ABSTRACT

The available Epstein Barr virus vaccine has tirelessly harnessed the gp350 glycoprotein as its target epitope, but the result has not been preventive. Right here, we designed a global multi-epitope vaccine for EBV; with special attention to making sure all strains and preventive antigens are covered. Using a robust computational vaccine design approach, our proposed vaccine is armed with 6-16 mers linear B-cell epitopes, 4-9 mer CTL epitopes, and 8-15 mer HTL epitopes which are verified to induce interleukin 4, 10 & IFN-gamma. We employed deep computational mining coupled with expert intelligence in designing the vaccine, using human Beta defensin-3-which has been reported to induce the same TLRs as EBV-as the adjuvant. The tendency of the vaccine to cause autoimmune disorder is quenched by the assurance that the construct contains no EBNA-1 homolog. The protein vaccine construct exhibited excellent physicochemical attributes such as Aliphatic index 59.55 and GRAVY - 0.710; and a ProsaWeb Z score of - 3.04. Further computational analysis revealed the vaccine docked favorably with EBV indicted TLR 1, 2, 4 & 9 with satisfactory interaction patterns. With global coverage of 85.75% and the stable molecular dynamics result obtained for the best two interactions, we are optimistic that our nontoxic, non-allergenic multi-epitope vaccine will help to ameliorate the EBV-associated diseases-which include various malignancies, tumors, and cancers-preventively.

ABSTRACT

Since its outbreak in December 2019 in China, COVID-19 has spread like wild fire to affect many communities of the world. The high infectivity and case fatality rates of the disease among the general population and the severely ill patients respectively drew the attention of the global community. Our review showed that socio-demographic and lifestyle-related risk factors and underlying comorbid diseases were directly and indirectly associated with increased susceptibility and severity of COVID-19. These factors included older age (⩾60 years), male gender, and ethnic minority groups (especially blacks), smoking, low serum level of vitamin D, unhealthy diet, physical inactivity (with poor exposure to sunlight), overweight/obesity, high blood pressure/hypertension, high blood cholesterol, cardiovascular diseases (like stroke and coronary heart disease), diabetes mellitus, chronic obstructive pulmonary disease, chronic kidney disease, chronic liver disease, and some cancers (like leukemia, lymphoma, or myeloma). The literature further revealed that the clinical progression of the majority of these associated risk factors can be modified through effective and comprehensive risk reduction through healthy living and lifestyle modification. COVID-19 preventive and treatment guidelines that give adequate attention to risk reduction and healthy lifestyle among people-either in the pre-, peri-, or post-COVID-19 stage, should be developed by public health policymakers and clinicians. This will play a significant role in the global effort to combat the pandemic, and reduce its negative impact on the life expectancy and socio-economic development of the world particularly in low- and middle-income countries (LMICs).

ABSTRACT

Maize (Zea mays L.) is an important staple, as well as cash crop, in sub-Saharan Africa (SSA). However, its production is severely constrained by low soil nitrogen (low N). Fifty-four early-maturing hybrids developed during three breeding periods, (2008-2010, 2011-2013 and 2014-2016) were evaluated under low (30 kg ha-1) and high (120 kg ha-1) soil nitrogen (N) in Ile-Ife and Mokwa, Nigeria, from 2017 to 2019. The study was designed to (i) determine the genetic gains in grain yield of the early-maturing maize hybrids developed during the three breeding periods, (ii) determine the relationship between grain yield and other agronomic traits and (iii) identify the highest-yielding and most stable hybrids under low- and high-N environments. The 54 hybrids were evaluated using a 6 × 9 lattice design with three replications. Mean squares for hybrids were significant for measured traits under low- and high-N environments, except the mean squares for stalk lodging and EPP under low N. Annual genetic gains in grain yield were 75 kg ha-1 year-1 (2.91%) and 55 kg ha-1 year-1 (1.33%) under low- and high-N environments, respectively, indicating that substantial gains were achieved in the genetic enhancement of the early-maturing hybrids. The hybrids TZdEI 314 × TZdEI 105, TZdEI 378 × TZdEI 173, ENT 12 × TZEI 48 and TZdEI 352 × TZdEI 315 were identified as the highest-yielding and most stable across test environments and should be tested extensively on farms and commercialized in SSA.

