We developed an assay that estimated the binding strength of HLA II-peptide interaction by measuring the cell-surface expression of HLA II-peptide fusion construct. In the present study, we used a newly developed HLA II-peptide interaction assay to identify HLA II-binding regions in SARS-CoV-2 Spike and the mutations that might affect the interaction with HLA II.
