The Fundação de Medicina Tropical Dr. Heitor Vieira Dourado (FMT-HVD), located in Manaus, the capital of the State of Amazonas (Western Brazilian Amazon), is a pioneering institution in this region regarding the syndromic surveillance of acute febrile illness, including arboviral infections. Based on the data from patients at the FMT-HVD, we have detected recurrent outbreaks in Manaus by the four dengue serotypes in the past 15 years, with increasing severity of the disease. This endemicity has culminated in the simultaneous circulation of all four serotypes in 2011, the first time this has been reported in Brazil. Between 1996 and 2009, 42 cases of yellow fever (YF) were registered in the State of Amazonas, and 71.4% (30/42) were fatal. Since 2010, no cases have been reported. Because the introduction of the yellow fever virus into a large city such as Manaus, which is widely infested by Aedes mosquitoes, may pose a real risk of a yellow fever outbreak, efforts to maintain an appropriate immunization policy for the populace are critical. Manaus has also suffered silent outbreaks of Mayaro and Oropouche fevers lately, most of which were misdiagnosed as dengue fever. The tropical conditions of the State of Amazonas favor the existence of other arboviruses capable of producing human disease. Under this real threat, represented by at least 4 arboviruses producing human infections in Manaus and in other neighboring countries, it is important to develop an efficient public health surveillance strategy, including laboratories that are able to make proper diagnoses of arboviruses.

Morphological evidence of hybridization between Ramphocelus flammigerus subspecies (Passeriformes: Thraupidae) in Colombia. Habitat modifications such as deforestation and the increase of agricultural activities, have led to uncommon faunal interactions. In Colombia, this condition have caused the secondary contact of subspecies of Ramphocelus flammigerus populations from Cauca valley and the Pacific coast; and some specimens with rumps of intermediate colors of the subspecies have been found and are thought as hybrids. The objective of this study was to assess the presence of morphological evidence that may suggest hybridization and may explain the origin of individuals with intermediate coloration. We predict that if subspecies hybridize, they will be more similar in morphology when coexisting than when separated. Alternatively, coexisting subspecies might diverge in sympatry, because of selection to reduce competition for resources (character displacement). For this, a survey in 15 localities was undertaken: 10 allopatric areas (five for each subspecies), and five sympatric areas. Mist nets were used to capture individuals and a total of seven morphological characters were measured. To identify the patterns of morphological variation, we compared morphology of subspecies, sympatric and allopatric populations and individuals of intermediate colors. Consequently, we performed discriminant analysis and test for differences between groups by using 95% confidence intervals for log-ratio tests. A total of 112 individuals were captured (46 intermediate-colored individuals, 20 R. f. flammigerus, and 46 R. f. icteronotus. Discriminant analyses showed that subspecies were well differentiated, and intermediate individuals overlapped with them. Log-ratio test, based on Mahalanobis distances, showed that intermediate individuals were morphologically more similar to both subspecies than subspecies themselves. In addition, log-ratio tests showed that subspecies sympatric populations were similar but allopatric ones were different, and that individuals of intermediate colors were more similar to sympatric than to allopatric populations of the two subspecies. Therefore, morphological evidence supports the predictions of a hybridization hypothesis among the subspecies of R. flammigerus. In conclusion, the analysis of morphological variation in R. flammigerus suggests that hybridization between subspecies is occurring and that a process of genetic introgression is probably in progress.


