ABSTRACT

The authors analyzed clinical experience gained in platelet concentrate transfusions to patients with blood diseases. A total of 155 patients received 974 transfusions of fresh or stored at 22 degrees C platelets. Isolation of platelets for transfusions' can be conducted by two methods: from platelet-enriched plasma, from leukothrombocytic layer. The latter is preferable because of higher functional capacity, low quantities of foreign leukocytes and red cells, decreased damage to platelets during isolation. Indications to the transfusions, their therapeutic effect, alloimmunization, resistance to platelet transfusions, prophylactic transfusions are covered.

ABSTRACT

The study of two flow methods (constant flow method and initial flow rate method) has been conducted to specify and compare methodological approaches to quantitation of red cell deformability. Washed red cells were resuspended in saline medium with glucose and albumin and examined for deformability by the above methods under different conditions. The parameters are presented to characterize flow curves obtained with the constant flow method. The authors have established optimal conditions for the measurements: the composition of the medium, the suspension HCT, leukocyte number, filtrate types, etc. With meeting of the above methodological requirements, both the methods permit quantitation of red cells deformability and can be utilized for stored blood and erythrocyte mass quality control.

ABSTRACT

The authors discuss possible ways for prevention of alloimmunization with HLA-antigen in acute leukemia patients after platelet transfusions. Prevention or delay of HLA-alloimmunization is possible when platelets of single random donor are used instead of pooled random donor platelets for transfusions. The effect of leucocyte removal from platelet concentrates and ultraviolet irradiation of platelet concentrates under conditions of blood bank on alloimmunization and refractoriness incidence has been considered.

ABSTRACT

Seven series of experiments (68 tests) were conducted on an isolated rat heart with the functioning left ventricle during its coronary vessels perfusion with suspension of human erythrocytes of different storage terms preserved in the solution containing citroglucophosphate with adenine. The main parameters of hemodynamics and oxygen regimen were evaluated. The results of the study have shown that washed human donor erythrocytes preserved in the above solution after 1, 7, 14, 21 and 28 days of storage present a highly efficient gas-carrying medium under conditions of acute anemic hypoxia.

ABSTRACT

The main parameters of hemodynamics and oxygen regimen were investigated in isolated rat heart (130 tests) during its perfusion with red blood cells preserved in glugicir (GGC) and citroglucophosphate (CGP). Short-term storage red blood cells did not show disorders in O2 transport to tissues. Long-term storage red blood cells (both in GGC and CGP) showed disorders in hemodynamics that were associated with appearance of free hemoglobin in the perfusion medium due to disruption of some red blood cells by the perfusion apparatus.

ABSTRACT

A new method has been developed for platelet concentrate (PC) preparation from the whole blood buffy coats. This method makes possible obtaining leukocyte-poor PC as compared to that prepared from platelet rich plasma. Functional characteristics of PC stored in Soviet plastic bags at room temperature (22 degrees C) under continuous agitation at circular rotator were studied during 7 days. Satisfactory maintenance of functional activity was noted during 5 days of PC storage. The method of PC preparation from the whole blood buffy coats can be also used for manual plateletpheresis of a single donor.

ABSTRACT

The isolated rat heart with the functioning left ventricle was used to measure the main parameters of hemodynamics and oxygen supply (60 experiments). Perfusion of the coronary vessels of the heart was performed with washed human red blood cells preserved for varying times (for 1, 7, 10, 14 and 21 days), stabilized on citroglucophosphate. The authors succeeded for the first time in demonstrating under artificially made conditions where the sole variable responsible for oxygen supply of the isolated organ was hemoglobin affinity for oxygen that there was no difference in the function of the isolated heart perfused with human red blood cells preserved for 7, 10 and 14 days as compared with freshly obtained ones. During perfusion of the heart with red blood cells preserved for an insignificant period of time (for 7 and 10 days), no physiological signs evidencing the impairment of oxygen supply to myocardial tissues were revealed.
