ABSTRACT

Since the coronavirus disease 19 (COVID-19) outbreak, the epidemiological analysis has raised a strong requirement for more effective and accurate contact tracing solution. However, the existing contact tracing solutions either lacked the evaluation of tracing proximity or the features used for the tracing proximity evaluation were susceptible to certain negative environmental factors (e.g., body shielding). In this article, we propose a novel distance estimation algorithm based on machine learning for contact tracing: DISTERNING, where we leverage machine learning algorithms including Learning Vector Quantization, Regression, and Deep Feed-forward (DFF) Neural Network, data processing methods, and digital filters to process the Bluetooth signal information collected by the mobile phone for contact distance estimation. A contact tracing scheme based on edge computing is also proposed for algorithm deployment due to the requirements of the computational power. Compared with the existing contact tracing solutions, our algorithm considers the factors that have significant negative influence on the Bluetooth signal for distance estimation in reality. The evaluation results show that when the collected Bluetooth signal is influenced by real-world negative environmental factors, employing our proposed algorithm DISTERNING can keep the accuracy of the estimated distance reliable. The output distance can be combined with some medical models to conduct infection risk assessments. IEEE

ABSTRACT

In order to decrease the infection of COVID-19 virus, people must frequency wash the hands or decrease the times of touching the public things. The blood pressure (BP) and weight are the important parameters under the healthy management. The goal of this study is to measure BP with a weight scale. The Ballistocardiogram and photoplethysmogram (PPG) were measured to extract the pulse transit time (PTT) that was used to estimate BP. The results show that the performances of proposed method were close to the reference method, using the electrocardiogram and PPG. Thus, the proposed method can decrease the infected risk when measuring BP with the cuff. © 2022 IEEE.

Background: Nurse is a high-risk groups work fatigue feeling, which seriously affects the quality of conventional work efficiency and bureden pressures for contradiction between nurses and patients especially during the COVID-19 pandemic.Normalized epidemic prevention and control during the preview triage nurse need to all patients to the hospital and the accompanying personnel carries on the preliminary screening.COVID-19 fxed point hospital preview triage nurse with an infected person contact, more prone to anxiety,depression, results in the decrease of efficiency, to treat the service object formulation work sense of fatigue performance, etc. Objectives: To explore the influencing factors of work burnout of pre-test and triage nurses under normal epidemic prevention and control. Methods: A total of 110 pre-test and triage nurses from 4 Grade-A hospitals in Shanxi Province were enrolled in this study. The general data questionnaire, Nurse Job Burnout Scale, Pittsburgh Sleep Quality Index Scale, Self-Rating Anxiety Scale and Self-Rating Depression Scale were investigated towork burnout of pre-examination and triage nurses. Comparison between groups using two Independent sample t-test and single factor variance analysis. Multiple regression were applied to analysis factors affecting nurse fatigue feeling dimensions by SPSS22.0. P values<0.05 were considered signifcant. Results: As shown in Table 1, different professional title, department, and the sleep quality of preview triage nurses emotional exhaustion dimension com-parison(P<0.001), different department nurses to personalized level dimension comparison(P<0.05), nurse personal accomplishment dimension comparison of different cultural levels(P<0.05). Professional title, working department, sleep quality and educational level were the influencing factors of job burnout of pretest and triage nurses. Conclusion: In the COVID-19 epidemic, managers should pay more attention to the main factors that affect the sense of exhaustion of pre-test and triage nurses, and take targeted intervention measures to alleviate the sense of exhaustion of nurses, so as to ensure the safety of nursing.

