ABSTRACT
OBJECTIVE:
To establish the characteristic chromatogram of Chinese eaglewood by HPLC-DAD.METHODS:
The analysis was performed on a Diamonsil C18 column(4.6 mm × 250 mm, 5 μm) with a gradient mobile phase of acetonitrile-0.1% formic acid at a flow rate of 0.7 mL · min-1. The detection wavelength was set at 252 nm and the column temperature was 30℃. Twenty-three batches of Chinese eaglewood samples were analyzed with the developed method, and similarity evaluation system for chromatographic fingerprint of traditional Chinese medicine(2004A Version) was used for the quality assessment.RESULTS:
Seventeen common peaks were found through the similarity evaluation system and 12 peaks were confirmed by comparison with the chromatograms of the standard substances. Among them, 9 peaks were identified as characteristic peaks of Chinese eaglewood. With peak 1 (agarotetrol) as the reference, the relative retention time of the other eight peaks were 1.033 6, 1.2193, 1.2486, 1.7833, 1.891 5, 1.9184, 2.404 6 and 2.4582, respectively.CONCLUSION:
The developed method conforms to the requirements of methodology validation, and can be used for the quality control of Chinese eaglewood.
