HPLC-DAD characteristic chromatogram of chinese eaglewood / 中国药学杂志

ABSTRACT

OBJECTIVE:

To establish the characteristic chromatogram of Chinese eaglewood by HPLC-DAD.

METHODS:

The analysis was performed on a Diamonsil C18 column(4.6 mm × 250 mm, 5 μm) with a gradient mobile phase of acetonitrile-0.1% formic acid at a flow rate of 0.7 mL · min-1. The detection wavelength was set at 252 nm and the column temperature was 30℃. Twenty-three batches of Chinese eaglewood samples were analyzed with the developed method, and similarity evaluation system for chromatographic fingerprint of traditional Chinese medicine(2004A Version) was used for the quality assessment.

RESULTS:

Seventeen common peaks were found through the similarity evaluation system and 12 peaks were confirmed by comparison with the chromatograms of the standard substances. Among them, 9 peaks were identified as characteristic peaks of Chinese eaglewood. With peak 1 (agarotetrol) as the reference, the relative retention time of the other eight peaks were 1.033 6, 1.2193, 1.2486, 1.7833, 1.891 5, 1.9184, 2.404 6 and 2.4582, respectively.

CONCLUSION:

The developed method conforms to the requirements of methodology validation, and can be used for the quality control of Chinese eaglewood.