ABSTRACT

Stimulator of interferon genes, namely STING, an adaptor protein located in the endoplasmic reticulum, has been recognized as a shining target for cancer and infection research. However, STING agonists cyclic dinucleotides (CDNs) have shown almost zero efficacy in phase I clinical trials as a monotherapy, likely due to poor cellular permeability and rapid diffusion despite intratumoral injection. These deficiencies further affect other applications of CDNs, such as pandemic SARS-CoV-2 prevention and therapy. Here, we rationally design a supramolecular cytosolic delivery system based on controllable recognition of calixarene, namely CASTING (CAlixarene-STING), to improve CDN druggability, including degradation stability, cellular permeability, and tissue retention. CASTING efficiently enhances the immunostimulatory potency of CDG(SF) [a chemically modified cyclic di-GMP (CDG)] to generate an immunogenic microenvironment for melanoma regression, anti-PD-1 response rate increase, and durable memory formation against tumor recurrence. More importantly, CASTING displays a superior adjuvant activity on SARS-CoV-2 recombinant spike/receptor binding domain vaccines, inducing robust and coordinated T-cell and antibody responses against SARS-CoV-2 infection in vivo. Collectively, the CASTING design represents an innovative advancement to facilitate the clinical translational capability of STING agonists. [GRAPHICS] .

ABSTRACT

Jinhongtang granule (JHT) is a traditional Chinese medicine formula used for treatment of infection diseases including severe COVID-19. However, pharmacokinetics of JHT was unknown, especially in infection condition. A liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed to simultaneously quantify ten active components form JHT in rat plasma. MS detection was performed by MRM scanning operating in the negative ionization mode. The method showed good linearity (r > 0.997). The accuracy, precision, matrix effect, recovery and stability were all satisfactory with current criterion. The method was successfully applied to compare the pharmacokinetic difference between normal and sepsis rats. The pharmacokinetic behaviors of analytes in sepsis rats were significantly different from those in normal rats. Cmax and AUC of rhein, emodin, aloe emodin, rhein-8-glucoside, aloe emodin 8-glucoside, protocatechuic acid, epicatechin and salidroside, were significantly increased in sepsis rats, except for 4-hydroxycinnamic acid and ferulic acid. In vitro intestinal absorption study using everted intestinal sac preparations indicated that the intestinal permeability was altered under sepsis. In conclusion, pharmacokinetic difference of JHT between normal and sepsis rats were evaluated for the first time, which provided useful information for the clinical application of JHT as an integrative therapy for severe and critical COVID-19.

ABSTRACT

This paper explores the possibility of combining sensory data of multiple individuals into a collective visualization. Using a smart cushion for office chairs that collects several stress-related parameters, namely: heart rate, respiratory rate, and heart-rate variability, individuals’ data can be aggregated into a collective stress visualization. Three different visualizations are designed which ly, grouped and aggregated, and metaphorically visualize the collective stress. Additionally, two more visualizations are explored for the ‘new way of working’ during the COVID-19 epidemic, where people work remotely and from the office. Through expert and user interviews, these visualizations are evaluated. Additionally, there is researched on whether measured heart-rate variability can predict perceived stress levels. ABSTRACT

Objective To analyze the genome characteristics and variations in nucleotides and amino acids of SARS-CoV-2 causing an outbreak in Henan Province in November 2021 and perform the traceability analysis.Methods In this study, throat swab specimens from cases in the acute phase were collected and tested for the nucleic acids of SARS-CoV-2 by real-time fluorescent RT-PCR.SARS-CoV-2 nucleic acid-positive samples were subjected to high-throughput genome sequencing and whole-genome alignment analysis.Results The median Ct values of ORF1ab gene and N gene in 70 positive specimens was 26.41 (15.58 to 39.27) and 24.43 (12.04 to 39.74), respectively.Compared with the sequence of Wuhan-Hu(NC_045512) reference strain, 47 to 49 nucleotide mutations sharing 47 nucleotide mutation and 41 amino acid mutations were found in 63 strains of successfully sequenced SARS-CoV-2.Nine nucleotide mutations and 12 amino acid mutations were found in the spike protein.The index case shared 47 mutations with the Russian imported cases in Henan Province on October 14 and the local cases in Jiangxi Province in October.Moreover, their genomes were highly homologous and they all belonged to the Delta variant (AY.122 evolutionary branch).Conclusions Continuous monitoring of imported COVID-19 cases and prolonging the period of quarantine were needed to reduce the risk of local outbreak and epidemic caused by imported COVID-19 cases.Analysis of the genomic characteristics of SARS-CoV-2 and the variations in nucleotides and amino acids was conducive to trace the origin of COVID-19 outbreak quickly and provide reference for precise control.

