ABSTRACT
Objective:
To study the relationship between the T cell clonotypes and RA,carried out the analysis of infiltrating T cell clonotypes in foot of a murine model of spontaneous RAMethods:
RT PCR and SSCP were used for this studyResults:
The identical rate of V?2 and V?8 2 clonotypes of T cells in different foot joints is 68 1 percent and 59 2 percent separately The assay of RT PCR/SSCP showed there were identical V?2 and V?8 2 clonotypes in four foot joints of all SKG mice(100%) DNA sequencing showed that the same DNA bands in samples from different joints by SSCP were the same T cell clonotypes The assay of T cell transformation suggested that clonotypes of T cells showed up in joints is related to morbid formation of RAConclusion:
This investigation indicated that T cells of V?2 and V?8 2 TCR types probably play an important role in SKG murine RA
