ABSTRACT

The purpose of this research was to demonstrate the potential of adding propolis (PR) to the diet of Nile tilapia (Oreochromis niloticus) to mitigate the harmful effect of cold stress (CS) on the growth performance, redox status, and immunological response. Two trials were conducted in this study. First, 210 Nile tilapia fingerlings (28.61±0.20 g) were used in a preliminary trial to determine the appropriate PR level and supplementation period to be applied for the main trial. Fish were assigned into 7 treatment groups (3 aquaria replicates × 10 fish per aquarium in each treatment group) according to the rate of PR supplementation in the fish diets at 0, 2, 4, 6, 8, 10, and 12 g/kg for 6 consecutive weeks. The average body weight and body weight gain were determined weekly. It was found that PR supplementation at 10 g/kg in fish diet for 4 weeks was enough to obtain significant results on the growth performance of Nile tilapia. For the main trial of the present study, 480 Nile tilapia fingerlings (average weight 29.93±0.11 g) were distributed into randomized 2 PR × 2 CS factorial treatment groups (6 replicate aquariums containing 20 fish in each group). Fish of PR groups received a basal diet for a feeding period of 4 weeks, included with 10 g/kg PR (+ PR group) or without PR inclusion (- PR group). Fish of the CS groups were either challenged with cold stress at 18°C (+ CS group) or maintained at a temperature of 26°C during the feeding period (- CS group). The results showed that CS challenge significantly (p < 0.05) impaired the growth indices, redox status, and immune response in the challenged fish compared to the non-challenged fish. On contradictory, the inclusion of PR into fish diets enhanced (p < 0.05) the feed intake, growth indices, antioxidant enzyme activity, and immunological parameters. Moreover, PR treatment alleviated the CS deterioration of fish weights, specific growth rates, feed efficiency, antioxidant enzyme activity, lymphocyte proliferation, and phagocytosis activity and alleviated the elevated mortality, H/L ratio, and malondialdehyde levels by cold stress. It is concluded that the inclusion of propolis at 10 g/kg in the diet of Nile tilapia fish could be approved as a nutritional approach to enhance their performance, especially when stressed by low-temperature conditions.

