ABSTRACT

The study focused on the neuroprotective role of Sorghum bicolor and vitamin C in the amelioration of oxidative stress and anxiety-like behavoiur induced by tramadol in male albino rats. The study design involved 7 groups and a control group with 5 male albino rats in each group. Tramadol (40 mg/kg) treatment was administered for 21 days. Tramadol 40mg/kg was administered in all groups. Pretreatment with varying doses of Sorghum bicolor and Vitamin C was done in three of the groups. Behavioral assessment of anxiety and locomotors actions of the groups were compared using Elevated Plus Maze (EPM) and Open Field Test (OFT). In conclusion, Sorghum bicolor and Vitamin C tramadol ameliorated oxidative stress and anxiety-like behaviour induced by tramadol. Pretreatment with Sorghum bicolor or vitamin C (100mg) can also reduced anxiogenic responses in male albino rats that are induced by chronic tramadol use.

ABSTRACT

Morin hydrate (MH) is the major flavonoid constituent of Morus alba acclaimed to have antioxidant, anti-inflammatory, anti-stress and neuroprotective properties. However, report on the effect of MH on memory performance and the underlying mechanism following chronic stress exposure is lacking. The current study aimed at investigating the neuroprotective effect of MH on chronic unpredictable stress (CUS)-induced memory impairment in mice using the Y maze test. Mice were subjected to unpredicted stress for 14 days, during which MH (5, 10 and 20 mg/kg i.p) or 25 mg/kg Ginseng was administered to them. On the 14th day, 1 h after treatment, learning and memory deficit was evaluated using the Y maze test and thereafter brains were harvested for the estimation of glutathione (GSH), lipid peroxidation product; malondialdehyde (MDA) and nitrite. Levels of inflammatory mediators tumor necrosis factor-alpha (TNF-α) and interleukin1-beta (IL-1ß), inducible nitric oxide synthase (iNOS) and nuclear factor-kappa B (NF-кB) expressions were also determined. The hippocampus was stained with hematoxylin-eosin (H&E) to examine any morphological changes in the neurons. Mice exposed to CUS showed evidence of impaired memory and increase levels of MDA, nitrite, TNF-α and IL-1ß. Furthermore, CUS reduced GSH level, increased the expressions of iNOS and NFкB immune-positive cells and produced loss of neuronal cells in the hippocampus. The MH treatment however improved memory, reduced MDA and nitrite levels, and enhanced brain GSH levels in CUS-mice. Besides, MH reduced brain levels of TNF-α and IL-1ß levels, down regulated the expressions of iNOS and NF-кB and rescue neurons in the hippocampal CA3 region of mice exposed to CUS. The results of the study indicate that MH improved CUS-induced memory impairment, which may be related to its ability to boost antioxidant defense system and suppress neuroinflammatory pathways.

BACKGROUND: Association between chlamydia trachomatis infection and male infertility is debated in literature. There is little or no information from Nigeria. The study aimed to determine the prevalence of chlamydial infection and its association with sperm quality parameters among a symptomatic men that present for infertility treatment in a Nigeria facility. METHODS: A cross-sectional study conducted at a private assisted conception clinic in Lagos, Nigeria among 138 men seeking infertility care. Seminal fluid analysis and IgG Chlamydial serology were performed for each participant. Data obtained were analysed using SPSS; p was significant at <0.05. RESULTS: Of the 138 men screened, 13.9% were Chlamydia-positive. Twenty-one per cent of clients who tested positive to Chlamydia had predominantly immotile sperm, compared with 10.2% without the infection; 26.3% with non-progressive motility had Chlamydia, compared with 2.8% men who were not infected. These differences were statistically significant (p = 0.001). More of those (57.9%) with Chlamydia (compared to 33.1% without) had significant leukocyte counts (p = 0.037). There were no statistically significant differences in sperm count and percent motility between serologically positive and negative men. CONCLUSION: Positive Chlamydia serology is associated with non-progressive motility and leukocytospermia in infertile Nigerian men
