ABSTRACT

With growing cross-disciplinary collaboration among researchers, it is increasingly important to record detailed methodology to prevent the repetition of preliminary experiments. The purpose of this paper is to explain the development of a coccidiosis challenge model for the investigation of dietary interventions to coccidiosis in broiler chickens. The objectives are to select a dose of mixed species coccidial vaccine and evaluate the suitability (ability to produce a consistent, marked change) of selected response variables important to nutritional studies at different times postinfection (PI). Coccivac-B and Coccivac-B52 (Merck Animal Health) were evaluated as the source of coccidia in three trials. Trials 1 and 2 were randomized complete block designs with four doses (0, 10, 20, or 30 times (×) label dose) of Coccivac-B administered to 12 replicate cages of six birds by repeater pipette (Trial 1) or gavaging needle (Trial 2). Trial 3 used a completely randomized design with 0× or 30× label dose of Coccivac-B52 administered by gavaging needle to six replicate cages of six birds. Birds were gavaged at 15 days of age, and response criteria were evaluated 7 days PI in all trials and again at 10 days PI in Trials 1 and 2. All means are reported in order of increasing coccidia dose with significance accepted at P ≤ 0.05. Broiler performance was not affected by coccidia in Trials 1 or 3 but grew poorer with increasing dose from 0 to 7 days PI in Trial 2 (body weight gain, 465, 421, 388, 365 g; feed to gain, 1.37, 1.47, 1.52, 1.58). As coccidia dose increased, nitrogen corrected apparent metabolizable energy decreased (Trial 1, 3387, 3318, 3267, 3170 kcal kg-1; Trial 2, 3358, 2535, 2422, 2309 kcal kg-1; Trial 3, not measured), while relative weight, length, and content for intestinal sections increased (Trials 1through 3). Gross lesion (duodenum, jejunum/ileum, ceca) and oocyst count scores (jejunum/ileum, ceca) increased with dose; however, gross scoring often suggested infection in unchallenged birds, a finding unsupported by oocyst count scores. At 7 days PI there was no correlation between midgut gross lesion score and midgut oocyst count score (r = 0.06, P = 0.705), but cecal scores were weakly correlated (r = 0.55, P < 0.001). Administering coccidia via repeater pipette (Trial 1) resulted in respiratory distress in some birds, while use of the gavaging needle (Trials 2 and 3) successfully induced intestinal damage in chickens without resulting in coccidia related mortality. Thirty times the label dose at 7 days PI resulted in the greatest number of response variables that produced a consistent, marked change. Therefore, consideration should be given to these conditions when designing future coccidiosis challenge models using vaccines as a source of coccidia.

Con la creciente colaboración interdisciplinaria entre investigadores, es cada vez más importante registrar la metodología detallada para evitar la repetición de experimentos preliminares. El propósito de este artículo es explicar el desarrollo de un modelo de desafío de coccidiosis para la investigación de intervenciones dietéticas para coccidiosis en pollos de engorde. Los objetivos son seleccionar una dosis de vacuna coccidial de especies mixtas y evaluar la idoneidad (capacidad de producir un cambio marcado y consistente) de las variables de respuesta seleccionadas que son importantes para los estudios nutricionales en diferentes momentos posteriores a la infección (PI). Las vacunas Coccivac-B o Coccivac B-52 (Merck Animal Health) se evaluaron como fuente de coccidias en tres ensayos. Los ensayos 1 y 2 fueron diseños de bloques completamente aleatorios con cuatro dosis (0, 10, 20 o 30 veces (×) la dosis indicada en la etiqueta) de Coccivac-B administradas a 12 jaulas repetidas de seis aves mediante una pipeta repetidora (ensayo 1) o por sonda oral. (Prueba 2). El ensayo 3 utilizó un diseño completamente aleatorio con una dosis de etiqueta de 0 × o 30 × de Coccivac-B52 administrada con una sonda oral en seis jaulas repetidas de seis aves. ABSTRACT