ABSTRACT

The current study was proposed to explore the role of dietary propolis (PR) supplementation in alleviating the negative effects of columnaris disease (CD) challenge on the growth performance, plasma biochemicals, antioxidant activity, stress indicators, and immunological reactions of common carp (Cyprinus carpio) fish. Five hundred forty common carp juveniles were evenly placed in thirty-six 100-L tanks and stocked for acclimatization to the lab conditions with a control diet within a started period of 14 days. Fish (average initial weight of 7.11±0.06 g) were randomly distributed into one of six treatment groups (6 replicate tanks × 15 fish per tank in each treatment group). Fish in the first group was assigned as a negative control without CD challenge or PR supplementation. Fish in the other five groups were challenged with CD by immersion of fish for 60 min into a 10-L water bath supplemented with 6×106 CFU/mL (median lethal dose, LD50) of pathogenic F. columnare bacteria. After infection, the fish were restored to their tanks and fed on a basal diet supplemented with PR at 0, 3, 6, 9, or 12 g/kg diet. The experimental period continued for 6 consecutive weeks in which the feed was introduced twice a day (8:00 and 15:00 h) at a rate of 2% of the fish biomass. Ten percent of water was siphoned and renewed after each meal every day, in addition to 50% of water refreshment after cleaning the tank every three days. The tanks were continuously aerated and provided with standard rearing conditions for carp fish (24.0±1.12°C, 7.7±0.22 pH, 6.3±0.16 mg/L O2, and 14L/10D photoperiod). The growth performance traits such as feed intake (FI), weight gain (WG), final weight (FW), specific growth rate (SGR), feed efficiency (FE), and cumulative mortality rates (CM) were recorded during the experimental period. At the end of the trial, blood samples were obtained from the fish to evaluate some plasma biochemicals, including aspartate aminotransaminase (AST), alanine aminotransferase (ALT), creatinine (CRE), alkaline phosphatase (ALP), and lactate dehydrogenase (LDH), antioxidant biomarkers, including total antioxidant capacity (TAOC), total superoxide dismutase (TSOD), reduced glutathione (rGSH), and catalase (CAT), stress indicators, including heterophil to lymphocyte (H/L) ratio, cortisol (COR), malondialdehyde (MDA), and myeloperoxidase (MPO), and immunological reactions, including peripheral blood leukocyte proliferation (PBLP), phagocytosis activity (PHG), lysozyme activity (LYS), alternative complement hemolytic action (ACH50), and total immunoglobulin concentration (TIG). In addition, samples of infected fish gills were taken to quantify the number of F. columnare in the PR-supplemented groups using the quantitative real-time polymerase chain reaction (qPCR) technique. The results showed that incorporating PR into the dietary ingredients of common carp has a protective effect against the challenge with F. columnare infection. There were linear and quadratic positive trends (P < 0.05) in most parameters of growth performance, plasma biochemicals, antioxidant activity, stress indicators, and immunological reactions with the increased PR-supplemented levels in the diet of infected fish. The best results were obtained when using PR at 9 g/kg in the diet, while higher levels (12 g/kg PR) showed an adverse trend in the evaluated parameters. The FI, WG, FW, SGR, and FE were improved by approximately 37, 104, 34, 73, and 49% in the fish treated with 9 g/kg PR compared to none-PR-infected fish. In addition, adding PR at the 9 g/kg diet level was the best dose that reduced the H/L ratio, COR, MDA, and MPO by about 14, 52, 48, and 29%, respectively, in the infected fish. Furthermore, the mortality rate was reduced by 94%, and the number of pathogenic bacteria cells adherent to the fish gills was lowered by 96% in the infected fish treated with 9 g/kg PR compared to none-PR infected fish. Our results concluded that dietary supplementation with 9 g/kg PR could be a promising nutritional approach for improving the growth performance, physiological profile, and health status of common carp fish, particularly when challenged with F. columnare or similar bacterial infections.

ABSTRACT

Sperm cryopreservation is an effective technique for conserving animal genetic diversity and transmitting superior genetic backgrounds, maintained via a non-invasive sampling and collection of huge quantities of sperm. Nevertheless, cryopreservation in avian species is not commercially viable because of the rooster sperm's susceptibility to damage. This study aims to estimate the impact of dimethylacetamide (DMA) as a cryoprotectant at different levels (3%, 6%, or 9%) on the post-thawed sperm quality, motility, antioxidant-biomarkers, and the expression of anti-freeze related genes. Semen samples were collected twice a week from twelve roosters aged 40 wk, weighing 3400 ± 70 g, and belonging to the Cairo-B2 chicken strain. Fresh semen samples were rapidly appraised, pooled, diluted with two volumes of a basic extender, and divided equally into three groups. The diluted groups were chilled at -20 °C for 7 min, then gently supplemented with 3, 6, or 9% pre-cooled DMA and equilibrated at 5 °C for a further 10 min. Semen pellets were formed by pipetting drops 7 cm above liquid nitrogen (LN2), which were then kept inside cryovials in the LN2. Thawing was performed 2 months later by taking 3-4 pellets of the frozen semen into a glass tube and warming it in a water bath for 8 s at 60 °C. The results showed that 3% DMA increased the proportion of total motile sperm, progressivity, viability, and plasma membrane integrity (%) compared to the 6% and 9% DMA groups. The lipid peroxidation and antioxidant enzyme activity were improved in the 3% group. At the same time, some anti-freeze-related genes' (including ras homolog family member A (RHOA), heat shock protein 70 (HSP70), and small nuclear ribonucleoprotein polypeptide A (SNRPA1)) expressions were upregulated within the 3% DMA group relative to other groups. In conclusion, the 3% DMA group maintained higher post-thawed sperm quality than the other tested groups.

