RESUMO

The aim was to investigate the proteolytic activity of plasmin and its long-term complications. Plasmin was injected into the vitreous cavity of rabbits' eyes. Slit lamp biomicroscopy and electroretinography were performed. Rabbits were serially sacrificed at four months, and globes fixated and prepared for light and transmission electron microscopy. In both the plasmin-injected and control eyes, electroretinography showed a transient decrease in the amplitude, but this recovered to the baseline level in a week. Under the light microscope, the plasmin-treated eyes had a smooth retinal surface, implying separation of the vitreous cortex from the retina. In the control eyes, the collagen fibers remained on the retinal surface. By transmission electron microscopy, the plasmin-treated eyes showed a vitreous-free retinal surface, but no vitreoretinal separation was observed in the control eyes. Plasmin induces a cleavage between the vitreous and the internal limiting membrane, with no long-term complications, so may be a useful pharmacologic adjunct to vitrectomy.

RESUMO

This study was designed with the aim of assessing the fibrinolytic status of patients suffering from obliterative arterial disease [OAD] as compared to healthy controls. Twenty-five patients with arterial obliterative diseases presented acutely were chosen for fibrinolytic assay and compared with 25 healthy controls. Twelve patients had critical ischemia and 15 patients were diabetics. Twenty-one patients had lower limb ischemia while, 4 patients had upper limb ischemia. The results of the study suggested that the fibrinolytic system is significantly impaired in cases of acute or severely ischemic patients when compared to controls. It was concluded that fibrinolytic assay could be added as a routine investigation in cases of critical ischemia and early onset atherosclerosis. Therapeutic options could be adjusted according to the fibrinolytic assay results

El objetivo de este estudio fue extraer y cuantificar el activador de plasminógeno AP en glándulas salivares de la rata. AP se extrajo por homogeneización de 1 vol de tejido com 1 vol de una solución de 2M KSCN. La solución con AP fue pipeteada por triplicado en placas de fibrina PF ricas y pobres en plasminógeno. La actividad fibrinolítica inducida por AP se observó como áreas circulares de licuefacción medidas en mm2, resultado del producto de dos diámetros perpendiculares de las zonas de lisis. El AP de la submaxilar produjo un término medio de actividad lítica de 198 mm2 + ou - 18ESM únicamente en PF ricas en plasminógeno. Extrapolando al área de lisis producida por 50 mg/ml de plasmina. No se observó lisis con AP extraído de la parótida y sublingual. La droga antifibrinolítica E-ACA, redujo significantemente, en una acción inhibitoria, dosis dependiente, la actividad fibrinolítica del AP de la submaxilar. El hecho de que AP indujo lisis únicamente en PF ricas en plasminógeno, la que fue significativamente reducida por E-ACA, indica claramente que la actividad lítica observada en el sustrato fibrina es debido a una fibrinolisis específica y no debido a una proteólisis no específica
