RESUMO

Abstract Herein the chemical constituents and the anti-pain properties of the essential oil from the stem bark of Casuarina equisetifolia L. (Casuarinaceae) grown in Nigeria were evaluated. The essential oil was obtained by hydrodistillation method in an all glass Clevenger-type apparatus, and characterized by gas chromatography (GC-FID) and gas chromatography-mass spectrometry (GC-MS). The hot plate method was used to determine the anti-nociceptive property whereas the anti-inflammatory activity was evaluated by carrageenan-induced and formalin experimental models. The pale-yellow essential oil was obtained in yield of 0.21% (v/w), calculated on a dry weight basis. The main constituents of the essential oil were methyl salicylate (30.4%), a-zingiberene (15.5%), (E)-anethole (9.5%), b-bisabolene (8.6%), b- sesquiphellandrene (6.9%), and ar-curcumene (6.2%). In the anti-nociceptive study, the rate of inhibition increases as the doses of essential oil increases with optimum activity at the 30th and 60th min for all tested doses. The essential oil displayed anti-nociceptive activity independently of reaction time at the highest tested dose (200 mg/kg). The essential oil of C. equisetifolia moderately reduced pain responses in early and late phases of the formalin test. The oil inhibited the paw licking in the neurogenic phase (60-63%) compared to the late phase of the formalin test. The carrageenan- induced oedema model revealed the suppression of inflammatory mediators within the 1st - 3rd h. Thus, C. equisetifolia essential oil displayed both anti-nociceptive and anti-inflammatory activities independent of the dose tested. The anti-inflammatory and anti-nociceptive activities of C. equisetifolia essential oil are herein reported for the first time

RESUMO

Abstract The Brazilian native species Cestrum intermedium, known as mata-boi, induces hepatotoxicity and death when ingested by cattle. While most studies on this species focus on toxicological features, our study is the first to describe the anatomy and in vitro biological activities of Cestrum intermedium. We investigated adult leaves and stems by histochemistry, described their anatomy, performed physical-chemical analysis, determined in vitro antioxidant and antimicrobial activities, and identified secondary metabolites. A few noteworthy anatomical features were the anomocytic stomata on the abaxial surface and the absence of trichomes, in addition to the circular shaped petiole with two projections on the adaxial surface. Histochemical analysis showed chemical markers such as alkaloids, usually reported as toxic, and terpenoids. Potassium nitrate (ATR-FTIR) and lupeol palmitate (NMR) were detected on the crude stem extract. Thermogravimetric and physical-chemical analysis provided fingerprint parameters for the species. Minimal Inhibitory Concentration (MIC) assay revealed that Staphylococcus aureus, Staphylococcus epidermidis, and Candida albicans were weakly inhibited by extract samples. Chloroform and ethyl acetate fractions presented high phenolic content, which resulted in in vitro antioxidant activity. These novel features expand the knowledge about this species, considering that previous studies mainly focused on its toxicity. Our study also provided characteristics that may help in avoiding misidentification between Cestrum members, especially when taxonomic keys cannot be employed, as in the absence of flowers and fruits.

RESUMO

Araujia odorata is a sub-shrub native from Argentina, Brazil, Paraguay and Uruguay, whose latex, roots and leaves are used in traditional medicine. The objective of this work is to study the foliar morpho-anatomy of six populations in an altitudinal gradient (359-2155 m.a.s.l.) of Northwestern Argentine and to determine the nature of the compounds present in the laticiferous of the stems and fruits using conventional techniques for plant anatomy. The populations under study did not show significant morpho-anatomical differences. They presented simple leaves, pinnated brochydodromous venation, amphiestomatic isolateral lamina, brachy, anomo and amphicyclocytic stomata, eglandular trichomes, midvein with bicolateral vascular bundle and non-articulated laticifers continuous in the petiole, stem and fruits. Differences in the quantified foliar parameters are observed, however, only the density of trichomes, stomata and the thickness of the cuticle are positively correlated with the altitudinal gradient, indicating phenotypic plasticity. Histochemical analysis of laticifers and other stem idioblasts of A. odoratawas performed for the first time.