This study was proposed to highlight the impact of dietary Spirulina platensis (SP) supplementation in alleviating the deterioration effect of Escherichia coli (EC) on the growth performance, redox biomarkers, immune reaction, and hindgut microbial counts and acidosis in broiler chickens. Four hundred Cobb500, one-day-old, broiler chickens were deposited in battery cages (10 chicks per cage). The chicks were distributed into totally randomized 2 × 2 factorial treatments (10 replicate cages per treatment) from the day 22 to the day 42 of age. Birds of two of the groups were fed on a basal diet without SP supplementation (-SP groups), while birds of the other two groups were fed on a basal diet supplemented with 10 g/kg SP (+SP groups). At day 36th of age, birds in one of the -SP and +SP groups were challenged by an intraperitoneal (i.p.) injection with 107 CFU/bird EC (O157:H7 strain) in 0.5 mL sterilized saline (+EC groups), whereas the other non-challenged groups were i.p. injected with 0.5 mL saline only (-EC groups). The current study results indicated that the boilers challenged with EC had a significant (p < 0.05) lower performance, poor antioxidant activity, immunosuppression, and higher numbers of pathogenic bacteria in the intestine when compared with the non-challenged birds. Dietary SP inclusion enhanced (p < 0.05) broiler growth, antioxidant activity, immune response, and intestinal beneficial bacteria and acidosis. Moreover, SP alleviated the reduction in all these parameters after exposure to EC infection. Therefore, diets containing 10 g/kg SP could be used as a promising approach to maximize broilers' production and support their health, particularly when challenged with EC infection.

ABSTRACT

Immunosuppression is a serious consequence of oxidative stress exposure that negatively affects the productivity and profitability of birds, as well as their well-being. Thus, the present investigation was designed to evaluate the potential of corn silk extract (CSE) supplementation to overcome the negative impacts of oxidative stress induced by corticosterone administration (CORT) in broiler chickens. A total of 280 one day old Cobb 500 male chicks were divided into four groups in 2 × 2 factorial arrangements. The experimental groups included CSE supplementation (0 or 500 mg/kg diet, from 20 to 35 days of age) and CORT administration (0 or 25 mg/kg diet, from 22 to 35 days of age) as independent factors. At the end of week five of age, production performance parameters were measured. The humoral and cell-mediated immune response parameters, redox status, and stress markers were determined. Data revealed deleterious effects of CORT administration on the broilers' body weight, body weight gain, and feed conversion ratio. Moreover, an exponential increase in stress marker levels, in addition to immunosuppression and redox imbalance, were associated with CORT administration. However, CSE supplementation, with its high total phenols content, partially alleviated the negative impacts of CORT administration, as shown by a significant improvement in immune response parameters and antioxidant activity, as well as a reduction in stress marker levels. Furthermore, CSE supplementation to non-stressed birds even significantly improved total antioxidant activity, total white blood cells (TWBCs) count, T-lymphocyte stimulating index, and wattle thickness. It can be concluded that, under stress conditions in commercial broiler farms, dietary CSE supplementation can strongly be recommended to modulate the negative impacts of stress. Therefore, CSE can be used as an effective immunomodulator and antioxidant agent to increase commercial broiler farm productivity and profitability.

Sperm cryopreservation is of great importance for the poultry industry but still needs to be optimized. The high susceptibility of poultry sperm to cryodamage leads to low fertility rates after cryopreservation. Therefore, the present study aimed at evaluating the effect of including a cryoprotectant, dimethylacetamide (DMA), in the chicken semen freezing extenders at a final concentration of 3%, 6%, or 9% on the post-thawed sperm motility, quality, antioxidant biomarkers, anti-freeze gene expression, and fertilizing ability. Results showed that the total motile sperm, progressivity, and viability were quadratically increased (p < 0.05) in the 6% DMA group. The antioxidant enzyme activity and lipid peroxidation were negatively (p < 0.05) affected by the increase in DMA concentration. Furthermore, some anti-freeze-associated genes such as heat shock protein 70 (HSP70) and ras homolog family member A (RHOA) were linearly and quadratically down-regulated (p < 0.05) with the high concentration of DMA. Finally, the fertility and hatchability rates did not indicate statistical differences between DMA groups. It can be concluded that using the low concentration of 3−6% DMA in the freezing semen extender is preferable to obtain acceptable results in the post-thawed sperm quality and fertility.