Las modificaciones de los hábitats naturales, tales como la deforestación y el incremento de las actividades agrícolas, han conducido a interacciones faunísticas inusuales. En Colombia, esta situación ha generado el contacto secundario entre las poblaciones de las subespecies de Ramphocelus flammigerus del Valle del Cauca y de la costa Pacífica; y se han encontrado individuos con rabadillas de colores intermedios entre las subespecies que se han catalogado como híbridos. El objetivo del presente estudio fue evaluar si existe evidencia morfológica que sugiera hibridación y que pueda explicar el origen de los individuos de coloración intermedia. Con este fin, se obtuvieron muestras de 15 localidades; 10 zonas alopátricas (cinco por cada subespecie) y cinco zonas simpátricas. Para la captura de individuos se utilizaron redes de niebla y fueron tomados siete caracteres morfológicos. Asimismo, se predijo que si las subespecies están hibridando, las mismas, podrían ser morfológicamente más similares cuando coexisten que cuando se encuentran separadas. Alternativamente, cuando las subespecies coexisten, éstas pueden divergir en simpatría debido a presiones selectivas para reducir la competencia por recursos (desplazamiento de caracteres). Para identificar los patrones de variación morfológica, se comparó la morfología de las subespecies, de poblaciones simpátricas y alopátricas de ambas subespecies y de los individuos de cloración intermedia. Consecuentemente, se realizó un análisis discriminante y se evaluaron las diferencias entre los grupos con la utilización de intervalos de confianza del 95% para las relaciones logarítmicas. Y se capturaron un total de 112 individuos (46 de coloraciones intermedias, 20 R. f. flammigerus y 46 R. f. icteronotus). Los análisis discriminantes mostraron que las subespecies se diferencian entre ellas y que los individuos de coloraciones intermedias se traslapan con estas. Las relaciones logarítmicas, basadas en las distancias cuadradas de Mahalanobis, mostraron que los individuos intermedios fueron más similares morfológicamente a ambas subespecies que las subespecies entre ellas. Adicionalmente, se encontró que las poblaciones simpátricas de ambas subespecies son similares pero las poblaciones alopátricas son diferentes, y que los individuos de coloraciones intermedias son más similares a las poblaciones simpátricas de las dos subespecies que a las alopátricas. Por lo tanto, la evidencia morfológica evidencia las predicciones derivadas de la hipótesis de hibridación entre las subespecies de R. flammigerus. En conclusión, el análisis morfológico de la variación en R. flammigerus sugiere que se presenta hibridación entre las subespecies y que probablemente está en progreso un proceso de introgresión genética.

This report describes the construction of integrated genetic maps in pearl millet involving certain purple phenotype and simple sequence repeat (SSR) markers. These maps provide a direct means of implementing DNA marker-assisted selection and of facilitating "map-based cloning" for engineering novel traits. The purple pigmentation of leaf sheath, midrib and leaf margin was inherited together 'en bloc' under the control of a single dominant locus (the 'midrib complex') and was inseparably associated with the locus governing the purple coloration of the internode. The purple panicle was caused by a single dominant locus. Each of the three characters (purple lamina, purple stigma and purple seed) was governed by two complementary loci. One of the two loci governing purple seed was associated with the SSR locus Xpsmp2090 in linkage group 1, with a linkage value of 22 cM, while the other locus was associated with the SSR locus Xpsmp2270 in linkage group 6, with a linkage value of 23 cM. The locus for purple pigmentation of the midrib complex was either responsible for pigmentation of the panicle in a pleiotropic manner or was linked to it very closely and associated with the SSR locus Xpsmp2086 in linkage group 4, with a suggestive linkage value of 21 cM. A dominant allele at this locus seems to be a prerequisite for the development of purple pigmentation in the lamina, stigma and seed. These findings suggest that the locus for pigmentation of the midrib complex might regulate the basic steps in anthocyanin pigment development by acting as a structural gene while other loci regulate the formation of color in specific plant parts.

A Pothomorphe umbellata (Piperaceae), é uma espécie medicinal nativa do Brasil, utilizada na indústria de cosméticos e protetores de pele contra raios UVA e UVB. Com o intuito de gerar informações aplicadas à propagação da espécie, o presente trabalho relacionou a coloração e a massa de sementes de P. umbellata a seu comportamento germinativo. A coloração e a massa de sementes de P. umbellata foram características adequadas para avaliar a homogeneidade fisiológica, o vigor, o potencial e o comportamento germinativo. Assim, conclui-se que, embora de germinação lenta, as sementes de coloração preta e mais densa devem ser as escolhidas quando de coleta ou de processo seletivo.


