Protective effect of spleen polypeptide in cyclophosphamide-induced myelosuppression in mice / 国际药学研究杂志

ABSTRACT

Objective:

To investigate the effect of spleen polypeptide on myelosuppression induced by cyclophosphamide in mice.

Methods:

The mice were randomly divided into 6 groupsthe normal control group,model group,spleen polypeptide〔25, 50,100 mg/ (kg•d)〕groups and positive drug IL-11〔250 μg/ (kg•d)〕group. Except the normal group,the other groups were intra- peritoneally injected with cyclophosphamide〔100 mg/ (kg•d)〕on the first,fourth,and seventh days. At the same time,the spleen polypeptide dose group was administered by tail vein injection every day. Positive drug group was given IL-11 by daily subcutaneous in- jections for 10 days. The organ index of mice,peripheral blood,bone marrow DNA content,number of nucleated cells,and pathologi- cal structure were measured.

Results:

Compared with the model group,the spleen polypeptide treatment group could improve the mental state and hair suppleness of myelosuppressed mice. Compared with the model group,the liver index of the spleen polypeptide 25,50 and 100 mg/kg groups were increased from 51.02% to 63.23%,60.06% and 59.93% respectively(all P<0.01),and the spleen index were increased from 2.10% to 6.35%,5.32% and 3.80% respectively(all P<0.01). Compared with the model group, the plasma WBC levels of the spleen polypeptide 25 and 100 mg/kg groups were increased from and 2.01×10 9 /L(P<0.01),the plasma RBC levels of the spleen polypeptide 50 and 100 mg/kg groups were increased from 7.06×10 12 / L to 8.40×10 12 /L and 8.00×10 12 /L(all P<0.01),the plasma HGB levels of the spleen polypeptide 50 and 100 mg/kg were increased from 103.25 g/L to 130.80 g/L and 125.86 g/L(all P<0.01),and the plasma PLT levels of the spleen polypeptide 100 mg/kg group was increased from 423.50×10 9 /L to 1285.40×10 9 /L(P<0.01). Compared with the model group,the DNA content of the bone marrow cells of the spleen polypeptide 50 and 100 mg/kg groups increased from 6.69 μg to 13.47 μg and 17.95 μg(all P<0.01),and the number of nucleated cells in the bone marrow cells increased from 1.82×10 9 /L to 6.38×10 9 /L and 9.90×10 9 /L(all P<0.01). Conclu- sion Spleen polypeptide could improve the bone marrow suppression induced by cyclophosphamide in mice.