ABSTRACT

This study aimed to investigate the role of dietary Spirulina platensis (SP) supplementation in relieving the negative impacts of heat stress (HS) on the productive performance, cholesterol profile, redox status, and inflammatory cytokines of laying hens. A total of 288, 45-wk-old and 1550.7 ± 2.3 g initial body weight, HY-Line W-36 laying hens were housed in two environmental-controlled compartments. Layers were allotted to eight treatments of a two × four factorial design, with six replicates containing six birds per treatment. The temperature in one of the compartments was kept at a thermoneutral condition (24 °C group), while the temperature in the other compartment was raised to a cyclic heat stress of 35 °C from 9:00 a.m. to 5.00 p.m. (35 °C group). Layers in each compartment were fed on one of four experimental diets, containing 0%, 3%, 6%, or 9% SP (SP groups). The trial continued for five weeks. As a result of this study, exposure of laying hens to cyclic HS resulted in a significant (p < 0.05) increase in the total cholesterol (CH), low-density lipoprotein-CH, liver- and egg yolk-CH, ceruloplasmin, malondialdehyde, interleukins (IL-1ß and IL-6), and tumor necrosis factor-α, and a significant (p < 0.05) decrease in the high-density lipoprotein-CH, total antioxidant capacity, and reduced glutathione levels. HS negatively (p < 0.05) affected the hen−day egg production (EP, 90.5% vs. 77.0%), egg weight (EW, 61.8 g vs. 56.8 g), feed intake (FI, 111.6 g vs. 101.5 g) and feed conversion ratio (FCR, 2.00 vs. 2.37). As SP levels increased in layer diets, a linear (p < 0.05) improvement response in most of the parameters was obtained in both HS and non-HS layers, recording the best results with 9% SP (e.g., 78.8% vs. 87.6% EP, 56.7 g vs. 61.9 g EW, 103.3 g vs. 110.2 g FI, and 2.38 vs. 2.04 FCR, in 0% vs. 9% SP, respectively). When incorporating SP into the diets of HS-layers, the negative impacts of HS were remarkably relieved (p < 0.05). Therefore, diets containing 9% SP could be used as a promising approach to improve the productive and physiological performance of laying hens, particularly under heat stress conditions.

ABSTRACT

Spirulina platensisis (SP) is a blue-green microalgae with a high value for animal and poultry nutrition. The study employed 250 40-week-old, HY-Line W-36 commercial laying hens. The layers received one of five experimental diet substitutes in five groups for 10 consecutive weeks (five replicates of 10 hens each group); a soybean-corn basal diet formulation without SP (Control group) or the soybean partially substituted with 3% SP, 6% SP, 9% SP, and 12% SP (for the remaining four groups). The results showed that dietary SP treatment significantly (p < 0.05) improved the productive performance, egg quality, blood metabolites, and hematological parameters of laying hens. In addition, there were linear and quadratic effects for increasing the levels of SP inclusion into the layer diets; however, the highest values of most parameters were observed when using 9% SP (90 g/kg of the layer diets). Furthermore, the results showed that 4.7% of the soybean meal ingredient in the layer diet could be replaced by 1% of SP. In conclusion, the partial replacement of soybean meal by SP into layer diets could be used as a promising nutritional approach to optimize the performance of laying hens.

ABSTRACT

The effects of heat stress on broiler performance and immunological response were explored using lighting-program manipulation as a potential tool. The study included 200 Cobb500 broiler chicks that were one day old at the time of recruitment. The birds were divided into four-compartment groups with similar environments (five cages per compartment, ten chicks per cage). Starting from the fourth day of age, birds of two compartments received a continuous lighting program (23L:1D a day; C.L. groups) while birds of the other two compartments received an intermittent lighting program (1L:3D 6 times per day; I.L. groups). Within each lighting program during 22-42 d of age, one group was subjected either to a thermoneutral temperature at 24 °C or heat stress at 35 °C. The results reveal that stress biomarkers, especially the plasma concentrations of corticosterone (CORT), tumor necrosis factor-alpha (TNF-α), and malondialdehyde (MDA) were relieved by 46%, 27%, and 51%, respectively, in the I.L. treatment groups compared to the C.L. program in broiler chicks subjected to heat stress. The liver function was also improved by 24% and 32% in AST and ALT levels, respectively, in the I.L. program compared to the C.L. program in stressed birds. Furthermore, the I.L. program positively influenced the immune response of the heat-stressed broilers. Eventually, the I.L. program increased the heat-stressed broilers' body weight gain and feed conversion ratio. It can be concluded that applying the I.L. program to broiler chickens can effectively improve their physiological balance and growth performance under heat-stress conditions.

