A galectina-3 pertence a uma família de lectinas endógenas que se ligam a -galactosídeos presentes na superfície celular e em glicoproteínas de matriz extracelular (ECM). No timo,essa lectina está envolvida com funções biológicas como adesão celular, migração e apoptose.Trabalhos recentes relacionaram a galectina-3 com o diabetes mellitus insulino-dependente.Essa doença é caracterizada pela destruição de células -pancreáticas mediada principalmente por linfócitos T autorreativos. Os camundongos NOD representam um modelo adequado para estudar o diabetes autoimune, pois desenvolvem a doença espontaneamente de modo similar aos humanos. Nosso grupo descreveu a presença de importantes alterações no timo destes animais como o desenvolvimento de espaços perivasculares (PVS) gigantes, caracterizados pela retenção de células T e B entremeadas por uma rede de ECM, associado a uma deficiência na expressão do receptor de fibronectina VLA-5. Com base nestes dados, o presente trabalho teve o objetivo principal caracterizar as células que expressam galectina-3 no timo, verificando sua interação com células e moléculas de ECM presentes nos PVS. Notamos um aumento significativo da colocalização da galectina-3 com células epiteliais e dendríticas corticais e medulares no timo dos camundongos NOD, em relação às células de camundongos BALB/c...
ABSTRACT
Galectin-3 belongs to a family of endogenous lectins which bind to -galactosides presentedon the cell surface and extracellular matrix (ECM) glycoproteins. In the thymus, this lectin isinvolved in biological functions such as cell adhesion, migration and apoptosis. Recent papersrelated galectin-3 to insulin-dependent diabetes mellitus. This disease is characterized bypancreatic -cell destruction mediated by autoreactive T lymphocytes. NOD mice represent asuitable model to study diabetes, as they develop the disease in a similar way as humans. Ourgroup described important thymic alterations in these mice such as the development of giantperivascular spaces (PVS), characterized by the retention of T and B cells, intermingled withan ECM network and associated with a defect in the expression of the fibronectin receptorVLA-5. Based on these results, the present work has the main objective:
to characterizegalectin-3-positive cells in the NOD mouse thymus, verifying its interaction with cells andECM molecules present in PVS. We noticed a significant enhancement of galectin-3colocalization with cortical and medullary epithelial and dendritic cells in NOD mice, inrelation to BALB/c cells. The quantitative analysis revealed a reduction of colocalization ofgalectin-3 with fibronectin and laminin, suggesting an accumulation of this lectin insidethymic microenvironmental cells...
