ABSTRACT
Objective:
To select the plasma miRNA (microRNA) associated with the clinical features of patients with colorectal cancer.Methods:
Differential expression of plasma miRNA was detected by TaqMan® Human MicroRNA Array from 7 patients with colorectal cancer and 6 healthy volunteers. The results of TaqMan® Human MicroRNA Array were validated by using plasma samples from 29 patients with colorectal cancer and 20 healthy volunteers by real-time fluorescence quantitative-PCR, and the relationship of differential expression with clinical features was analyzed.Results:
In the condition of fold change > 1.0 and P < 0.05, sixteen plasma-specific miRNAs were identified from patients with colorectal cancer. Hsa-miR-125b, hsa-miR-375, hsa-miR-150 and hsa-miR-206 validated by real-time fluorescence quantitative-PCR were consistent with those detected by TaqMan® Human MicroRNA Array, and the differential expression was statistically significant between colorectal cancer patients and healthy volunteers (P < 0.05). Furthermore, the expression of plasma hsa-miR-150 and hsa-miR-375 had a significant impact on tumor invasion and vascular cancer embolus.Conclusion:
Plasma hsa-miR-150 and hsa-miR-375 are expected to be non-invasive biomarkers to give assistance in tumor staging and to evaluate the prognosis of colorectal cancer. Copyright © 2012 by TUMOR.