Academic advising plays an important role in the process of international student learning in a new culture. The article discusses the COVID-19 impacts on international student learning and the growing needs for academic support at the Dongbei University of Finance and Economics. By reviewing the literature on academic advising services and conducting an academic advising service needs analysis, the university builds an academic guidance system to support international student academic development during the COVID-19 pandemic. The survey was conducted to explore international students’ satisfaction levels with the academic guidance system. In this article, we present the results of the satisfaction survey, its effects and actions and offer suggestions for further improvement of academic advising services. © Journal of International Students.

ABSTRACT

Chest radiographs clearly present the characteristics of lung lesions in patients with new coronary pneumonia, thus they can be leveraged to build a new coronary pneumonia detection model to provide doctors with favorable auxiliary diagnosis results. This paper proposes a COVID-19 localization and identification approach based on yolov5 and EfficientNet. Due to inherent reasons such as computational complexity and network structure, the features of a single model are usually limited in representation, and EfficientNet provides yolov5 with competitive feature expression through BiFPN and other advantages, and the ensemble of EfficientNet and yolov5 recognition results will significantly improve the performance of a single model. In order to evaluate the robustness of the approach proposed in this paper, we trained our network on COVID-19 Detection datasets from Kaggle platform. Evaluations and comparisons demonstrate that the ensemble approach achieves better performance on various scenes. © 2022 IEEE.

ABSTRACT

After the first large-scale outbreak of pneumonia (COVID-19) caused by novel coronavirus (SARS-CoV-2) infection in December 2019, it was declared a global pandemic by the World Health Organization in March 2020. Significant infectivity and lethality rate pose a huge threat to the life and safety of all mankind, and we urgently need to grasp the infection/transmission behavior of the virus at the molecular level. Novel coronavirus and the 2003 outbreak of severe acute respiratory syndrome coronavirus (SARS-CoV) belong to bat SARS-like coronavirus (SARSR-COV) species, and their surface spike glycoproteins are highly homologous. The key step of their invasion into human cells is the binding of the receptor binding domain (RBD) part of the surface spike protein and human angiotensin converting enzyme 2(ACE2). Many experimental studies have reported that SARS-CoV-2 possesses a stronger binding ability than SARS-CoV. In order to explain the experimental results, molecular dynamics simulations, steered molecular dynamics simulations and umbrella sampling were performed on the binding of wild-type RBD (RBDWT) of the two viruses and ACE2. The results showed that different behaviors of the β-loop region lead to differences in key residues and interactions, and further result in differences in binding/unbinding mechanisms. The binding affinity of SARS-COV-2RBD and ACE2 is nearly 30 times that of SARS-COV-RBD. © 2022. All rights reserved.

ABSTRACT

COVID-19 pandemic captured the full attention of the world in 2020, and the government declared a series of non-pharmacological interventions (NPIs) to curb the influence of social movement on transmission. In different countries, different policies bring about different results. Quantifying the effect of the movement becomes a vital issue for evaluating the effectiveness of these actions. The transmission rate changes and is hard to computer after altering activity. Therefore, this research sets some European countries as the research objects, collects mobility data and daily cases during some periods, and proposes a mobility-susceptible-exposure-infectious-recovery (M-SEIR) model. Unlike the SEIR model, the movement change is quantified as a variable (σ(t)) and added in the M-SEIR model. With random sampling to get the number of people in different initial states, this research iterates the model. The iterative filtering ensemble adjustment Kalman filter (IF-EAKF) is used to adjust the subsequent iterative results. In the research, it receives the changing trend of parameters and the daily new estimation in the end. Set the first round as the fitting period and repeat the experiment 100 times in the fitting part. The result confirms the feasibility and robustness of the model. In addition, this study makes a reasonable forecast for European countries about the second round. By controlling the strength and the time point of applying non-pharmacological interventions, the research predicts the impact of these actions on the pandemic and provides some suggestions for the deployment of relevant policies in the future. Finally the study eliminates the external factors such as motion and temperature, and obtains an interesting discovery: Despite the daily case in the third round higher than that in the first round, the transmission parameter in the former appears lower than that in the latter. ABSTRACT

