney, Kruskal-Wallis and Spearman correlation. Results were considered statistically significant when p<0,05. Few caregivers participated in activities outside the home and the average number of people they had a bond was 4,4 relatives and 3,6 friends. Caregivers who reported pain and those who had a partner had higher average number of relatives who to trust. The average number of friends was higher in the group that reported use of medication for depression. Total and per capita incomes correlated with the social network. It was found that family members are the primary caregiver’s social network.

Estudio transversal que utiliza el Índice de la Red Social y el genograma para evaluar la red social de los 110 cuidadores familiares de enfermos dependientes atendidos por un servicio de cuidados en el hogar, en São Paulo. Los datos fueron analizados por las pruebas de Mann-Whitney, Kruskal-Wallis y la correlación de Spearman. Los resultados se consideraron estadísticamente significativos cuando p<0,05. Pocos cuidadores participaban en actividades fuera del hogar y el número promedio de personas con las cuales tenían vínculo fueran 4,4 personas de la familia y 3,6 amigos. Los que informaron dolor en el cuerpo y los que tenían una pareja tenían mayor número medio de familiares en que confiar. El número medio de amigos fue mayor en el grupo que informó el uso de medicación para la depresión. Los ingresos totales y per cápita se correlacionaron con la red social. Se encontró que los miembros de la familia son la principal red social del cuidador.

Objetivo Avaliar a rede social de 110 cuidadores familiares de pacientes dependentes atendidos por um Serviço de Assistência Domiciliária no município de São Paulo. Método Estudo transversal, que utilizou o Social Network Index e o genograma. Os dados foram analisados pelos testes U de Mann-Whitney, Kruskal-Wallis e correlação de Spearman. Foram considerados estatisticamente significantes quando p <0,05. Resultados Poucos cuidadores participavam de atividades extradomiciliares e o número médio de pessoas com quem mantinham vínculo era de 4,4 familiares e 3,6 amigos. Cuidadores que referiram dor no corpo e aqueles que possuíam companheiro apresentaram maior número médio de parentes em quem confiar. A média de amigos foi superior no grupo que referiu uso de medicamentos para depressão. As rendas total e per capita mostraram correlação com a rede social. Conclusão Verificou-se que os familiares são a principal rede social do cuidador. .

RESUMO

The anti-tumor effect of R-Phycoerythrin (R-PE) from Porphyra haitanensis was studied using cell line HeLa as an in vitro model and Sarcoma-180 (S180) tumor-bearing mice as an in vivo model. The results showed that the combination treatment of R-PE and photodynamic therapy PDT) significantly inhibited the growth of HeLa cells up to 81.5%, with a fair dose-effect relationship, but did not inhibit endothelial cells. The annexin v-fitc/PI fluorescence staining experiments demonstrated that at doses between 0~60µg/mL, apoptosis cells and later stage apoptosis cells or necrosis cells increased significantly as the R-PE dosage increased. DNA electrophoresis showed that after R-PE+PDT treatment of HeLa cells for 24 hours, a light "smear" band between 100~400bp appeared to indicate the degradation of genomic DNA. The QRT-PCR results showed that R-PE+PDT treatment increased caspase-3 and caspase-10 gene expression and decreased the Bcl-2 gene expression level significantly as the R-PE dose increased, implying that R-PE promoted HeLa cell apoptosis. Compared with untreated S180 tumor-bearing mice, R-PE injection significantly inhibited the growth of S180 in tumor-bearing mice up to 41.3% at a dose of 300mg-kg-1. Simultaneously, the significant increase of superoxide dismutase (SOD) activity in serum (p < 0.01) and the decrease of the malondialdehyde (MDA) level in liver suggests that R-PE improved the anti-oxidant ability of the S180 tumor-bearing mice, which may related to its antitumor effect. In addition, the R-PE caused a significant increase (p < 0.05) in the spleen index and thymus index, and a significant increase (p < 0.01) in lymphocyte proliferation, NK cell kill activity and the TNF-α level in the serum of S180 tumor-bearing mice. These results strongly suggest that the antitumor effect of R-PE from Porphyra haitanensis functioned by increasing the immunity and antioxidant ability of S180 tumor-bearing mice, promoting apoptosis by increasing protease gene expression and TNF-α secretion.

OBJECTIVES: Apoptosis is the process of programmed cell death (PCD) that occurs in both animal and plant cells. Protozoan parasites possess metacaspase and these caspase-related proteases could be involved in the PCD pathways in these organisms. Therefore we analyzed the activities of metacaspase and PARP genes in Leishmania infantum (MCAN/IR/96/LON49) treated with miltefosine. MATERIALS AND METHODS: Anti-leishmania activity of miltefosine was studied by treatment of cultured promastigotes with various concentration of miltefosine. MTT assay and Annexin-V FLUOS staining by using FACS flow cytometry methods were used. Cytotoxic potential of HePC on the amastigots of L.infantum was evaluated in J774 cell line. In addition, metacaspase and PARP genes expression of treated L. infantum were studied. RESULTS: Miltefosine led to dose-dependent death of L. infantumwith features compatible with apoptosis. Over expression of metacaspase and PARP was seen 6 hr after treatment. CONCLUSIONS: Our study showed that miltefosine exerts cytotoxic effect on L. infantum via an apoptotic-related mechanism.

RESUMO

We postulated that apolysis was processed in accordance with apoptotic changes occurring in a cestode, Spirometra erinacei (Pseudophyllidea). We cloned the novel putative apoptosis-associated gene from S. erinacei via screening of a S. erinacei cDNA library with a ced-3 gene (activator of apoptosis) probe from Caenorhabditis elegans. We identified a 261-bp cDNA sequence, which encodes for an 86-amino acid protein. The cloned gene expression was observed in the neck and gravid proglottids via Northern blotting, using cloned cDNA inserts as probes, but the clone was not expressed in any of other tissues. We suggest that this gene may be involved in the apolysis of S. erinacei during normal tissue development and differentiation in cestode parasites.

OBJECTIVE@#To investigate the expression of caspase-3 in different posttraumatic intervals and severity of brain injury.@*METHODS@#The study examined brain tissue samples of slight (n = 24), severe (n = 24) brain injury and control (n = 6) of rat, using immunohistochemical staining, western-blot and RT-PCR method.@*RESULTS@#Up-regulating of caspase-3 expression was found in tissue from traumatic brain injury compared with controls in early 1 hour after TBI, and lasted for 14 days. The gray degree and threshold area of caspase-3 positive cells is different in different severity of brain injury.@*CONCLUSION@#The increasing of caspase-3 expression indicates that TBI exists. The gray degree, threshold area of caspase-3 positive cells and the cleavage degree of pro-caspase-3 have association with the severity of brain injury.
