STUDY ON THE INHIBITION EFFECT OF PROCYANIDINS ON PROLIFERATION OF HUMAN BREAST CANCER CELLS / 营养学报

ABSTRACT
Objective To investigate the effect of grape procyanidins (GPC) and the regulation of estrogen receptor(ER)on proliferation of human breast cancer(MCF-7)cells. Method MCF-7 cells were cultured with GPC,and divided into six groups tumor control,high,middle and low dose GPC treated,estrogen antagonist ICI 182. 780 (ICI) control,ICI and GPC combined treated. The proliferation activity level of MCF-7 cells was determined by MTT assay,and the levels of the proliferating cell nuclear antigen (PCNA) and Bax protein were measured by immunohistochemistry method. Results Compared with tumor control group,the level of proliferation activity and the expression of PCNA were significantly decreased in high dose GPC group,while the expression of Bax protein was increased significantly. Compared with high dose GPC group,both the proliferation activity and the PCNA expression were increased,and Bax protein expression decreased significantly in ICI and GPC combined treated group. Conclusion GPC can inhibit the proliferation and induce the apoptosis of human breast cancer cells effectively,and its mechanism is closely related to the regulation of ER.
