ABSTRACT

Bacterial drug resistance has become a global public health threat, among which the infection of carbapenem-resistant Enterobacterales (CRE) is one of the top noticeable issues in the global anti-infection area due to limited therapy options. In recent years, the prevalence of CRE transmission around the world has increased, and the transmission of COVID-19 has intensified the situation to a certain extent. CRE resistance can be induced by carbapenemase, porin, efflux pump, penicillin-binding protein alteration, and biofilm production. Deletion, mutation, insertion, and post-transcriptional modification of corresponding coding genes may affect the sensitivity of Enterobacterales bacteria to carbapenems. Clinical and laboratory methods to detect CRE and explore its resistance mechanisms are being developed. Due to the limited options of antibiotics, the clinical treatment of CRE infection also faces severe challenges. The clinical therapies of CRE include single or combined use of antibiotics, and some new antibiotics and treatment methods are also being developed. Hence, this review summarizes the epidemiology, resistance mechanisms, screening and clinical treatments of CRE infection, to provide references for clinical prevention, control and treatment of CRE infection. © 2022 Elsevier GmbH

ABSTRACT

The 3C-like protease (3CLpro) is an essential enzyme for the replication of SARS-CoV-2 and other coronaviruses and thus is a target for coronavirus drug discovery. Nearly all inhibitors of coronavirus 3CLpro reported so far are covalent inhibitors. Here, we report the development of specific, noncovalent inhibitors of 3CLpro. The most potent one, WU-04, effectively blocks SARS-CoV-2 replications in human cells with EC50 values in the 10-nM range. WU-04 also inhibits the 3CLpro of SARS-CoV and MERS-CoV with high potency, indicating that it is a pan-inhibitor of coronavirus 3CLpro. WU-04 showed anti-SARS-CoV-2 activity similar to that of PF-07321332 (Nirmatrelvir) in K18-hACE2 mice when the same dose was administered orally. Thus, WU-04 is a promising drug candidate for coronavirus treatment.

ABSTRACT

The high nutritional value and diverse functional properties of egg yolk proteins have led to its widespread use in the fields of food, medicine, and cosmetics. Various extraction methods have been reported to obtain the proteins from egg yolk, however, their utilization is limited due to the relatively low extraction efficiency and/or toxic solvents involved. Several simpler and greener technologies, especially physical fields (ultrasound), have been successfully developed to improve the extraction efficiency. The egg yolk proteins may exert multiple biological activities, enabling them to be a promising tool in improve human health and wellbeing, such as anti-obesity, anti-atherosclerosis, anti-osteoporosis, diagnosis and therapy for SARS-CoV-2 infections. This article summarizes the novel extraction technologies and latest applications of the egg yolk proteins in the recent 5 years, which should stimulate their utilization as health-promoting functional ingredients in foods and other commercial products. © 2022, The Korean Society of Food Science and Technology.

ABSTRACT

This study explored the degradation behavior of three types of disposable face masks in simulated seawater via the accelerated aging experiments. Microplastics (MPs) and dissolved organic carbon (DOC) were monitored in UV- and thermal-treated mask suspensions and their concentrations increased slowly in the early stage at 50 ℃ and 58 ℃. Owing to the high energy supply, the release rates of MPs and DOC at 76 ℃ were much faster than the above two temperatures. The time-temperature superposition principle (TTSP) was used to superpose the MPs/DOC release kinetics from other tested temperatures to the reference temperature and its applicability was verified by the similar activation energy. Then, a release kinetics model was established and fitted well with the superposed MP data (R2 ≥ 0.96). Since less than 0.1 % of carbon was leached, the superposed DOC data was roughly modelled by the exponential function (R2 ≥ 0.90). According to the TTSP and the established kinetics models, about 15 years were estimated to decompose half of a certain marine mask waste, together with leaching 0.21 ± 0.02 mg∙g-mask−1 of DOC. If mask consumption remains the same before 2025, they would contribute 40000–230000 tonnes of MPs and 13–97 tonnes of DOC to the ocean by 2040. © 2022

