ABSTRACT

OBJECTIVE:To optimize the technical conditions of alprostadil liposome.METHODS:Alprostadil liposome was prepared by instillation technique,and technical conditions were optimized with particle size and entrapment efficiency as indexes.RESULTS:The optimized technical conditions for the preparation of alprostadil liposome were the followingthe concentration of ethanol was 12.5% at a stirring speed of 600 r?min-1;the instilling time was 45 min and the pH of the buffer solution was 4.05;the size distribution of the prepared alprostadil liposome was 60～200 nm and the entrapment efficiency was 88.0%～90.5%.CONCLUSION:The instillation method for the preparation of alprostadil liposome is simple and reliable,and the technical conditions were reasonable and feasible.
