ABSTRACT
Objective:
To investigate the proliferation inhibitory effect of curcumin on human ovary cancer cell line SKOV 3 and the mechanisms.Methods:
The MTT method was used to assay the biologic activities of curcumin at different time and in different doses.The cell cycle distribution was detected by flow cytometric analysis.The cell ultrastructure was observed by electron microscope.The expression of apoptosis-associated genes protein was detected with immunohistochemical technique.Results:
Curcumin could effectively inhibit SKOV 3 in a dose- and time-dependent manner.IC 50 of curcumin to SKOV 3 was 79.96?mol/L in 72 hours.The cell growth was arrested at G 2/M stage.Curcumin could lead to apoptosis.When treated with curcumin for 72 hours,the expression of MTP53 and Bcl-2 were down-regulated,while the expression of Bax and Fas were up-regulated.Conclusion:
Curcumin can inhibit the proliferation of SKOV 3.The apoptosis is related to the down-regulation of MTP53 and Bcl-2 and up-regulation of Bax and Fas.