The pandemic of COVID-19 threatens the health and safety of the people all over the world. COVID-19 vaccine is the key public product to establish population immune barrier and achieve the global contain of the pandemic. The World Health Organization, the Global Alliance for Vaccines and Immunization and the Coalition for Epidemic Preparedness Innovations established COVID-19 Vaccines Global Access Facility (COVAX) in 2020, aiming to enable the fair access to COVID-19 vaccine by all countries in the world, especially the low- and middle-income countries. Although COVAX has facilitated the production and research of COVID-19 vaccine by coordinating the global supply chain, the implementation of COVAX is still facing many difficulties in financing, implementation and the awareness of public, revealing the problems of global health governance. Taking COVAX as an example, this paper analyzes the difficulties faced by global health governance and explore the underlying causes, so as to suggest feasible short and long-term paths for China's participation in global governance.

ABSTRACT

Coagulopathy is associated with both inflammation and infection, including infection with the novel SARS-CoV-2 (COVID-19). Endothelial cells (ECs) fine tune hemostasis via cAMP-mediated secretion of von Willebrand factor (vWF), which promote the process of clot formation. The e xchange p rotein directly a ctivated by c AMP (EPAC) is a ubiquitously expressed intracellular cAMP receptor that plays a key role in stabilizing ECs and suppressing inflammation. To assess whether EPAC could regulate vWF release during inflammation, we utilized our EPAC1 -null mouse model and revealed an increased secretion of vWF in endotoxemic mice in the absence of the EPAC1 gene. Pharmacological inhibition of EPAC1 in vitro mimicked the EPAC1 -/- phenotype. EPAC1 regulated TNFalpha-triggered vWF secretion from human umbilical vein endothelial cells (HUVECs) in a phosphoinositide 3-kinases (PI3K)/endothelial nitric oxide synthase (eNOS)-dependent manner. Furthermore, EPAC1 activation reduced inflammation-triggered vWF release, both in vivo and in vitro . Our data delineate a novel regulatory role of EPAC1 in vWF secretion and shed light on potential development of new strategies to controlling thrombosis during inflammation. KEY POINT: PI3K/eNOS pathway-mediated, inflammation-triggered vWF secretion is the target of the pharmacological manipulation of the cAMP-EPAC system.

This paper analyzed clinical studies, systematic reviews and clinical practice guidelines on the treatment of COVID-19 with antiviral drugs. It elaborated on the evidence ecosystems in these studies following the "efficacy, evidence-based practice, effectiveness" (3E) model that considered five elements: research hypothesis, research evidence, big data from healthcare facilities, real-world data, and real-world evidence. Finally, this paper summarized the experience in the production, transformation and application of evidence. This paper could help clinicians conduct highquality clinical research and provide good clinical practice based on the best currently available evidence.

The diagnosis of COVID-19 has become a highly focused research area that captures researchers' attention worldwide. Although the results of RT-PCR have been regarded as the golden standard for diagnosing COVID-19, CT-based diagnostic systems also have their unique advantages, attracting numerous researchers continuously into the area of developing deep learning-based diagnostic systems that utilize CT images. This paper is committed to presenting a comprehensive review, including current dynamics, generalized framework and useful resources. To capture the pattern of the developed methods, this paper introduces a generalized framework containing two stages: segmentation and classification. Furthermore, various valuable online resources have also been collected to provide more datasets, existing implementations of diagnostic systems, and commonly adopted evaluation metrics to researchers that are new to this area for their better adaptation and contribution to this meaningful, life-changing field. © 2021 IEEE.
