ABSTRACT
Objective:
To observe the effect of rosiglitazone on serum MMP-9 con-centration and expression of peripheral monocyte MMP-9 in rats with diabetes.Method:
Thirty SD rats were randomly divided into 3 groupscontrol group(n=10),diabetes group(n=10)and rosiglitazone group(n=10).The rat diabetic model was made by the intra- peritoneal injection of streptozocin.After the model was made successfully,the rats of rosightazone group were treated with rosiglitazone(5 mg?kg~(-1)?d~(-1)for 28d)by garage and the rats of control group and diabetes group were treated with normal saline.The concentration of serum MMP-9 were detected by Enzyme-linked Immunosorbent Assay(ELISA).The expres- sion of peripheral monocyte MMP-9 mRNA were measured by RT-PCR.Result:
The concentration of serum MMP-9 were lowered by rosiglitazone treatment.The expression of peripheral monocyte MMP-9 mRNA were inhibited by rosightazone treatment too.Conclusion:
Rosiglitazone may be stable artery atherosclerosis plaque through inhibit the mRNA expression of MMP-9.