ABSTRACT

Background: Mental health is a global issue requiring global attention. Depression and anxiety are two of the most common mental disorders (CMDs) and are characterized by high incidence and high comorbidity. In recent years, the prolonged COVID-19 pandemic and exacerbated social instability have posed significant challenges to the mental resilience and mental health outcomes of the global population. Now more than ever, with an increase in mental health needs, it has become even more crucial to find an effective solution to provide universal mental healthcare. Psychotherapy is of vital importance for those coping with symptoms of depression and anxiety and is used to enhance mental resilience. However, such therapy can be difficult to access in reality. In this context, the Micro-Video Psychological Training Camp (MVPTC) platform will be developed. Objectives: As an online self-help platform for psychological intervention, the MVPTC platform was developed for those who suffer from mild to moderate symptoms of depression and/or anxiety and is tasked with the goal of reducing depressive and anxious symptoms while improving mental resilience. Thus, this study will be carried out to verify its efficacy and applicability. Methods: In this parallel-group, randomized controlled trial, a total of 200 mild to moderately depressed and/or anxious adults seeking self-help will be randomly recruited and assigned to either the micro-video psychological intervention group or the wait list control group. Online measurements by self-assessment will be taken at baseline, post-intervention, 1-month, and 3-month follow-up. Results: The primary results will involve symptoms of depression and anxiety. The secondary results will involve mental resilience. An analysis will be conducted based on the intention-to-treat principle. Discussion: This trial will examine whether the MVPTC platform for the relief of symptoms and the enhancement of resilience in a population screened for depression and anxiety symptoms proves effective and applicable. Large-scale resilience enhancement may benefit public mental health in terms of preventive interventions, managing depressive and anxiety symptoms, and promoting mental health. With the MVPTC-based method being applied, a brief, efficient, and structured intervention model can potentially be established, having the potential to provide necessary and accessible mental support for an extensive target group. Background The worldwide COVID-19 pandemic has turned into a global catastrophic public health crisis,and the conclusion about the risk factors of hospital death in COVID-19 patients is not uniform. Objective To explore risk factors of in-hospital death in patients with COVID-19 by a meta-analysis. Methods Case-control studies about risk factors of in-hospital death in COVID-19 patients were searched from databases of the Cochrane Library,ScienceDirect,PubMed,Medline,Wanfang Data,CNKI and CQVIP from inception to October 1,2021. Literature screening,data extraction and methodological quality assessment were conducted. Meta-analysis was performed using Stata 15.1. Meta-regression was used to explore the potential sources of heterogeneity. Results Eighty studies were included which involving 405 157 cases〔349 923 were survivors(86.37%),and 55 234 deaths(13.63%)〕,that were rated as being of high quality by the Newcastle-Ottawa Scale. Conclusion The risk of in-hospital death due to COVID-19 may be increased by 25 factors(including being male,older age,dyspnoea,fatigue,obesity,smoking,stroke,kidney disease,cardiovascular disease,hypertension,diabetes,cancer,pulmonary disease,liver disease,elevated levels of white blood cells,neutrophil count,total bilirubin,aspartate transaminase,alanine transaminase,lactate dehydrogenase,blood urea nitrogen,serum creatinine,C-reactive protein,interleukin-6,and erythrocyte sedimentation rate),and may be decreased by 13 factors(including myalgia,cough,vomiting,diarrhoea,headache,asthma,low body mass index,decreased lymphocyte count and platelet count,increased D-dimer,longer prothrombin time,lower albumin,increased procalcitonin and cardiac troponin). ABSTRACT

As the world's largest trading bloc, the agreement of RCEP, which was formalized in September 2020, is believed to play a non-neglectable role in the post-pandemic recovery. Real economies and the capital markets of the participating countries will have greater interactions due to tariff reduction and negative lists. By looking into the shocks in early 2020 that affect the stock markets of RCEP participating countries, we measure the stock market reaction to common risks just before the RCEP agreement was formalized. Following return-based, volume-based and liquidity-based event-study approaches, we use daily data from 11 Asia-Pacific countries to examine the stock market reactions. We find that RCEP economies for which the agreement took effect on January 1st, 2022 showed better risk resistance in response to COVID-19 shocks. In the long run, trading benefits brought by the RCEP agreement are expected to form and strengthen a system of circular flow of international trading activities among the participating countries, which will in turn increase the risk resistance ability of their stock markets. © 2022 Elsevier Inc.