High dietary protein and the use of poorly digested protein sources have been suggested to negatively impact broiler health, possibly because of protein fermentation in the distal intestinal tract. The effect of dietary protein levels with low or high indigestible protein fractions (LIP or HIP) on male and female broiler performance were evaluated. The trial was completely randomized with a 2 × 3 × 2 factorial arrangement, where gender, dietary protein levels (24, 26, and 28%), and dietary protein digestible fractions were the main factors. Ross 308 male (1944) and female (2232) were allocated to 72 pens with 54 males or 62 females per pen. Six grower diets 24-LIP, 24-HIP, 26-LIP, 26-HIP, 28-LIP, and 28-HIP were fed from 0 to 32 D of age. Birds were vaccinated with Coccivac-B52 on day 5, and feed intake and BW were recorded on 0, 12, 22, and 32 D. On day 32, 24 birds per treatment were processed for meat yield. Males were heavier than females at all post-hatch ages and the LIP birds were heavier than their HIP counterparts on 32 D. On day 22, birds fed 24 and 26% CP were heavier than those fed 28% CP. Birds fed the 28-LIP diet consumed less total feed than their 24 and 26-LIP equivalents. Birds fed 24% CP had the highest total feed to gain ratio, whereas LIP fed birds had a lower total feed to gain than those fed HIP diets. LIP diets resulted in higher total mortality than the HIP diets. Carcass yield was higher for females than males, increased with CP level, and was lower in HIP than LIP birds. An interaction between CP level and protein digestibility resulted in the 26 and 28-LIP having higher breast yield than all other diets. In conclusion, broiler growth performance and meat yield were affected by dietary indigestible protein alone or in combination with gender and dietary CP level.

ABSTRACT

The rate and extent of protein digestion are relevant to broiler performance and health, but information is lacking on the rate of digestion and the characteristics of the undigested fraction for common protein feed ingredients. Therefore, this study evaluated the digestion kinetics and the distal ileum (DI) digesta protein characteristics of protein meals fed to broiler chickens. Using a completely randomized design, 360 male broilers at 14 D of age were assigned to 60 battery cages and fed semi-purified diets composed of wheat starch (N-free) or wheat starch with either corn distillers dried grains with solubles (CDDGS), corn gluten meal, meat and bone meal, soybean meal, fish meal (FM), porcine meal (PCM), canola meal, blood meal (BM), or feather meal. At day 21, the protein digestion kinetics and total and soluble protein of the DI content were determined. Differences were considered significant when P ≤ 0.05. Protein source affected the extent of amino acid (AA) and CP digestibility at the DI. The results demonstrated differences in digesta mean retention time (MRT) and the rate of digestion of AA and CP among protein sources. FM had the shortest MRT of 46 min, whereas CDDGS had the longest at 142 min. Both FM and PCM had the highest digestion rates for most of the AA evaluated among the protein sources, whereas CDDGS had the lowest. In turn, the total and soluble CP in the distal ileal contents ranged from 54 to 1466 mg and 6 to 347 mg, respectively. In conclusion, dietary protein source influences the amount and solubility of the undigested protein in the DI and the digestion kinetics of AA and CP along the small intestine of broilers. These parameters may contribute to the effects of protein source on muscle deposition and could influence the impact protein sources may have on gut health through protein fermentation.

ABSTRACT

Wheat is the primary grain fed to poultry in western Canada, but its nutritional quality, including the nature of its starch digestibility, may be affected by wheat market class. The objectives of this study were to determine the rate and extent of starch digestibility of wheat market classes in broiler chickens, and to determine the relationship between starch digestibility and wheat apparent metabolizable energy (AME). In vitro starch digestion was assessed using gastric and small intestinal phases mimicking the chicken digestive tract, while in vivo evaluation used 468 male broiler chickens randomly assigned to dietary treatments from 0 to 21 d of age. The study evaluated 2 wheat cultivars from each of 6 western Canadian wheat classes: Canadian Prairie Spring (CPS), Canadian Western Amber Durum (CWAD), CW General Purpose (CWGP), CW Hard White Spring (CWHWS), CW Red Spring (CWRS), and CW Soft White Spring (CWSWS). All samples were analyzed for relevant grain characteristics. Data were analyzed as a randomized complete block design and cultivars were nested within market class. Pearson correlation was used to determine relationships between measured characteristics. Significance level was P ≤ 0.05. The starch digestibility range and wheat class rankings were: proximal jejunum - 23.7 to 50.6% (CWHWSc, CPSbc, CWSWSbc, CWRSab, CWGPa, CWADa); distal jejunum - 63.5 to 76.4% (CWHWSc, CPSbc, CWSWSbc, CWRSab, CWGPa, CWADa); proximal ileum - 88.7 to 96.9% (CWSWSc, CPSbc, CWHWSbc, CWRSb, CWGPb, CWADa); distal ileum - 94.4 to 98.5% (CWSWSb, CWHWSb, CPSb, CWRSab, CWGPab, CWADa); excreta - 98.4 to 99.3% (CPSb, CWRSb, CWHWSb, CWSWSab, CWGPab, CWADa). Wheat class affected wheat AMEn with levels ranging from 3,203 to 3,411 kcal/kg at 90% DM (CWRSc, CWSWSc, CPSb, CWGPb, CWADa, CWHWSa). Significant and moderately strong positive correlations were observed between in vitro and in vivo starch digestibility, but no correlations were found between AME and starch digestibility. In conclusion, rate and extent of starch digestibility and AME were affected by western Canadian wheat class, but starch digestibility did not predict AME.