ABSTRACT

Northern corn leaf blight (NCLB) incited by the fungus Exserohilum turcicum is a foliar disease that significantly limits maize production and productivity in West and Central Africa (WCA), particularly in the mid-altitudes but during the last decade it has become a menace in lowland agro-ecologies. The most economical and environmentally friendly disease management strategy is the cultivation of maize varieties resistant or tolerant to NCLB. However, no early maturing (EM) and extra-early maturing (EEM) NCLB resistant varieties are commercially available in WCA. One hundred inbred lines each of EM and EEM derived from tropical maize germplasm were inoculated with a virulent isolate of E. turcicum at five locations in Nigeria during the 2017 and 2018 growing seasons. The objective of the study was to identify promising NCLB resistant lines and to investigate inter-relationships among the traits. Analysis of variance revealed highly significant genotype and genotype by environment (G × E) interactions for disease severity, grain yield (GYLD), and other agronomic traits. The average disease severity (TURC) values ranged from 1.9 to 5.8 and 2.9 to 5.7 for the EM and EEM inbred lines, respectively. The levels of reaction of the inbred lines to NCLB ranged from highly resistant to highly susceptible. Stepwise regression analysis showed that ears per plant, ear and plant aspects were significantly influenced by the disease scores. Ears per plant, ear and plant aspects, TURC and GYLD traits were employed to develop a base index (BI) for selecting NCLB resistant inbred lines for hybrid development. TZEI 135 and TZEEI 1 were outstanding in GYLD and also had the highest positive BI values in the EM and EEM inbred lines, respectively. The identification of NCLB resistant lines in this study has set the premise for development of NCLB resistant hybrids for WCA as well as the improvement of tropical maize breeding populations for NCLB resistance.

ABSTRACT

Although the practice of strengthening the immune system may not guarantee that people will not contract severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), there is evidence that suggests that the likelihood and severity of many viral infections can be considerably reduced if appropriate measures are taken to increase the efficacy of the immune response to invading organisms. Evidence-based public health measures to reduce viral spread include-personal isolation, physical distancing, wearing facial masks, frequent hand washing with soap and running water, not touching the face, vaccination, etc. However once infected, recovery relies on host immunity to eradicate the foreign invaders, with symptomatic management, which is the core management strategy in COVID-19 management guidelines. Researchers have observed that severe COVID-19 prevalence and mortality is highest in people with preexisting conditions. Eighty percent of these preexisting diseases are lifestyle related. Also, the science of strengthening the immune system by adopting appropriate lifestyle changes is still evolving, especially among the suspected and confirmed COVID-19 cases. This article briefly highlights the immune response to viral infections including COVID-19 and refers to evidence that healthy lifestyle practices, enshrined as core aspects of lifestyle medicine, can strengthen the immune response to infections. This may improve outcome in COVID-19. We therefore recommend specific evidence-based lifestyle intervention measures that should be considered in the management of COVID-19.

ABSTRACT

Objective: To assess the occurrence and pattern of Toll-like receptor 4 (TLR4) co-segregated genotypes among children with Plasmodium falciparum malaria in Nigeria. Methods: In this case-control study, a total of 79 Plasmodium falciparum infected children aged 2–7 years and 105 age-matched uninfected controls of Yoruba descents in Lagos were studied. The extracted DNA samples were used for TLR4 genotyping at codons 299 (Asp> Gly) and 399 (Thr >Ile) by PCR-restriction fragment length poly-morphism. Malaria infection was diagnosed by blood smear microscopy and infected children were stratified into asymptomatic, uncomplicated and severe malaria sub-groups. Malnutrition was determined by measuring the mid upper arm circumference and anemia was defined as hemoglobin0.05) in frequency between infected and non-infected children. However, low and high occur-rences of the TLR4 Asp299Asp/Thr399Thr and Asp299Gly/Thr399Thr genotypes were observed in the severe malaria subgroup. Conclusions: This study reveals a protective role for TLR4 Asp299Gly/Thr399Ile and Asp299Asp/Thr399Thr genotypes against severe malaria in Nigerian children.
