ABSTRACT
Objective To investigate the role of interleukin-1? (IL-1?) in regulating the gene expression of matrix metalloproteinase-13 (MMP-13) in the human hyaline chondrocyte. Methods Human hyaline chondrocytes were harvested enzymatically and cultured in DMEM supplemented with 20% fetal bovine serum, varying concentration of IL-1? was added to the medium. The MMP-13 mRNA level of passaged monolayer cell cultures of human hyaline chondrocyte was assessed by reverse transcriptase polymerase chain reaction (RT-PCR) and fluorescent quantitative PCR (FQ-PCR). Chondrocyte cDNA was amplified byGeneAmp 5700 sequence detector system with equal quantitative genomic cDNA as templates respectively.