Whole corn mash fermentations infected with industrially-isolated Brettanomyces yeasts were not affected even when viable Brettanomyces yeasts out-numbered Saccharomyces yeasts tenfold at the onset of fermentation. Therefore, aeration, a parameter that is pivotal to the physiology of Dekkera/Brettanomyces yeasts, was investigated in mixed culture fermentations. Results suggest that aeration strategy plays a significant role in Dekkera/Brettanomyces-mediated inhibition of fuel alcohol fermentations. Although growth of Saccharomyces cerevisiae was not impeded, mixed culture fermentations aerated at rates of > or =20 ml air l(-1) mash min(-1) showed decreased ethanol yields and an accumulation of acetic acid. The importance of aeration was examined further in combination with organic acid(s). Growth of Saccharomyces occurred more rapidly than growth of Brettanomyces yeasts in all conditions. The combination of 0.075% (w/v) acetic acid and contamination with Brettanomyces TK 1404W did not negatively impact the final ethanol yield under fermentative conditions. Aeration, however, did prove to be detrimental to final ethanol yields. With the inclusion of aeration in the control condition (no organic acid stress) and in each fermentation containing organic acid(s), the final ethanol yields were decreased. It was therefore concluded that aeration strategy is the key parameter in regards to the negative effects observed in fuel alcohol fermentations infected with Dekkera/Brettanomyces yeasts.

ABSTRACT

Growth rates determined by linear regression analysis revealed that Saccharomyces cerevisiae consistently grew more rapidly than Brettanomyces yeasts under a wide array of batch fermentative conditions, including acetic acid stress, in normal gravity (ca. 20 degrees Plato) mashes made from ground corn. Brettanomyces yeasts only grew more rapidly than S. cerevisiae when acetic acid concentrations were elevated to industrially irrelevant levels (>0.45%, w/v). Furthermore, the three Brettanomyces isolates used in this study failed to produce significant quantities of acetic acid under pure culture fermentative conditions. In fact, the small amounts of acetic acid which accumulated in pure culture fermentations of whole corn mash were below the concentration required to inhibit the growth and metabolism of S. cerevisiae. Acetic acid concentrations in pure culture Brettanomyces fermentations exceeded 0.05% (w/v) only in media containing low levels of glucose (<4%, w/v) or when aeration rates were elevated to at least 0.03 vol. air vol.-1 mash min-1. Consequently, it was concluded that Brettanomyces yeasts would not be capable of competing with S. cerevisiae in industrial batch fermentations of whole corn mash based solely on growth rates, nor would they be capable of producing inhibitory concentrations of acetic acid in such fermentations.

ABSTRACT

This study is an evaluation of the self-competence, academic performance, behavioral problems, and parent-child relations of boys who had been raised in single-parent families headed by their mothers and who had weekly contact with an adult friend or companion through a midwestern affiliate of the Big Brothers/Big Sisters of America. Results indicated that participation in such a program was not related to changes in the areas investigated. These findings are not consistent with the social support literature suggesting that an adult companion or friend may benefit children in single-parent families. Further study with a larger sample, over a longer time frame, is recommended.

ABSTRACT

We investigated differences in self-concept between 360 only children and 555 sibling children in the sixth grade of three urban schools and three rural schools in the People's Republic of China. Peer perceptions and teacher perceptions were also collected using sociometric measures. Self-concept was determined by the Self-Perception Profile for Children (Harter, 1985). Comparisons of sibling children and only children showed no differences on self-concept; however, urban residents had higher self-concept scores than children from rural areas, and boys scored higher than girls. No significant interaction effects between sibling status, gender, and residence were found.

ABSTRACT

The mobilities of coenzyme A and coenzyme A derivatives of cholate, chenodeoxycholate, deoxycholate, lithocholate, and their 5 alpha analogs were studied in reversed-phase high-performance liquid chromatography. With a C18 Radial-PAK A cartridge (10-micron particles) and a solvent mixture of 2-propanol/10 mM phosphate buffer (pH 7.0, 140:360), separation of the chenodeoxycholyl and deoxycholyl coenzyme A derivatives was not observed. An increase in ionic strength of the buffer to 50 mM afforded separation, which was markedly augmented with a C18 Radial-PAK A cartridge with 5-micron particles. Lowering the pH of the buffer to 5.5 did not materially change the separations regardless of the ionic strength. Quantitation was carried out to a lower level of 8.5 X 10(-12) mol.