(1) Background: Propolis and bee pollen have natural bioactive compounds that may support the performance and immunological response of broilers. (2) Methods: The study included 300 1 d old Cobb-500 broiler chicks. Starting from 22−42 d of age, chicks were divided according to a 2 × 2 factorial design into one of the four treatment groups (5 replicates × 15 chicks per replicate); a basal diet without supplementation (CONT) or supplemented with 1 g/kg of propolis (PR) or bee pollen (BP) separately or in an even combination (PR + BP). (3) Results: A significant (p < 0.05) increase was obtained in the body-weight gain of broilers treated with PR, BP, and PR + BP compared to the CONT. The total antioxidant capacity and superoxide dismutase were highly (p < 0.05) activated in all treated groups compared to the CONT. Immunological parameters, especially the leukocyte cell viability, T- and B-lymphocyte proliferation, immunoglobulins (IgA and IgM), antibody titers, and wattle-swelling test were significantly (p < 0.05) enhanced in the treated broilers with PR and/or BP compared to the CONT. (4) Conclusions: The dietary supplementation of PR and/or BP could be beneficial for broiler growth through maximizing the antioxidant- and immune-system defenses.

ABSTRACT

Late blight disease of potato and tomato, caused by Phytophthora infestans, results in serious losses to Egyptian and global potato and tomato production. To understand the structure and dynamics of the Egyptian population of P. infestans, 205 isolates were collected from potato and tomato plants during three growing seasons in 2010-2012. The characterization was achieved by mating-type assay, metalaxyl sensitivity assay, and virulence pattern. Additionally, genotyping of 85 Egyptian isolates and 15 reference UK isolates was performed using 12 highly informative microsatellite (SSR) markers David E. L. Cooke and five effector (RxLR) genes. Mating-type testing showed that 58% (118 of 205) of the isolates belonged to mating type A1, 35% (71 isolates) to mating type A2, and the rest 8% (16 isolates) were self-fertile. The phenotype of metalaxyl response was represented as 45% resistant, 43% sensitive, and 12% as intermediate. Structure analysis grouped the 85 identified genotypes into two main clonal lineages. The first clonal lineage comprised 21 isolates belonging to A2 mating type and 8 self-fertile isolates. This clonal lineage was identified as Blue_13 or EU_13_A2. The second main clonal lineage comprised 55 isolates and was identified as EU_23_A1. A single isolate with a novel SSR genotype that formed a distinct genetic grouping was also identified. The effector sequencing showed good correspondence with the virulence data and highlighted differences in the presence and absence of loci as well as nucleotide polymorphism that affect gene function. This study indicated a changing population of P. infestans in Egypt and discusses the findings in the context of late blight management.

