ABSTRACT
[Objective] To determine the contents of imperatorin and isoimperation in Angelicae dahuricae from different sources by HPLC,and prove some scientific evidence for processing standardization and quality control of Angelicae dahuricae decoction pieces.[Methods] An HPLC method was established.XB-C18 column was used, with mixture of acetonitrile and water in a gradient mode. The flow rate was 1.0mL?min-1.[Result] The calibration curve of imperatorin was in good linear over the range of 0.0552~0.276?g with the average assay recovery 99.5%.The calibration curve of isoimperatorin was linear over the range of 0.048~0.24?g with the average assay recovery 98.6% (RSD=1.7%).The contents of imperatorin from different sources in Angelicae dahuricae were between 0.046%~0.145%,isoimperatorin was between 0.038%~0.087%,the total was between 0.084%~0.232%.[Conclusion] This method was proved to be rapid, reproducible and accurate. It is suitable for the quality control of Angelilae dahuricae and its preparation.
