RESUMO

The chemical composition of the oil extracted from the seeds of Sapindus saponaria L., (Sapindaceae), was investigated. Cyanolipids constituted 5 percent hexane extract of the seeds, whereas triacylglycerols (TAG) accounted for 90 percent. The oil contains type III cyanolipids (CL) 1-cyano-2-hydroxymethylprop-1-en-3-ol-diesters. Structural investigation of the oil components was accomplished by chemical, chromatographic (TLC, CC, GC-MS), and spectroscopic (IR, NMR) means. GC-MS analysis showed that fatty acids were dominant in the CL components of the oil from S. saponaria L., with cis-11-eicosenoic acid, cis-11-octadecenoic acid and eicosanoic acid as the only esterified fatty acyl chains respectively. This being the first report of this kind of natural products (CL), located in the seeds of this plant.

Tetranychus urticae Koch (1836) (Acari: Tetranychidae), known as spider mite is a pest that can reduce fruit production in strawberry up to 80%. The control of the pest by synthetic acaricides has provided the destruction of beneficial organisms and development of resistant populations. As an alternative to traditional control, this study aimed to evaluate the effects of the extract from Sapindus saponaria and soap solution on the oviposition and mortality of females of T. urticae. The mites were sprayed with the aid of a Potter Tower, using formulations of extract/soap solution at 1, 2, 3, 4, 5% (w / v) and assessed at 24, 72 and 120h. It was found that the extract efficiency (LC50, 1.18%) than the soap solution (LC50, 1.31%). Regarding the ovicidal activity, the greatest effect was observed with the formulation at 5% (w / v) after 120 hours for both the extract (60.9%) as for soap solution (84%). The results observed in laboratory conditions are favorable to the use of the extract and soap solution to control T. urticae in pest management programs for the strawberry crop.

RESUMO

<p><b>OBJECTIVES</b>To investigate the protective effects of Sapindus saponins in spontaneously hypertensive rats, and the possible cellular and molecular mechanisms.</p><p><b>METHODS</b>Thirty-two 16-week-old spontaneously hypertensive rats were randomly divided into four groups (8 in each group): model group (placebo), positive control group (27 mg/kg of Captopril Tablets), Sapindus saponins groups (27 mg/kg and 108 mg/kg, respectively). Another 8 healthy Wistar-Kyoto strain (WKY) rats were used as the normal group. The animals were treated for 8 weeks. Blood pressure of rats was determined by non-invasive blood pressure meter (BP-6). Furthermore, the contents of angiotensin II (Ang II) in plasma and myocardial tissue were determined by enzyme-linked immunosorbent assay (ELISA), the gene expression of receptor angiotensin type 1 (AT1R) in aorta was determined by quantitative realtime polymerase chain reaction (qRT-PCR). The protein expression of transforming growth factor-β1 (TGF-β1) and AT1R in heart was determined by immunohistochemical staining. The protein expression of p-phosphorylation of p38 mitogen-activated protein kinase (p-p38MAPK) was determined by Western blotting. The contents of interleukin (IL)-1, IL-6 and tumor necrosis factor (TNF) in serum were determined by radioimmunoassay. And the histopathological and morphological changes of aorta and heart tissue samples were assessed semi-quantitatively by hematoxylin-eosin (HE) or Masson staining.</p><p><b>RESULTS</b>Thirty minutes after single or continuous treatment, systolic blood pressure (SBP) was reduced significantly in Sapindus saponins groups. And the contents of AngII, IL-1, IL-6 and TNF-α in serum, the expression of AT1R mRNA, p-p38MAPK and TGF-β1 were significantly suppressed dose-dependently (P<0.05 or P<0.01). With the Sapindus saponins treatment, compared with those of the model group, the cardiac and aortic pathological changes were ameliorated significantly.</p><p><b>CONCLUSIONS</b>Our findings suggest that Sapindus saponins might have protective effects in spontaneously hypertensive rats, the cellular and molecular mechanisms of which might be relevant to the regulation of inflammatory responses mediated by p-p38MAPK signal pathway based on activated Ang II and AT1R.</p>

RESUMO

Leishmaniasis and trypanosomiasis are parasitic diseases with a high infection rate, being a serious public health issue in the new world. Unfortunately, there are few available commercial drugs, poorly efficient and with increasing parasite resistance. Under these condi- tions, there is a need for new molecules to develop new and better drugs. One approach to carry out this search is using traditional medicine as information source to obtain new molecules or extracts to control these parasite diseases. Sapindus saponaria (Sapindaceae) fruit resin is used in Colombia to treat ulcers caused by Leishmaniasis. In a bioguided study, we have analyzed the in vitro effect of fruit resin, chroma- tographical fractions from fruit resin and also pure compounds against Leishmania species (L. panamensis, L. braziliensis, L. amazonensis and L. donovani) and Trypanosoma cruzi. The in vivo antileishmanial effect was established under the hamster model for cutaneous leish- maniasis by L. panamensis; refined extract of S. saponaria and pure saponins displayed high in vitro and in vivo activity as leishmanicides. In addition, extracts caused low viability on T. cruzi amastigotes. The use of the crude extract can be a good alternative against cutaneous leishmaniasis, due to its activity, reduced hemolytic effect, and easy production procedures.