This study aimed to analyze stem anatomical characteristics of Trapa natans from five Skadar Lake locations (L1 - Milovica bay, L2 inflow of the Moraca river, L3 - Kamenik, L4 - Grmozur, L5 - Lipovik) with different concentrations of Cu, Mn, Zn, Co and Pb, during the summer period of the year 2012. Cross sections of stem were made using cryotechnic procedure. For all analyzed quantitative anatomical parameters, the minimum values at the location L2 were recorded, with the presence of maximum content for all investigated metals in stem of Trapa natans. On the other hand, except for cobalt, at the location L4 the minimum concentration for all investigated metals were recorded. Plants collected from this location have the largest average values of the most measured anatomical parameters. The results of Discriminant Analysis showed that plants from different location could be clearly classified into three groups according to their stem quantitative anatomical characteristics which corresponds with heavy metals content. Our research also showed that there is no statistically significant correlation between the content of most investigated metals (Co, Cu, Pb and Zn) and the values of anatomical parameters. Statistically significant negative correlation was found between Mn content in stem and values of two anatomical characters (stem cross-sectional area and Mn content, r = - 0.88; p < 0.05; number of hypodermal cell layers and Mn content, r = - 0.90; p < 0.05).

RESUMO

BACKGROUND: The biomechanical properties of maize stalks largely determine their lodging resistance, which affects crop yield per unit area. However, the quantitative and qualitative relationship between micro-phenotypes and the biomechanics of maize stalks is still under examined. In particular, the roles of the number, geometry, and distribution of vascular bundles of stalks in maize lodging resistance remain unclear. Research on these biomechanical properties will benefit from high-resolution micro-phenotypic image acquisition capabilities, which have been improved by modern X-ray imaging devices such as micro-CT and the development of micro-phenotyping analysis software. Hence, high-throughput image analysis and accurate quantification of anatomical phenotypes of stalks are necessary. RESULTS: We have updated VesselParser version 1.0 to version 2.0 and have improved its performance, accuracy, and computation strategies. Anatomical characteristics of the second and third stalk internodes of the cultivars 'Jingke968' and 'Jingdan38' were analyzed using VesselParser 2.0. The relationships between lodging resistance and anatomical phenotypes of stalks between the two different maize varieties were investigated. The total area of vascular bundles in the peripheral layer, auxiliary axis diameter, and total area of vascular bundles were revealed to have the highest correlation with mechanical properties, and anatomical phenotypes of maize stalk were better predictors of mechanical properties than macro features observed optically from direct measurement, such as diameter and perimeter. CONCLUSIONS: This study demonstrates the utility of VesselParser 2.0 in assessing stalk mechanical properties. The combination of anatomical phenotypes and mechanical behavior research provides unique insights into the problem of stalk lodging, showing that micro phenotypes of vascular bundles are good predictors of maize stalk mechanical properties that may be important indices for the evaluation and identification of the biomechanical properties to improve lodging resistance of future maize varieties.

RESUMO

Abstract The quality of forage production is a prerequisite to raising livestock. Therefore, income losses in this activity, primarily cattle raising, can result in the impossibility of economic activity. Through the qualitative and quantitative anatomical study of Urochloa hybrida cv. Mulato II and U. brizantha cv. Marandu, we searched for descriptions and compared changes in the individual vegetative body from populations with death syndrome pastures (DPS). Specimens were collected at different physiological stages from farms in northern Mato Grosso. After collection, the individuals were fixed in FAA50 and stored in 70% alcohol. Histological slides were prepared from the middle third of the sections of roots, rhizomes, and leaves, and the proportions and characteristics of tissues were evaluated in healthy, intermediate, and advanced stages of DPS. Changes were compared between cultivars. With the advancement of the syndrome, the following changes were observed: a more marked decrease in the length of roots in U. hybrida; disorganization of the cortical region of the roots and rhizome cultivars; fungal hyphae in roots and aerenchyma formation in U. hybrida; a decrease in sclerenchyma fiber proportions in roots and leaves; sclerification of the epidermis of U. brizantha rhizomes; and an increase in pericyclic fibers in U. hybrida. Furthermore, there was a decrease in the volume of epidermal cells of the abaxial face of the leaves of both cultivars, with a greater reduction in U. hybrida; a gradual decrease in thickness in the midrib of leaves similar to leaf mesophyll; conduction system obstructions; partial or total cell lysis in roots and rhizomes affected by the syndrome. Obstructions in sieve tube element and companion cells, and sometimes obstruction in xylem vessel elements. The evolution of DPS in cultivars was similar, but there were variations, arising probably from the physiological response to stress, such as aerenchyma formation in the root and increased pericycle in the rhizome of U. hybrida.

