Effect on hyaluronic acid as substitute of thrombin mixture in application of platelet rich plasma

ABSTRACT
Platelet rich plasma(PRP) is commonly used in aspect of containing autogenous source of several growth factors that could preventing from infection and enhancing bone formation capacity and easily handling to clinician. In PRP application, we had additionally applied thrombin mixture which had gained from allogeneic or xenogeneic donors is very expensive and, it could not be excluded transmission of disease from donors. This study was designed to evaluate possibility of substitution hyaluronic acid(HA) for thrombin mixture in clinical, radiographic, and histological aspects. Twenty-four rabbit were included in this randomized, blinded, prospective pilot study. In experimental group, Mandibular bone defects on which size were 6 mm diameter round shape were created and immediately were grafted with Bio-oss soaked with PRP+HA. In control group, same bone defect were prepared and grafted with Bio-oss soaked with PRP+thrombin mixture. All the specimen were sacrified at 1st, 2nd, 4th, 6th week postoperatively, proceeded for HandE and M/T staining, and examined under light microscope. All groups showed almost similiar hardness of grafted site clinically postoperatively. PRP with HA applied experimental groups showed increased radiographic bone density after 6th week postoperatively and hitologically experimental group showed better bone formation capacity such as osteoblastic activity and new bone formation since 4 week postoperatively. Therefore, HA has a sufficient potential of substitution for thrombin mixture in clinically PRP application.