ABSTRACT

Heat stress (HS) induces deleterious effects on the performance of laying hens and causes economic losses for poultry industry. This study was carried out to investigate the organic effect of selenium-enriched yeast (SY) on relieving the performance, immunity and physiological deterioration induced by heat stress in laying hens. A total of 324, 28-week-old, Hy-Line Brown commercial chicken layers were randomly distributed into 4 treatments according to a 2 × 2 factorial design, with 9 hens × 9 replicates per treatment (n = 81). From 30 to 34 weeks of age, layers were exposed to 2 temperature treatments (the HS treatment groups): a thermoneutral temperature at 24°C and a heat stress at 35°C. Layers were further assigned into the 2 subgroups according to dietary supplementation with organic selenium-enriched yeast (the SY treatment groups) at either 0 or 0.4 mg/kg diet. Results indicated that all the aspects of the layer performance during the experimental period were impaired by exposure to HS, while SY supplementation improved the layer performance in both the HS and non-HS layers. Intestinal villi disruptions and liver necrotic hepatocytes were observed in the layers exposed to HS, while villi integrity and hepatocytic normality were enhanced by SY treatment. A significant (P < 0.05) decrease in the total leukocyte count, sheep red blood cell (SRBC) antibody titer, and T- and B-lymphocyte proliferation along with an increase in the heterophils/lymphocytes (H/L) ratio were observed in the HS layers compared to non-HS layers. On the contrary, SY treatment significantly (P < 0.05) improved the immune function traits in both the HS layers and non-HS layers. Furthermore, the SY treatment plays an important role in mitigating the oxidative stress and inflammation induced by HS, displaying lower levels of plasma corticosterone, lipid peroxidation, interleukin-1ß, and tumor necrosis factor-α in HS layers supplemented with SY compared to HS layers without SY supplementation. These results conclude that addition of SY to the diet of laying hens could be applied as a potential nutritional approach to relieve the deterioration effects of heat stress on the immunity, physiological status, and productive performance of laying hens.

Oxidative stress is a detrimental physiological state that threatens birds' productivity and general health. Colocynth is an herbal plant known for its bioactive properties, and it is mainly known for its antioxidant effects. This study's purpose was to investigate how effective colocynth seed is at lowering the detrimental impact of acute oxidative stress caused by paraquat (PQ) injection in laying hens. A total of 360 Hy-Line Brown chickens, aged 39 weeks, were gathered and divided into four equal groups (10 hens × 9 replicates) in a 2 × 2 factorial design. The experimental groups were given either a basal diet or the basal diet supplemented with colocynth seed (1% of diet). Starting from week 40 of age and for 7 successive days, the experimental groups were either injected daily with paraquat (5 mg/kg body weight) or with saline (0.5 mL, 0.9% NaCl). Egg production performance with selected stress biomarkers and immunological response parameters were investigated at the end of week 40 of age. Our data revealed a significant reduction in egg production with an increase in blood stress biomarkers (i.e., HSP-70, corticosterone, and H/L ratio) in PQ-injected groups compared with non-stressed groups. Furthermore, an unbalanced redox state was detected in acute oxidative stress groups, with a significant rise in lipid peroxidation level, a reduction in total antioxidant capacity (TAC), and a drop in superoxide dismutase (SOD) and catalase enzyme activity. Supplementing PQ-injected hens with colocynth seed reduced the deleterious effects of acute oxidative stress. There was a significant drop in stress biomarkers with a significant rise in antioxidant enzyme activity and TAC observed in the PQ-injected group provided with colocynth seed supplementation. Remarkably, supplementation of colocynth in the non-stressed group resulted in a significant 27% increase in TAC concentration and 17% higher SOD activity when compared with the non-stressed control group. Colocynth supplementation in the PQ-injected group elevated the total white blood cell count by 25% and improved the B-lymphocyte proliferation index (a 1.3-fold increase) compared with the PQ-injected group that did not receive supplementation. Moreover, the non-stressed colocynth-supplemented group had significantly higher cell-mediated and humoral immune responses than the non-stressed control group. This study demonstrated that colocynth seed supplementation in birds exposed to acute oxidative stress may effectively alleviate its negative impacts on production performance, immunological responses, and redox status. We also inferred that, under normal conditions, colocynth seed can be added to laying hens' diets to stimulate production and ameliorate immune responses.