In Venezuela, Atriplex is represented by A. cristata and A. oestophora, the latter being endemic; they inhabit coastal areas with high temperatures, high solar radiation and sandy soils with high salt content. This work aimed to provide information to facilitate and clarify these species taxonomic delimitation, throughout the study of the anatomy of their vegetative organs; this may also clarify our understanding of their adaptability to soil and climatic conditions prevailing in areas they inhabit. The plant material was collected from at least three individu- als of each species in Punta Taima Taima and Capatárida, Falcon. Segments of roots, located near the neck and towards the apex, apical, middle and basal internodes of stems, were taken; and of leaves, located in the middle portion of plants. This material was fixed in FAA (formaldehyde, acetic acid, 70% ethanol) until processing. Semipermanent and permanent microscope slides were prepared with transverse or longitudinal sections, made using a razor (free-hand) or a rotation microtome, in this latter case, after paraffin embedding; besides, additional plates were mounted with portions of leaf epidermis, obtained by the maceration technique. The sections were stained with aqueous toluidine blue (1%) or safranin-fast-green, and mounted in water-glycerin or in Canada balsam. In order to calculate the vulnerability index, the vessel diameter in the vascular rings of roots, as well as their density, were quantified. Our results revealed structural features in the different organs, that resulted of taxonomic value and allowed the distinction of the species: in the leaf, the presence of aquifer tissue, the number of vascular bundles and their organization in the midrib, and the collenchyma differentiation in this part of the leaf; in the roots, the xylem and phloem arrangement in the growth rings, the nature of conjunctive tissue, and the presence of included phloem in one species. In addition, the species showed typical anatomical features of halophytes and xerophytes, such as: high density of trichomes on leaves and young stems which act as salt secreting glands, abundant sclerenchyma in stems and roots, water storage tissue and Kranz anatomy in leaves, narrow cortical region in young roots, presence of cambial variants in stems and roots, as well as short and narrow xylem vessels. Vulnerability index calculations indicated that both species tend to assure conduction but not the efficiency of the system. Atriplex species have anatomical characters which facilitate their adaptation to the special conditions prevailing in their habitats and that may be used for taxonomic delimitation.

Woodson, Apocynaceae is a herb native to South America employed in folk medicine as an analgesic, anti-inflammatory and to inhibit snake venom effects. This work was carried out to study the morpho-anatomical characters of the leaf and stem in order to advance knowledge on this medicinal plant and on pharmacognostic quality control. The plant material was fixed and prepared according to light and scanning electron microtechniques. Its leaves are simple, alternate and ovate-obovate. The epidermis is uniseriate and coated with a thick and striate cuticle. The stomata are paracytic and occur on both foliar surfaces. The dorsiventral-like mesophyll has a sub-epidermal parenchymatic layer containing phenolic substances and is traversed by minor collateral vascular bundles. The midrib is biconvex and the petiole is concave-convex, both presenting bicollateral vascular bundles in an open arc. The stem is circular in transverse section and the epidermis remains in incipient secondary growth. A sub-epidermal parenchymatic layer containing phenolic substances, a discontinuous sclerenchymatic sheath of non-lignified fibres and cylinders of external phloem, xylem and internal phloem occur. Numerous branched laticifers and idioblasts with phenolic substances are present in the leaf and stem.

A nervura central é biconvexa e o pecíolo é côncavo-convexo, ambos apresentando feixes vasculares bicolaterais em arco aberto no parênquima fundamental.