Pothomorphe umbellata (Piperaceae) is a medicinal species native to Brazil, which has been used in the cosmetic industry and in products that protect the skin against UVA and UVB rays. To generate information applied to the species propagation, the present work related the coloration and the mass of P. umbellata seeds to their germinative behavior. The coloration and the mass of seeds of P. umbellata were characteristic appropriate to evaluate physiologic homogeneity, vigor, potential and germinative behavior. Thus, although of slow germination, seeds of black and denser coloration should be chosen during collection or selective process.

RESUMO

Anopheles stephensi is an important urban malaria vector, which is widely distributed throughout the Indian subcontinent. The said vector species has developed resistance to various insecticides and therefore, it is desirable to develop alternative strategies including genetic methods for its control. One of the requirements for such strategy is to establish morphological mutants and to establish the genetic basis of the same. Such mutant markers could be used in the construction of genetically modified strain/s in the genetic control programme of An. stephensi. The dark colour expresses in all larval stages and pupae with full penetarence and uniform expression in both the sexes. The viability of the mutant is as good as the wild type. The genetic studies of the dark larva revealed that the gene da is mono-factorial, autosomal and recessive to the wild type. The gene da is an excellent marker for An. stephensi.

The effects of sublethal concentrations of a synthetic pyrethroid, beta-cyfluthrin (Bulldock 025 SC) were investigated in the F1 generations of white and sepia mutants of Drosophila melanogaster after various cross combinations of the parents which were exposed separately to the insecticide mixed diet for 24 hours. The experiment was carried out under the laboratory conditions at a temperature of 25+/-5 degrees C and 60+/-5% relative humidity. The insecticide had deleterious effect on the growth and development of both the mutant flies. Larval, pupal and total developmental periods were found to be increased as compared to controls, whereas the number of adult emerged decreased in all the treatment sets. Larval period index (LPI), pupal period index (PPI) and growth index (GI) were observed to be decreased in the entire cross combinations. Maximum effects were observed in those sets where both the sexes were treated, crossed and F1 generation was studied on the treated food having sublethal concentration of the insecticide, followed by similar treatment sets but on the fresh untreated food. Females were found to be resistant as compared to the males, whereas white mutant files were more susceptible to the test insecticide beta-cyfluthrin as compared to sepia mutant flies.

BACKGROUND & OBJECTIVES: Anopheles stephensi, an important vector of malaria continues to be distributed widely in the Indian subcontinent. This vector species has developed resistance for various insecticides. Therefore, it is desirable to develop alternate strategy, which does not involve resistance. In order to develop such strategy, it is mandatory that genetic studies of concerned vector species should be established. This paper describes the isolation and genetic studies of an eye colour mutant, ruby-eye (ru), and linkage studies involving another autosomal recessive mutant greyish brown larva (grb ru) in A. stephensi. METHODS: The stocks of mutants ruby-eye (ru), greyish brown (grb ru) and wild type mosquitoes were maintained in the laboratory. Crosses were made between the wild type and mutant to determine the mode of inheritance of ruby-eye. For linkage studies crosses were made between the mutant ruby-eye and another autosomal recessive mutant greyish brown larva. The percentage cross over was calculated for the genes linkage relationship for ru and grb ru. RESULTS: Results of crosses between mutant and wild type show that the inheritance of ruby-eye in A. stephensi is monofactorial in nature. The ru allele is recessive to wild type and is autosomal. The linkage studies showed no linkage between grb and ru. INTERPRETATION & CONCLUSION: The mutant ru represents an excellent marker for A. stephensi as it expresses in all the life stages with complete penetrance and high viability. This mutant can be used extensively to conduct basic and applied research.

RESUMO

Se realizaron 54 tratamientos de eliminación melánica fisiológica gingival; 10 pacientes mostraron repigmentación clínica leve al término de un año del tratamiento. Ocho de estos individuos resultaron ser fumadores crónicos. El método mucoabrasivo es una técnica quirúrgica sencilla sin grandes riesgos; se trató la región anterosuperior e inferior debido a que es el área de la sonrisa y afecta estética y psicológicamente.
