The clinical-pathogical significance and the expression of maspin mRNA and protein in human gastric carcinoma / 重庆医科大学学报

ABSTRACT

Objective:

To investigate the expression of maspin mRNA and protein in gastric cancer and the relationship between clnical pathological parameters. In order to explore their roles in tumorigenesis and progression of gastric cancer.

Methods:

Immunohistochemistry SP method was used to detemine expression of maspin in gastric adenocarcinoma,signet-ring cell carcinoma and normal gastric mucosa. The reverse transcriptase PCR was carried out to detect the level of maspin mRNA from the above mentioned 30 gastric adenocarcinoma and 20 normal samples.

Results:

The expression rate of maspin protein was 50% in gastric adenocarcinoma ,46.7% in signet-ring cell carcinoma,and 90% in normal gastric mucosa ,respectively. The relative transcription level of maspin mRNA in normal gastric mucosa was higher than that in gastric adenocarcinoma .Maspin protein expression was significantly related to the depth of invasion and lymph node metastasis of gastric carcinoma,but did not relate to tumor size and TNMstaging. The relative mRNA transcription level of maspin significantly correlated with the lymph node metastasis and the depth of invasion of carcinoma,but did not relate to TNM staging or tumor size.

Conclusion:

Down-regulated expression of maspin play an important role in gastric tumor. It may have inhibitory effects on invasion and metastasis of gastric tumor. Maspin might be objective indicator for pathobiological behaviors of gastric tumor.
