RESUMO

PURPOSE: To detect whether chitin and sepia ink sponge (CS) can promote wound healing and elevate impact of CS on phagocytosis ability of macrophages. METHODS: Forty-eight rats were assigned to four groups: Normal group (Normal), negative control group (Con), chitin and sepia ink sponge group (CS) and positive control Surgicel Gauze(r) group (SG). Deep second-degree burn model was created in rats. Wound area was recorded by digital imaging and determined using Image J software. Samples were collected and kept at -80oC on 3d, 7d, 14d and 21d for cytokines detecting. Transforming growth factor (TGF)-β1, interleukin (IL)-6, matrix metalloproteinase (MMP)-1, hydroxyproline (Hyp) and macrophage activity reflected by tumor necrosis factor (TNF)-α were determined by enzyme-linked immunosorbent assay (ELISA). RESULTS: Comparing to Con and SG, scabs in CS group fell off and basically healed on 21 day. TGF-β1, IL-6, MMP-1 and Hyp were significantly increased by CS and SG comparing to Con (p < 0.05), CS had more apparently adjustment on TGF-β1 and MMP-1 compared to SG; results in vitro indicated CS significantly promoted phagocytosis ability of macrophages reflected in TNF-α (p < 0.05). CONCLUSION: CS improved wound healing through exerting significant influences on secretion of kinds of cytokines and activating macrophages.

RESUMO

El ungüento QL es un ungüento rectal en fase de investigación destinado al tratamiento de afecciones rectales. La combinación por primera vez de quitina y clorhidrato de lidocaína en un producto semisólido de base oleaginosa, requiere del estudio del comportamiento de estabilidad. Para obtener información con respecto a su estabilidad se realizó un estudio en las condiciones establecidas. Se analizaron 3 lotes pilotos QL01, QL02 y QL03, almacenados a temperatura de 40 ± 2 °C. Los estudios se realizaron a tiempo 0, 1, 2, 3, 6 meses. No se observaron cambios en las propiedades organolépticas, en la estabilidad sedimentacional, ni en el área de extensibilidad. Se mantuvo el flujo pseudoplástico tixotrópico característico de este tipo de sistema al cabo de los 6 meses. Los principios activos se mantuvieron estables en las condiciones ensayadas.


QL ointment is a rectal ointment under development stage devoted to the treatment of rectal affections. The combination for the first time of quitine and lydocaine chloride in a semisolid product of oleaginous base requires the study of the stability behaviour. To obtain information on its stability, a study was carried out under the established conditions. 3 pilot batches QL01, QL02 and QL03, stored at a temperature of 40 2 C, were analyzed. The studies were conducted at 0, 1, 2, 3 and 6 months. No changes were observed neither in the organoleptic properties, nor in the sedimentation stability and the extensibility area. The pseudoplastic thixotropic flow characteristic of this type of system was maintained after 6 months. The active principles were stable under the assayed conditions.

RESUMO

Las úlceras de extremidades inferiores constituyen una patología muy común en la población, sobre todo en mujeres de edad avanzada, siendo su etiología en un alto porcentaje de origen vascular. Se demuestran los efectos de la quitina como agente cicatrizante contraponiéndola, en un estudio doble ciego (30 pacientes), aun agente tópico de uso frecuente en el manejo de esta etiología, la sulfadiazina de plata; paralelamente se propone rescatar un elemento que ha caído en desuso en el arsenal terapéutico dermatológico y que consideramos aún tiene vigencia: La Bota de Unna
