Protective effect of paeoniflorin on Abeta25-35 -induced PC12 cell injury / 中国中药杂志

ABSTRACT
<p><b>OBJECTIVE</b>To investigate the protective effect of paeoniflorin on PC12 cell injury induced by Abeta25-35.</p><p><b>METHOD</b>Abeta25-35-induced PC12 cell injury model was established. The effect of cell proliferation was detected by the MTT method. We also measured the apoptosis with the FITC-Annexin V/PI staining, observed the mitochondrial membrane potential and changes in intracellular calcium using JC-1 and Fluo-3/AM staining respectively, and studied the protein expression of Bax, Bcl-2, cleaved Caspase-3 by Western blotting analysis.</p><p><b>RESULT</b>Paeoniflorin could inhibit Abeta25-35-induced PC12 cytotoxicity and apoptosis and show dose dependence. Its protective effect may be realized by increasing mitochondrial membrane potential Bcl-2 protein expression, decreasing Ca(2+) concentration and Bax protein expression and inhibiting activation of apoptosis-related protein Caspase-3.</p><p><b>CONCLUSION</b>Paeoniflorin shows a protective effect on Abeta25-35-induced PC12 cell injury.</p>