ABSTRACT

The current study investigates the mental health condition of Mainland Chinese in Canada and identifies the associated sociodemographic and COVID-19-related predictors. A sample of 471 Mainland Chinese aged 18 or older completed an online survey that collected information on demographics, experience, cognition, and behaviours related to the COVID-19 pandemic and mental health condition. Mental health condition was assessed with the Depression, Anxiety, and Stress Scale-21 (DASS-21) for the depression, anxiety, and stress levels of Mainland Chinese during the pandemic. Moderate to severe depression, anxiety, and stress levels were respectively reported by 11.30%, 10.83%, and 5.10% of respondents. Univariate analysis of variance models (ANOVAs) were conducted to assess mental health condition variance as stratified by independent sociodemographic-or COVID-19-related explanatory variables, to identify possible predictors to be entered into the subsequent regression models. The regression models identified age, income level, health status, and perceived discrimination as significant sociodemographic predictors (absolute value of beta s = 1.19-7.11, ps < 0.05), whereas self-infection worry, attitude towards Canadian measures, information confusion, food/goods stocking, and room cleaning/sanitizing were identified as significant COVID-19-reltaed predictors (absolute value of beta s = 1.33-3.45, ps < 0.05) for mental health outcomes. The results shed light on our understanding of the major factors associated with the mental health condition of Mainland Chinese in Canada during the COVID-19 pandemic.

ABSTRACT

Targeting the interaction between the spike protein receptor binding domain (S-RBD) of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and angiotensin-converting enzyme 2 (ACE2) is a potential therapeutic strategy for treating coronavirus disease 2019 (COVID-19). However, we still lack small-molecule drug candidates for this target due to the missing knowledge in the hot spots for the protein-protein interaction. Here, we used NanoBiT technology to identify three Ginkgolic acids from an in-house traditional Chinese medicine (TCM) library, and they interfere with the S-RBD/ACE2 interplay. Our pseudovirus assay showed that one of the compounds, Ginkgolic acid C17:1 (GA171), significantly inhibits the entry of original SARS-CoV-2 and its variants into the ACE2-overexpressed HEK293T cells. We investigated and proposed the binding sites of GA171 on S-RBD by combining molecular docking and molecular dynamics simulations. Site-directed mutagenesis and surface plasmon resonance revealed that GA171 specifically binds to the pocket near R403 and Y505, critical residues of S-RBD for S-RBD interacting with ACE2. Thus, we provide structural insights into developing new small-molecule inhibitors and vaccines against the proposed S-RBD binding site. © 2022

ABSTRACT

The ongoing SARS-CoV-2 pandemic represents a brutal reminder of the continual threat of mucosal infectious diseases. Mucosal immunity may provide robust protection at the predominant sites of SARS-CoV-2 infection. However, it remains unclear whether respiratory mucosal administration of DNA vaccines could confer protective immune responses against SARS-CoV-2 challenge due to insurmountable barriers posed by the airway. Here, we applied self-assembled peptide-poloxamine nanoparticles with mucus-penetrating properties for pulmonary inoculation of a COVID-19 DNA vaccine (pSpike/PP-sNp). The pSpike/PP-sNp not only displays superior gene transfection and favorable biocompatibility in the mouse airway, but also promotes a tripartite immunity consisting of systemic, cellular, and mucosal immune responses that are characterized by mucosal IgA secretion, high levels of neutralizing antibodies, and resident memory phenotype T-cell responses in the lungs of mice. Most importantly, immunization with pSpike/PP-sNp completely eliminates SARS-CoV-2 infection in both upper and lower respiratory tracts and enables 100% survival rate of mice following lethal SARS-CoV-2 challenge. Our findings indicate PP-sNp is a promising platform in mediating DNA vaccines to elicit all-around mucosal immunity against SARS-CoV-2. © 2022 Elsevier Ltd

ABSTRACT

Problem, research strategy, and findings: Many towns and cities have reallocated street space in response to the COVID-19 pandemic. This was done to allow more social distancing for physical activity and to provide space for restaurants to offer outdoor dining. We used data collected via an online Qualtrics panel of New Jersey residents (n = 1,419) to evaluate how these street closures were viewed. Did people take advantage of the opportunity to dine outside? Was walking more attractive with more space? How did respondents feel about changes in traffic patterns due to the closures? Did people walk more frequently? Results suggested that there was substantial public support for these types of interventions that allowed for more walking and more lively town centers (about 40%–45% of respondents expressed support, and only 35% had negative views;a large share was neutral). Those with negative views believed that street closures increased congestion and that outdoor dining made it more difficult to walk. Takeaway for practice: In New Jersey, many towns are considering making COVID-inspired street changes permanent. There is broad support for this, though transportation agencies remain an impediment. We suggest that the current groundswell of support for street changes represents a rare opportunity to implement street design changes that support pedestrians and outdoor activities. © 2022 American Planning Association, Chicago, IL.