Desmoncus orthacanthos is a Neotropical climbing palm that resembles rattan and therefore has similar potential applications. The genus Desmoncus (subfamily Arecoideae, subtribe Bactridinae) is distributed throughout the Americas, from veracruz, Mexico, to Brazil and Bolivia. The anatomical characteristics of its support tissue have not been thoroughly studied, although some observations from Central American artisans suggest that the stems collected from undisturbed sites possess better characteristics; these include a good capacity to withstand bending without breaking (i.e. higher fracture strength than plants from disturbed sites). Stem samples were collected from individuals from disturbed and undisturbed sites, at three points along the length of the stem (basal, medium and apical). Collections were made of one ramet from five individuals (n=5) at both sites. Each ramet was divided into three sections: basal, from soil surface to a height of 0.5 m; medium, from a height of 0.5 to 5.0 m; and apical, from a height 5.0 to 10.0 m. An anatomical analysis including vascular bundles, parenchyma elements and fibers was performed in the radial direction and also along the longitudinal direction of the stems. The amount of vascular bundles was greater for samples from undisturbed site stems; the amount of parenchyma cells differ between samples from both sites and the amount of fibers was greater for samples from disturbed site stems. The anatomical structural dimensions were smaller for samples from the undisturbed site stems. These findings partially confirm the artisans’ belief and supports the conclusion that microclimatic conditions affect plant anatomical structure. Rev. Biol. Trop. 56 (2): 937-949. Epub 2008 June 30.

A medula é parenquimática.

Commonly known as erva-de-colégio in Portuguese, Elephantopus mollis Kunth (Asteraceae) is a perennial herb, with slightly lignified base, short branches and purple flowers. Its leaves are used as emollient, vulnerary and diaphoretic, and to treat bronchitis, coughs and influenza in folk medicine. The aim of this work was to study leaf and stem anatomical characters, in order to expand the knowledge for this medicinal plant identification. Adult leaves and stem fragments were fixed, sectioned and stained either with astra blue/basic fuchsine or toluidine blue. Microchemical essays and scanning electron analysis were also carried out. The leaf has wavy-contoured cells in surface view, forming a single layer and coated with striate cuticle. Non-glandular and glandular trichomes occur. The former is erect, multicellular and uniseriate. The glandular ones are capitate and biseriate. The mesophyll is dorsiventral and the midrib is traversed by collateral vascular bundles arranged in open arc. The stem, in initial secondary growth, has uniseriate epidermis coated with striate cuticle and trichomes similar to the leaf. In the cortex, annular collenchyma alternates with chlorenchyma and an endodermis with Casparian strips is found. Sclerenchymatic caps adjoin the phloem, which is formed outwards by the vascular cambia, while the xylem is produced inwards. The pith is parenchymatic. Small calcium oxalate crystals are present in the leaf and stem.

RESUMO

Information on anatomical structure is needed by breeders working on improvement for drought tolerance. For studying the effect of polyploidy on cassava anatomy and its significance to tolerance to drought, we induced a polyploidy type of a selected clone (UnB 530) by applying an aqueous solution of 0.2% colchicine on lateral buds for a period of 12 h. The stem identified as tetraploid was propagated to produce the whole plant. Free-hand cross-sections of the median portion between stem internodes were made. They were clarified using 50% sodium hypochlorite solution, stained with 1% safranin-alcian blue, passed through an ethanol series and butyl acetate and mounted in synthetic resin. The tetraploid type showed more prismatic and druse crystals in the cortical parenchyma, and its pericycle fibers had thicker walls. The secondary xylem of tetraploid types was wider than diploid ones, having thinner walls and less starch.

RESUMO

Wood and bark anatomy and histochemistry of Acacia bilimekii Humb. & Bonpl., Acacia cochliacantha Mcbride., Conzatia multiflora (Rob) Stand. and Guazuma ulmifolia Lam.are described from stem samples collected in a tropical dry forest (Morelos,Mexico). Enzyme activities were tested in tangential, radial and transverse cuts of fresh material. Histochemistry and stem anatomy were studied on similar cuts previously softened in a solution of water-glicerol-PEG. Our results show that the anatomical patterns of bark and wood, as well as the histochemical patterns and specific gravity, are influenced by water accessibility and climate; these patterns could guarantee mechanical and anti-infection strategies to support extreme conditions. Enzyme cytochemistry reveals biochemical activities probably related to lipid utilization routes for the lignification processes and for synthesis of extractives; these results suggest that the formation and maturation of woody tissue is very active at the beginning of the rainy season. These species are widely used by the local population. Traditional uses include firewood, dead and live fences, fodder, construction, supporting stakes, handcrafts, farming tools, extraction of tanning products, and medicine. There is no relationship between use and abundance. Alternative uses are proposed according to a density index
