Effect of angiogenesis inhibitor on human gastric cancer xenotransplanted model of nude mice / 中华消化杂志

ABSTRACT
Objective To study the effect of an novel angiogenesis inhibitor, NM-3, on primary tumor growth of human gastric cancer in vivo, and to examine apoptosis of gastric cancer cells induced by NM-3. Methods We established xenotransplanted model of a human gastric cancer cell SGC-7901 in the dorsal skin of nude mice. NM-3 was injected peritoneally at dose of 10 mg/kg, 20 mg/kg or 40 mg/kg every other day for 5 weeks, combined with carboplatin (5 mg/kg) every third day for 4 weeks. As controls of combined treatment, the another two groups of mice were injected with NM-3 or carboplatin alone. Seven weeks after implantation, tumor size, tumor growth inhibition rate, and apoptotic index (AI) were evaluated respectively after sacrifice of the mice. Results Compared with the untreated controls, growth of the tumor implanted subcutaneously was significantly reduced in size in the mice treated with NM-3 10 mg/kg, 20 mg/kg, or 40 mg/kg , with an inhibition rate of 23.0%,40.6% and 56.2%, respectively. The AI increased significantly in the mice treated with NM-3 combined with carboplatin (1.37%?0.19% versus 6.96%?0.65%, 10.65%?1.43%, and 21.66%?2.96% ). Conclusions NM-3, as an angiogenesis inhibitor, can induce apoptosis in gastric cancer and has strong inhibitory effect on tumor growth of human gastric cancer implanted in nude mice.