The long-term semen cryopreservation is increasingly crucial for conservation of endangered livestock and poultry species. Glycerol is the most widely used cryoprotectant for freezing chicken semen. Continuous improvement in details with glycerol may help increase the fertility of post-thawed semen. Two experiments were performed in the present study to investigate the effects of glycerol concentration, removal method, and straw type on the quality of post-thawed sperm. In experiment 1, glycerol concentration (3%, 5%, 7%, 9%, 11%, and 13%) and glycerol removal method (final dilution ratio 1:1, 1:2, 1:4, 1:8, 1:16, and 1:20) combination groups were investigated for post-thawed sperm quality, residual glycerol concentration, and fertility to find the best combinations. Experiment 2 was performed to evaluate the effects of straw type (0.25 and 0.5 mL) and glycerol concentration (3%, 5%, 7%, 9%, 11%, and 13%) on the post-thawed sperm quality. Results showed that post-thawed sperm motility of 6 glycerol concentration groups were different (P < 0.01). Sperm motility of 5%, 7%, 9%, 11% and 13% was higher than that of 3% (P < 0.01). There was no difference among different concentrations of glycerol in VSL, VCL, VAP, ALH, WOB, BCF, LIN, or STR (P > 0.05). As for the glycerol removal method, sperm motility of 1:8 dilution was the highest, followed by 1:1 and 1:2, while the difference among groups was not statistically significant (P = 0.11). Glycerol concentration and removal method had no interaction effect on sperm motion parameters (P > 0.05). The highest fertility (48.70%) was found for the 5% and 1:2 combination. There was no difference for sperm motility between 0.25 and 0.5 mL straws (P > 0.05). Glycerol concentration and straw type had no interaction effect on the sperm motion parameters (P > 0.05). It can be concluded from these observations that the combination of 5% glycerol and 1:2 dilution rendered higher fertility should be suggested in practice, and that both 0.25 and 0.50 mL straws fit the present procedure.

ABSTRACT

There is an extensive search for natural products that can be introduced to broiler rations to improve performance, especially during the unfavorable breeding conditions. Under heat-stress conditions, the immune response seriously deteriorates, which consequently impairs broiler production performance. Thus, the present study aimed to investigate the potentials of Citrullus colocynthis seeds (CCs) supplementation to modulate the immune response of broilers subjected to chronic heat stress. A total of 300 Cobb-500 male broiler chickens aged 21 days were randomly divided into two equal groups and reared under either thermo-neutral condition (24 ± 1 °C) or subjected to cyclic heat stress (34 ± 1 °C for 8 h). Each group was further divided into two groups (5 replicate × 15 chicks) and was fed either the basal diet or the basal diet with 0.1% CCs supplementation. The results showed that heat stress impaired the production performance by lowering the final body weight and feed intake as well as impairing feed conversion. The levels of stress markers (i.e., malondialdehyde, tumor necrosis factor-α and corticosterone) increased (p < 0.05), whereas the activity of antioxidant enzymes decreased in broilers exposed to heat stress. Further, heat stress caused direct suppression of broiler humoral and cell-mediated immune responses. The stimulating index of T and B lymphocytes proliferation, as well as the antibody titer against sheep red blood cells, were significantly (p < 0.05) reduced by heat-stress exposure. However, CCs supplementation to broilers subjected to heat stress improved (p < 0.05) the final body weight, feed intake, and feed conversion ratio (FCR), compared to the non-supplemented stressed group. The cellular and cell-mediated immune response indicators significantly enhanced (p < 0.05) with CCs supplementation. Supplementation of CCs to broilers reared under similar environmental conditions elevated the total white blood cells (TWBCs) count and the broiler stimulating index of T and B lymphocytes. It can be concluded that CC seeds can be effectively used to stimulate the immune response and improve the production performance of broilers reared under heat-stress condition.

