ABSTRACT
OBJECTIVE:
To determine the content of ?-aminobutyric acid in Rhizoma Pinelliae Praeparatum.METHO-DSIn alkaline condition,amino acids reacted with phenyl isothiocyanate and resulted in the derivatives of benzene isothiocyanate-amino acid complex.A Venusil-AA amino acid analysis column(250 mm?4.6 mm,5 ?m)was used and the absorbance at 248 nm was measured.The mobile phase A is 0.05 mol?L-1 sodium acetate solution(pH6.5)and the mobile phase B consisted of acetonitrile-water(80∶20).RESULTS:
The linear range of ?-aminobutyric acid was from 2.056 ?g?mL-1 to 12.336 ?g?mL-1,and the recovery was 99.0%(RSD=0.92,n=6).CONCLUSION:
This method is simple,accurate and reproducible,and it can be used for quality control of Rhizoma Pinelliae Praeparatum.
