Simultaneous Determination of Paeoniflorin, Tetrahydropalmatine and Osthole in Gujinwan Capsules by HPLC / 中国药师

ABSTRACT

Objective:

To develop an HPLC method for the content determination of paeoniflorin, tetrahydropalmatine and osthole in Gujinwan capsules.

Methods:

The determination was performed on an Eclipse XDB-C18 column (250 mm × 4. 6 mm,5 μm) with the mobile phase consisting of 0. 1% phosphoric acid (adjusting pH to 6. 6 with triethylamine)-acetonitrile with gradient elution. The flow rate was 1. 0 ml·min-1 . The column temperature was 30 ℃. The detection wavelengths were set at 230 nm and 280 nm. Re-sultsThe calibration curves were found to be linear within the range of 9. 56-33. 65 μg· ml-1 for paeoniflorin(r=0. 9998), 3. 65-12. 86 μg· ml-1for tetrahydropalmatine(r=0. 9999) and 5. 81-20. 45 μg· ml-1for osthole(r=1. 0000). The average recovery of paeoniflorin, tetrahydropalmatine and osthole was 98. 8%(RSD=1. 1%), 98. 4%(RSD=0. 8%) and 99. 1%(RSD=1. 4%)(n=6),respectively.

Conclusion:

The method is simple, accurate and reproducible, which can be used for the quality control of Gujinwan capsules.