ABSTRACT

The modern broiler industry faces huge challenges to keep high production quality and quantity, especially under environmental heat stress conditions. The negative effect of heat stress on broiler productivity is mediated by oxidative stress induction. The blue-green alga (Spirulina platensis) has many applications in poultry nutrition with the high levels of bioactive antioxidant compounds, which can alleviate the oxidative stress damage induced by high ambient temperature. The current study was designed to investigate the effects of dietary Spirulina inclusion at different levels on growth performance, redox status, carcass traits, meat quality, blood hematology, and metabolites profile of broilers subjected to cyclic heat stress. A total of 300 one-day-old Cobb-500 broiler chicks were recruited. Starting from day 21 to 42 of age, birds were randomly divided into five treatment groups with 6 replicates × 10 birds per group, where the first one was provided with the basal diet and reared under normal thermal conditions (23 ± 1 °C) to serve as a negative control. Meanwhile, the other four groups were exposed to cyclic heat stress (34 ± 1 °C for 8 h per day) and were fed a basal diet supplemented with Spirulina at a concentration of 0, 0.5, 1 or 1.5%. Spirulina supplementation to heat-stressed broilers was able to alleviate the negative impacts of heat stress on the final average daily gain, body weight and feed conversion ratio, with the best impact observed among the chickens fed 1% Spirulina. Hematological results indicate increasing hemoglobin and hematocrit levels with Spirulina supplementation compared to the non-supplemented stressed group. Further, Spirulina supplementation significantly influenced blood lipid metabolites marked by reduced serum cholesterol and low-density lipoprotein (LDL), and increased high-density lipoprotein (HDL) levels. The lipid peroxidation level was reduced (p < 0.05), while the antioxidant enzyme activity was increased with Spirulina supplementation to the heat-stressed group. Spirulina supplementation at 0.5 or 1% improved carcass dressing, breast and leg percentages. It can be concluded that dietary Spirulina supplementation at 0.5 or 1% to broiler reared under heat stress conditions can effectively improve broiler production performance and balance the redox status.

This study examines the effect of dietary supplementation with Lactobacillus acidophilus (LA) on the cholesterol levels, immune response, and productive performance of laying hens. A total of 216, 40-week-old, commercial Hy-Line brown chicken layers were randomly assigned into four treatment groups (18 birds × three replicates per group) and fed diet supplemented with 0 (control), 1 × 109, 21 × 109, and 31 × 109 colony forming units (CFUs) of Lactobacillus acidophilus (LA) per kg of feed for six consecutive weeks. Results show that plasma triglycerides, low-density lipoprotein (LDL) and total cholesterols became lesser, while high-density lipoprotein (HDL) cholesterol became higher in LA-supplemented groups compared to the control. In addition, a significant reduction occurred in the liver and egg yolk cholesterol by LA supplementation. Moreover, the immunological parameters including antibody titer against sheep red blood cells (SRBCs), phytohemagglutinin (PHA)-wattle swelling test, and T- & B-lymphocyte proliferation were enhanced in laying hens supplemented with LA compared to the control hens. While the heterophil to lymphocyte (H/L) ratio decreased with LA supplementation, indicating low stress conditions in the treated hens. These positive effects for LA were further reflected on the productive performance of laying hens and improved egg production, egg weight, egg mass, and feed efficiency. Our findings indicate that LA probiotic could be recommended in laying hens' diets for lowering egg yolk cholesterol with positive impacts on health and performance.

Propolis (PR) is a resin product of bee colonies that has rich bioactive antioxidant and bactericidal compounds. Endotoxin, a byproduct of bacterial growth, is reported to cause progressive induction of endogenous oxidative stress and has negative impacts on individual health and wellbeing. Hereby, we investigated the ability of PR to alleviate the oxidative stress and immunosuppression imposed by avian pathogenic Escherichia coli using laying hen as a based model. In this study, PR was dietary supplemented to hens for 4 weeks at a concentration of 0.1%. At the beginning of the 4th week of the experiment, hens from control and PR treatment were injected with E. coli (O157:H7; 107 colonies/hen) or saline. The results showed significant (p < 0.05) negative impact of E. coli challenge on antioxidant status, immune response and productive performance. PR supplementation reduced (p < 0.05) inflammation markers levels (tumor necrosis factor α (TNFα) and interleukin 1ß (IL-1ß)) and plasma corticosterone concentration. The antioxidant status was ameliorated with dietary PR supplementation to challenged hens, showing significant (p < 0.05) reduction in malondialdehyde (MDA) levels and increasing total antioxidant capacity (TAC) concentrations. Cell mediated, as well as, humeral immune response improved significantly (p < 0.05) with dietary PR verified by the enhancement of T- and B-lymphocyte proliferation and the positive respond to phytohemagglutinin (PHA). Leucocyte cells viability increased significantly and the apoptotic factor forkhead box O3 (Foxo3) was reduced with PR supplementation. The current study revealed that dietary PR supplementation can effectively be used as an organic feed additive to overcome the endogenous oxidative stress induced by endotoxins challenge.
