ABSTRACT

BACKGROUND: Combined chondroitin sulfate (CS) and glucosamine (GlcN) has been widely used in oral formulations to prevent and treat osteoarthritis. CS is effective for controlling pain in osteoarthritic patients, whereas GlcN can stimulate glycosaminoglycan synthesis, thus reducing extracellular matrix degradation. Although several studies have been published on this topic, the effectiveness of treatment with oral CS and GlcN remains uncertain. The objective of this study was to analyze the progression of experimentally induced osteoarthritis in horses and verify the effectiveness of an oral compound based on CS and GlcN to treat and/or modulate this disease. The study analyzed the metacarpophalangeal joint of the left thoracic limb of 16 horses divided into two groups, with eight horses treated with CS and GlcN in the treated group (GT) and eight untreated horses in the control group (GC). Chondral lesions were induced through arthroscopy, which was defined as time-point zero (T0). Physical, ultrasonographic, and radiographic examinations and synovial fluid biomarkers measurements were performed on days 0, 30, 60, 90, and 120. At the end of the experiment (T4), arthroscopy was performed again to macroscopically evaluate the joints and collect material for microscopic analysis. RESULTS: Significant differences were observed between groups in some evaluated parameters, such as visual lameness assessment, synovial concentrations of prostaglandin E2, and ultrasound examination. However, the GT still presented slightly improved results for joint flexion angle, analysis of lameness using sensors, and histopathological analysis of chondral repair tissue, however, without the statistical significance (p>0.05). CONCLUSIONS: The treatment was considered effective in the clinical modulation of experimental osteoarthritis, with improvement of some parameters in the GT. However, this type of treatment may not be entirely effective to change the catabolic process in articular cartilage and the progressive induced chondral damage.

ABSTRACT

Racehorses are exposed to repetitive overload during training and competition, causing joint hyperextension, tissue fatigue, and ultimately skeletal failure. Some degree of bone changes, such as sclerosis, are expected in equine athletes, as adaptation to the biomechanical rigors of training and racing. Understanding the imaging characteristics of the equine joint surface and subchondral bone would allow earlier detection of injuries or adaptation, improving prognosis and training programs. This study sought to describe the joint surface structural patterns and the periarticular structures of the third metacarpal bone (MC3). Both forelimbs of eight horses engaged in daily training programs, aged 3-5 years, which were euthanized for reasons unrelated to the metacarpophalangeal (MCP) joints, were collected. Specimens were evaluated through macroscopic inspection, radiography, ultrasonography, and microscopic examinations, such as optical microscopy and microtomography. Analysis of the microtomography images showed that 50% of the samples had higher trabecular thickness in the lateral condyle. Comparison of each imaging examination revealed that ultrasound images were most closely related to the histological examination (p = .29) in terms of sensitivity, while macroscopic and radiographic examinations differed most between evaluators. Finally, the irregularities and modifications observed in the articular cartilage surface and subchondral bone were normal adaptations of the anatomical structures of trained racehorses, which should be considered during clinical examination.

ABSTRACT

BACKGROUND: Mesenchymal stem cells (MSCs) represent a promising therapy for the treatment of equine joint diseases, studied due to their possible immunomodulatory characteristics and regenerative capacity. However, the source of most suitable MSCs for producing cartilage for regenerative processes in conjunction with biomaterials for an enhanced function is yet to be established. AIM: To compare the chondrogenicity of MSCs derived from synovial fluid, bone marrow, and adipose tissue of horses, using the aggrecan synthesis. METHODS: MSCs from ten horses were cultured, phenotypic characterization was done with antibodies CD90, CD44 and CD34 and were differentiated into chondrocytes. The 3D cell culture system in which biocompatible nanoparticles consisting of gold, iron oxide, and poly-L-lysine were added to the cells, and they were forced by magnets to form one microspheroid. The microspheroids were exposed to a commercial culture medium for 4 d, 7 d, 14 d, and 21 d. Proteoglycan extraction was performed, and aggrecan was quantified by enzyme-linked immunosorbent assay. Keratan sulfate and aggrecan in the microspheroids were identified and localized by immunofluorescence. RESULTS: All cultured cells showed fibroblast-like appearance, the ability to adhere to the plastic surface, and were positive for CD44 and CD90, thus confirming the characteristics and morphology of MSCs. The soluble protein concentrations were higher in the microspheroids derived from adipose tissue. The aggrecan concentration and the ratio of aggrecan to soluble proteins were higher in microspheroids derived from synovial fluid than in those derived from bone marrow, thereby showing chondrogenic superiority. Microspheroids from all sources expressed aggrecan and keratan sulfate when observed using confocal immunofluorescence microscopy. All sources of MSCs can synthesize aggrecan, however, MSCs from synovial fluid and adipose tissue have demonstrated better biocompatibility in a 3D environment, thus suggesting chondrogenic superiority. CONCLUSION: All sources of MSCs produce hyaline cartilage; however, the use of synovial liquid or adipose tissue should be recommended when it is intended for use with biomaterials or scaffolds.

