Effect of Dan Shen Pill on expression of Fas/FasL proteins in cultured neonatal rat cardiomyocytes during hypoxia and reoxygenation / 中国病理生理杂志

ABSTRACT

AIM:

To observe the effect of Dan Shen Pill on expression of Fas/FasL proteins in cultured neonatal rat cardiomyocytes during hypoxia and reoxygenation.

METHODS:

The neonatal rat cardiomyocytes were isolated and cultured in normal condition for 24 hours before hypoxia and reoxygenation experiments were conducted. The expressions of Fas/FasL proteins in cultured neonatal rat cardiomyocytes were determined by immunohistochemistry and positive expression index (PEI) was calculated.

RESULTS:

(1) The PEI of Fas/FasL proteins in hypoxia group was higher, but the PEI in Dan Shen Pill treated group was significantly lower than that in control. (2) Under the condition of hypoxia for 30 min then reoxygenation for 4.5 h or 10.5 h, the PEI of Fas/FasL oprotein in cultured neonatal rat cardiomyocytes was also much higher than that in control. However, in the presence of Dan Shen Pill, PEI reduced remarkably compared to cells in the absence of Dan Shen Pill under the same hypoxia and reoxygenation conditions.

CONCLUSION:

The results indicated that the enhancement of Fas/FasL protein expression emerged in the hypoxia and reoxygenation injury in cultured neonatal rat cardiomyocytes. Dan Shen Pill treatment reduced apoptosis during hypoxia and reoxygenation by decreasing the Fas/FasL protein expression.
