As Doenças do Espectro Autístico incluem o Autismo, o Transtorno Invasivo do Desenvolvimento Sem Outra Especificação e a Síndrome de Asperger. A etiologia é bastante discutida, devido a sua variação e complexidade. São doenças presentes desde os 30 meses de idade, caracterizadas por comportamento ritualístico, fala ausente ou pouco desenvolvida, além de problemas graves de relacionamento social e deficiência mental na maior parte dos casos. Podem ocorrer isoladamente ou como parte das manifestações de afecções específicas, como doenças gênicas e cromossômicas, com a participação de praticamente todos os cromossomos. Há relatos de pacientes com alterações subteloméricas, inclusive, envolvendo a extremidade distal (subtelomérica) do braço longo do cromossomo 2 (2q37). Este estudo teve como objetivos a avaliação do cariótipo pela técnica de bandamento GTG e da região 2q37 em alta resolução de indivíduos com doenças do espectro autístico. Foram estudados 15 pacientes. Um deles (6,67%) apresentou deleção em 2q37 (região subtelomérica), quando analisado em alta resolução. Este achado não foi confirmado pela técnica Fluorescencein situ Hybridization (FISH). Portanto, a análise citogenética em alta resolução de 2q37 pode ser incluída na investigação destes pacientes, contudo, os achados devem ser confirmados por técnicas complementares.
ABSTRACT
Autistic Spectrum Disorders (ASD) include Autism, Pervasive Developmental Disorder Not Otherwise Specified (PDD-NOS) and Aspergers syndrome. The causes have been much discussed due to the variations and complexity. Onset can occur before the age of 30 months characterized by ritualistic behavior, lack of or delayed speech, as well as severe disorders of reciprocal social interactions and, in most of cases, mental impairment. ASD may occur in isolation or as a manifestation of specific diseases. Furthermore, there have been reports of patients with ASD and subtelomeric alterations, even involving the distal end of the long arm of chromosome 2, more specifically the 2q37 region. This study aimed at evaluating the karyotype using of the GTG-banding technique, and the subtelomeric region of long arm of chromosome 2 (2q37) by highresolution GTG-banding in patients with autistic spectrum disorders. Fifteen patients were studied. A deletionin 2q37 region was observed in one patient (6.67%), when analyzed in high resolution. This find was not confirmed by the FISH (Fluorescence in situ Hybridization) technique. Therefore cytogenetic analysis in high resolution of the 2q37 may be included in investigation of these patients, however, the findings may be confirmed by supplementary techniques.
