ABSTRACT
Objective:
To evaluate the effect of NF-? B inhibitor-PDTC on TNF-? induced cytotoxicity in breast cancer cells.Methods:
The cell survivals of breast cancer cell lines MCF-7 and MDA-MB-435s were measured by MTT assay in vitro.MDA-MB-435s cells were implanted into BALB/C mice. Animal tumor models were treated with TNF-?, PDTC and TNF-? plus PDTC.Results:
The growth of two breast cancer cell lines was not inhibited by either TNF-?(2000U/mi or less)or PDTC.However,both breast cancer cells treated with TNF-?(2000U/mi)plus PDTC(50? mol)showed significant growth inhibition as compared with controls. In vivo, the growth of implanted tumor was significantly reduced in volume in mice treated with TNF-?(7.5?10 6U/kg,ip,bid) plus PDTC(50mg/kg,ip,bid) as compared with controls( P