BACKGROUND Socioeconomically disadvantaged groups disproportionately reported experiencing adverse circumstances resulting from the COVID-19 pandemic's socioeconomic impacts. Overarching factors associated with differentiated risks in the United States include race and ethnicity. OBJECTIVE We aim to examine: (1) the differentiated risk of experiencing adverse circumstances by race and ethnicity in the United States and (2) the trend in adverse outcomes and racial/ethnic differences in the past two years. METHODS The study utilized 49 data cycles from the Household Pulse Survey from April 2020 to September 2022. The outcomes are adverse experiences, including loss of employment income, food scarcity, housing insecurity, and unmet needs for mental health services. The racial and ethnic groups are non-Hispanic White, non-Hispanic Black, non-Hispanic Asian, non-Hispanic other minorities, and Hispanic. We compared weighted percentages of the total population and racial and ethnic groups reporting having experienced adverse circumstances during every data collection period. RESULTS We found that except for non-Hispanic Asians, racial and ethnic minorities were more likely to report loss of employment income, food scarcity, housing insecurity, and unmet needs for mental health services. Prevalence estimates by race/ethnicity for each cycle illustrated the persistent racial/ethnic disparities from April 2020 to the present. CONCLUSIONS The adverse socioeconomic impacts of the COVID-19 pandemic tended to be disproportionately higher for most racial and ethnic minorities compared to non-Hispanic Whites, and this trend continues. CONTRIBUTION This paper analyzes real time population survey data to demonstrate the extent of unequal and adverse socioeconomic impacts of the COVID-19 pandemic on minority groups and highlights the persistence of these trends in adverse socioeconomic outcomes © 2022, Demographic Research.All Rights Reserved.

ABSTRACT

It is considerably significant to recognize the psychology of undergraduates especially during the period of COVID-19 outbreak. This letter proposes an Information-Centric Networking (ICN) driven psychology recognition and health conduction method based on the active data collection mode, that is, all undergraduates submit their psychology data to manger without protest by the special sensor tool. Meanwhile, the involved undergraduates and the management center construct a large network model. In order to support the mobility of undergraduates, this letter exploits ICN paradigm to help the management and transmission of psychology data. In addition, this management center deploys Deep Belief Network (DBN) to analyze the submitted psychology data. Finally, this letter makes a practical simulation on campus, and the experimental results show that the proposed psychology control method not only has good recognition effect but also contributes to health conduction. To be specific, the proposed method can improve about 2.6% psychology recognition accuracy and decrease about 46.5% psychology recognition time. © 2021 John Wiley & Sons, Ltd.

ABSTRACT

Automatic generation of medical reports can provide diagnostic assistance to doctors and reduce their workload. To improve the quality of the generated medical reports, injecting auxiliary information through knowledge graphs or templates into the model is widely adopted in previous methods. However, they suffer from two problems: 1) The injected external information is limited in amount and difficult to adequately meet the information needs of medical report generation in content. 2) The injected external information increases the complexity of model and is hard to be reasonably integrated into the generation process of medical reports. Therefore, we propose an Information Calibrated Transformer (ICT) to address the above issues. First, we design a Precursor-information Enhancement Module (PEM), which can effectively extract numerous inter-intra report features from the datasets as the auxiliary information without external injection. And the auxiliary information can be dynamically updated with the training process. Secondly, a combination mode, which consists of PEM and our proposed Information Calibration Attention Module (ICA), is designed and embedded into ICT. In this method, the auxiliary information extracted from PEM is flexibly injected into ICT and the increment of model parameters is small. The comprehensive evaluations validate that the ICT is not only superior to previous methods in the X-Ray datasets, IU-X-Ray and MIMIC-CXR, but also successfully be extended to a CT COVID-19 dataset COV-CTR. The global lockdown measures implemented due to the COVID-19 pandemic have nearly always had negative consequences for patients with multiple sclerosis (MS). OBJECTIVE: We compared the social and professional effects of confinement on patients with MS in 2 very different populations, from Spain and China. METHODS: Questionnaires were administered to a group of patients with MS who consulted at the MS unit of Vithas hospital (DINAC Foundation) in Seville, and patients with MS attended in several provinces of China in April 2020, with the aim of analysing the differences and similarities between populations in the social and professional effects of confinement. To this end, a database was created and subsequently analysed. RESULTS: The Chinese population includes a higher proportion of younger patients and no differences were identified regarding sex. Most of the variables studied behaved in the same way in both patient populations. Spanish patients presented a lesser impact (30.7%) on their socio-economic situation than Chinese patients (44%) (P < .05). There were no significant differences between populations in the remaining variables. Social networks were widely used in the majority of patients from both populations. CONCLUSIONS: The social and professional consequences of the pandemic were very similar in both groups; the use of social networks and family support was also similar. Spanish patients seem to present greater economic stability, perhaps due to the social support they receive.
