RESUMO
Os receptores Toll-Like são moléculas de superfície, presentes nas células de defesa do hospedeiro, responsáveis pelo reconhecimento deestruturas microbianas e na geração de sinais, que levam à produção de citocinas proinflamatórias essenciais para a ativação das respostasimunes inatas. Em seres humanos, dez homólogos da proteína Toll já foram identificados e assim classificados (TLR1-10) cada um com diferentes funções específicas para determinado componente microbiano. Embora os receptores Toll-Like desempenhem um papel primordial na defesa do hospedeiro contra os processos infecciosos e inflamatórios, deve haver um equilíbrio entre a ativação e inativação destes receptores para evitar uma resposta inflamatória ou imunológica excessiva, como ocorre nas doenças crônicas inflamatórias e autoimunes sistêmicas, como o lúpus eritematoso sistêmico e a artrite reumatóide. Dessa forma, diversas pesquisas neste campo têm procurado alcançar um melhor entendimento dos mecanismos de ativação dos receptores Toll-Like. O objetivo principal deste estudo é fazer uma revisão de literatura sobre os receptores Toll-Like, ressaltando a geração dos sinais e as repostas imunológicas desenvolvidas no reconhecimento das diversas estruturas microbianas por estes receptores.
ABSTRACT
Toll-like receptors are surface molecules that are present in the host?s defence cells. They play an important role in detection and recognition ofmicrobial pathogens. They generate signals that induce the production of inflammatory cytokines for activation of innate immune responses.In humans, ten Toll-like receptors have been identified and designated (TLR1-10) with different functions specific to particular microbialcomponents. Although Toll-like receptors play a key role in protecting the host against infectious and inflammatory processes, and there mustbe a balance between the activation and inactivation of these receptors to avoid an excessive inflammatory or immune response, as it occurs insystemic autoimmune and chronic inflammatory diseases, such as systemic lupus erythematosus and rheumatoid arthritis. Thus, much research in this has sought to achieve a better understanding of the mechanisms by which Toll-like receptor signalling may be regulated. Themain objective of this study is to review the literature on Toll-like receptors, emphasizing the generation of signals and the immune responsesdeveloped in recognition of microbial components for these receptors.
