UPLC Fingerprint, Cluster and Principal Component Analysis of Xiao'er Chaigui Tuire Granules / 中国药房

ABSTRACT

OBJECTIVE:

To establish UPLC fingerprint of Xiao'er chaigui tuire granules, and to conduct cluster analysis (CA) and principal component analysis (PCA).

METHODS:

UPLC method was adopted. The determination was performed on Waters Acquity UPLC BEH Ci8 column with mobile phase consisted of methanol-0. 05% formic acid solution (gradient elution) at the flow rate of 0. 5 mL/min. The detection wavelength was 260 nm, the column temperature was 40 ℃, and sample size was 0. 5 μL. Using puerarin as reference, UPLC fingerprints of 14 batches of Xiao'er chaigui tuire granules were determined. The similarity of 14 batches of samples was evaluated by using TCM Chromatographic Fingerprint Similarity Evaluation System (2012 edition), and common peak was identified. CA and PCA were conducted by using SPSS 19. 0 software.

RESULTS:

There were 15 common peaks in UPLC chromatograms of 14 batches of samples. The similarity of UPLC fingerprints of 14 batches of samples was higher than 0. 90, and UPLC fingerprint was in good agreement with control fingerprint. The 14 batches of samples could be clustered into 2 categories.

CONCLUSIONS:

Established fingerprint can provide reference for quality evaluation of Xiao'er chaigui tuire granules.