The aim of this study was to evaluate the allelopathic potential of aqueous extract of Sapindus Saponaria L. (soapberry) root on diaspore germination and seedling growth of Echinochloa crus-galli (L.) Link (barnyardgrass) and Ipomoea grandifolia (Dammer) O'Donell (morningglory). The aqueous extract was prepared in a proportion of 100 g of dry plant material in 1000 mL of distilled water, resulting in 10% concentration. The dilutions were made with distilled water to 7.5, 5.0 and 2.5%. The root extract of S. saponaria did not inhibit the germination of the barnyardgrass and morningglory diaspores. The highest allelopathic activity occurred on seedlings growth, being evidenced by the high rate of abnormal seedlings, reduction in shoot length and mainly on root length. The inhibitory effect was concentration-dependent and varied with the receptor species studied.

RESUMO

Endophytic fungi inhabit vegetable tissues or organs, without causing them any harm. Endophytes can co-evolve with plant hosts and possess species-specific interactions. They can protect the plant from insect attacks and diseases, and are also able to produce substances of biotechnological interest. In folk medicine, the bark, roots and fruits of Sapindus saponaria is used to produce substances with anxiolytic, astringent, diuretic and expectorant properties, as well as tonics, blood depuratives and cough medicine. This study evaluated the diversity of endophytic fungi present in the leaves of S. saponaria L. and observed the colonization of host plants by endophytes, using light and scanning electron microscopy. We verified that these fungi are found in intercellular and intracellular spaces. The genera of some isolates of S. saponaria were identified mainly by sequencing of ITS region of rDNA and, when possible, also by their microscopic features, as follows: Cochliobolus, Alternaria, Curvularia, Phomopsis, Diaporthe and Phoma. Phylogenetic analysis showed the existence of genetic variability of the genera Phomopsis and Diaporthe and interspecific variation among the Curvularia, Alternaria and Phoma, belonging to family Pleosporaceae.

RESUMO

Extracts from the dried pericarp of Sapindus saponaria L. (Sapindaceae) fruits were investigated for their antifungal activity against clinical isolates of yeasts Candida albicans and C. non-albicans from vaginal secretions of women with Vulvovaginal Candidiasis. Four clinical isolates of C. albicans, a single clinical isolated of each of the species C. parapsilosis, C. glabrata, C. tropicalis, and the strain of C. albicans ATCC 90028 were used. The hydroalcoholic extract was bioactivity-directed against a clinical isolate of C. parapsilosis, and showed strong activity. The n-BuOH extract and one fraction showed strong activity against all isolates tested. Further column-chromatography on silica gel separation of this fraction afforded two pure triterpene acetylated saponins: 3-O-(4-acetyl-beta-D-xylopyranosyl)-(1->3)-alpha-Lrhamnopyranosyl-(1->2)-alpha-L-arabinopyranosyl-hederagenin (1) and 3-O-(3,4-di-acetyl-beta-D-xylopyranosyl)-(1->3)-alpha-L-rhamnopyranosyl-(1->2)-alpha-L-arabynopyranosyl-hederagenin (2). The structures of the compounds were based on spectral data (¹H and 13C NMR, HSQC, HMBC and MS), and on with literature. The saponins isolated showed strong activity against C. parapsilosis.

As saponinas triterpênicas isoladas (1) e (2) apresentaram forte atividade contra C. parapsilosis.

The larvicidal potential of the crude ethanolic extracts (CEE) of the stem peel of Sapindus saponaria was evaluated against Rhipicephalus sanguineus. Lethal concentrations (LC), were calculated by preparing CEE solutions at different concentrations in distilled water. Larvae fasted for 14-21 days were utilized in the bioassays, after incubation of engorged females collected from infested environments frequented by dogs in several neighborhoods of Goiânia, GO. Bioassays were performed in a specially constructed biological chamber for testing botanical acaricides, acclimatized to 27±1°C, RH>80 percent. The larvae were counted on filter paper envelopes impregnated with the solutions or distilled water and larval mortality observed after 48h. S. saponaria showed good larvicidal activity (LC50 and LC99 of 1994 and 3922ppm, respectively) and the results demonstrated its potential as a botanical acaricide and an alternative control measure for R. sanguineus.

