ABSTRACT

Objective:

To find the possible mechanisms of endovascular radiation in the prevention of restenosis in rabbits following carotid endarterectomy and to observe the dose effect relationship.

Methods:

Twenty four rabbits were randomly divided into three groups following carotid endarterectomy (each n=8) and were allocated to receive a radiation dose of 0,10,20Gy( 32 P)respectively.Changes in the levels of plasma nitric oxide(NO) and endothelin 1(ET 1)were measured 3 day before operation and 3,7,14,28 day after operation.

Results:

The plasma NO was markedly increased and ET 1 was markedly decreased in endovascular radiation groups compared to control groups. The results were similar in the two groups(10Gy versus 20Gy).

Conclusion:

The endovascular radiation may inhibit smooth muscle cell proliferation and migration, and thus prevent restenosis by increasing NO and decreasing the level of ET 1.Changes of the levels of plasma NO and ET 1 is a practicable methods to detect the effect of therapy.
