ABSTRACT
BACKGROUND:
It is likely to find that ABCE1 plays an important part in the occurrence and progression of cancers through in-depth study.OBJECTIVE:
To investigate the effects of ABCE1 silencing on the proliferation and sensitivity toβ-eiemene of ovarian cancer stem cel s via electrotransfection.METHODS:
siRNA sequence of ABCE1 was designed and synthesized, and then was electrotransfection into ovarian cancer stem cel s. Subsequently, expressions of ABCE1 mRNA and protein in ovarian cancer stem cel s after ABCE1 silencing were detected by RT-PCR and western blot assay;the cel cycle was detected using flow cytometry;the sensitivity of ovarian cancer stem cel s toβ-eiemene after ABCE1 silencing was analyzed by MTT assay and colony-formation assay. RESULTS ANDCONCLUSION:
After ABCE1 silencing, expressions of ABCE1 mRNA and protein in ovarian cancer stem cel s were significantly reduced. And the cel cycle was arrested in G0/G1 phase, and the number of cel s in S phase significantly decreased. Furthermore, the sensitivity of ovarian cancer stem cel s toβ-eiemene after ABCE1 silencing was significantly enhanced. In conclusion, ABCE1 silencing cannot only significantly inhibit the proliferation of ovarian cancer stem cel s, but also enhance the sensitivity of cancer stem cel s toβ-eiemene.
