ABSTRACT
Objective To investigate the effect of periostin on bone remodeling and autophagy during the period of tooth replacement and to investigate the mechanism of action of periostin in regulating the balance of bone metabolism during the replacement of primary and permanent teeth. Methods Fifteen cases each of experimental and control groups were collected from the periapical periodontal disease and multiple teeth extracted for periapical adhesions in the author's hospital, the mRNA levels of periostin, bone metabolism genes, and autophagy genes were detected in the specimens by in situ hybridization.Osteoblasts were cultured in vitro, periostin was added to the culture medium of the experimental group, and the expression of bone metabolism proteins[receptor activator of nuclear factor-κb ligand(RANKL),osteoprotegerin(OPG)]and autophagy proteins(LC3,Beclin-1)in osteoblasts was detected by Western blot. Results In situ hybridization results showed that periostin expression in the experimental group was lower than that in the control group(P<0.05),RANKL,LC3,BECN1 expression and RANKL/OPG values were higher than those in the control group(all P<0.05),and no significant differences were seen in OPG expression(P>0.05);protein blotting showed that RANKL,RANKL/OPG,LC3Ⅱ,Beclin-1,LC3Ⅱ/LC3Ⅰ were lower in the experimental group than in the control group(all P<0.05),and no significant differences were seen in OPG and LC3Ⅰ than in the control group(P>0.05).Conclusion Periostin reduces inflammation-induced bone resorption by reducing RANKL expression and improving hyperautophagic state.