At present, plants are the main source of obtention of most of the drugs that may be useful for treating cancer. A mixture of polysaccharides was extracted from the leaves of Argemone mexicana L. plant, also known as Cardo Santo, collected in the flowering period. The method of incorporation of tritiated Thymidine was used to determine the effect of the mixture on the cellular proliferation in human cellular lines (H125 and U1906). The antitumoral activity of this crude of polysaccharides administered alone or combined with 2 known cytostatic agents was evaluated in mice inoculated with tumoral cells of lymphocytic leukemia P-388 and sarcoma 37 and their indirect antitumoral activity was assessed in Ehrlich's ascitic tumor. In all the cases, it was calculated the increase of survival time in relation to the positive or negative pattern of comparison. The mixture of polysaccharides caused a rejection to Elrich's tumor implantation, and its administration together with 5-Fluoracyl produced a moderate increase of the antitumoral activity in the mice carriers of the Sarcoma 37 tumor. These results may be related to the possible stimulating effect of this mixture on the immune system of the animals with tumor. It is necessary to study the effect of the mixture of polysaccharides obtained from A. Mexicana species on the immune system, and the effectivity of its mixture with other cytostatic agents to determine its possible use as a chemotherapy adjuvant for the treatment of cancer.

RESUMO

Phytotherapies have offered alternative sources of therapy for migraine and gained much importance in prophylactic treatment. Sapindus trifoliatus is a medium-sized deciduous tree growing wild in south India that belongs to the family Sapindaceae. The pericarp is reported for various medicinal properties. A thick aqueous solution of the pericarp is used for the treatment of hemicrania, hysteria or epilepsy in folklore medicine. We have investigated the antihyperalgesic effects of the lyophilized aqueous extract of S. trifoliatus in animal models predictive of experimental migraine models using morphine withdrawal-induced hyperalgesia on the hot-plate test and on 0.3 percent acetic acid-induced abdominal constrictions in adult male Swiss albino mice. The extract significantly (N = 10, P < 0.05) increased the licking latency in the hot-plate test when administered ip at 10 mg/kg (6.70 ± 0.39 s in saline control vs 18.76 ± 0.96 s in S. trifoliatus-treated animals) and significantly (N = 10, P < 0.001) reduced the abdominal constrictions when administered ip at 2 and 10 mg/kg (40.20 ± 1.36 in saline control vs 30.20 ± 1.33 and 23.00 ± 0.98 for 2 and 10 mg/kg, ip, respectively, in S. trifoliatus-treated animals). Furthermore, when administered ip at 20 and 100 mg/kg, the extract significantly (N = 10, P < 0.05) inhibited the apomorphine-induced climbing behavior in mice (climbing duration 15.75 ± 5.0 min for saline control vs 11.4 ± 1.28 and 3.9 ± 1.71 min for 20 and 100 mg/kg, respectively, in S. trifoliatus-treated animals). In receptor radioligand-binding studies, the extract exhibited affinity towards D2 receptors. The findings suggest that dopamine D2 antagonism could be the mechanism involved in the antihyperalgesic activity of the aqueous extract of S. trifoliatus.

<p><b>AIM</b>To observe the alterations in the biochemical and biophysical changes in the sperm membrane during sperm maturation in male rats treated with the water extract of the fruit pericarp of S. mukorossi.</p><p><b>METHODS</b>Adult male Sprague-Dawley rats were gavaged the aqueous extract of the fruit pericarp of S. mukorossi at a dose of 50 mg/kg/d for 45 days. On day 46, the sperm parameters were observed in different sections of the epididymis and the sperm superoxide dismutase and the lipid peroxidation was determined and compared with the controls. The testis and epididymis were routinely prepared for histological examination under the light microscope.</p><p><b>RESULTS</b>No significant differences in the sperm number and morphology were observed between the control and treated groups. However, a significant inhibition (P<0.05-0.01) of sperm motility in the caput, corpus and cauda regions of the epididymis was seen in the treated group. No significant histopathological changes were found in the testis and epididymis. The important finding was that in the treated animals, the spermatozoa showed an abnormal distribution of the superoxide dismutase activity, being minimum in the caput and maximum in the corpus, which was just opposite to that of the controls.</p><p><b>CONCLUSION</b>The study provides a unique observation where the plant extract alters the sperm membrane physiology without change the testicular and epididymal morphology.</p>