ABSTRACT

Several studies in human and equine medicine have produced controversial results regarding the role of dimethylsulfoxide (DMSO) as a therapeutic agent. This study aimed to evaluate the effect of joint lavage with different DMSO concentrations on biomarkers of synovial fluid inflammation and cartilage degradation in joints with lipopolysaccharide (LPS)-induced synovitis. Twenty-six tibiotarsal joints of 13 horses were randomly distributed into four groups (lactated Ringer's solution; 5% DMSO in lactated Ringer's; 10% DMSO in lactated Ringer's; and sham). All animals were evaluated for the presence of lameness, and synovial fluid analyses were performed at 0 h, 1 h, 8 h, 24 h, and 48 h (T0, T1, T8, T24, and T48, respectively). The white blood cell counts (WBC), total protein (TP), urea, prostaglandin E2 (PGE2), interleukin (IL)-1ß, IL-6, IL-10, tumor necrosis factor-α (TNF-α), hyaluronic acid (HA), and chondroitin sulfate (CS) concentrations were measured. The WBC counts and PGE2, IL-1ß, IL-6, and TP concentrations increased in all groups at T8 compared to baseline values (p<0.05). At T48, only the 5% DMSO and 10% DMSO groups showed a significant decrease in WBC counts (p<0.05). Furthermore, the 10% DMSO group had lower concentrations of PGE2 and IL-1ß at T48 than at T8 (p<0.05) and presented lower IL-6 levels than the5% DMSO and lactated Ringer's groups at T24. All groups showed an increase in CS concentration after LPS-induced synovitis. Joint lavage with 10% DMSO in lactated Ringer´s has anti-inflammatory but not chondroprotective effects.

OBJECTIVE: The aim of this study was to verify whether transient inflammatory reactions induced by intra-articular medicinal ozone administration affect joint components, by in vivo evaluation of inflammatory (prostaglandin E2, Substance P, Interleukin-6, Interleukine-1, Tumor Necrosis Factor), anti-inflammatory (Interleukin-10) and oxidative (superoxide dismutase activity and oxidative burst) biomarkers and extracellular matrix degradation products (chondroitin sulphate and hyaluronic acid) in synovial fluid. METHODS: The effects of medicinal ozone were analyzed at two ozone concentrations (groups A and B, 20 and 40 µg/ml, respectively), using oxygen-injected joints as controls (group C); each group received ten treatments (15 ml gas per treatment). Physical evaluation, evaluation of lameness, ultrasonography, and synovial fluid analysis were performed. RESULTS: All joints presented mild and transient effusion throughout the study. Group B exhibited the highest lameness score on day 14 (P<0.05), detected by the lameness measurement system, probably because of the higher ozone concentration. All groups exhibited increased ultrasonography scores on day 14 (P < 0.05). Groups A and B exhibited increased proteins concentrations on day 21 (P<0.05). There was no change in hyaluronic acid concentration or the percentage of high-molecular weight hyaluronic acid throughout the experiment. Chondroitin sulfate concentrations decreased in group B, and did not change in group A and C, indicating that neither treatment provoked extracellular matrix catabolism. Cytokine and eicosanoid concentrations were not significantly changed. CONCLUSIONS: The ozonetherapy did not cause significant inflammation process or cartilage degradation, therefore, ozonetherapy is safe at both evaluated doses.

BACKGROUND: Bone marrow and adipose tissues are known sources of mesenchymal stem cells (MSCs) in horses; however, synovial tissues might be a promising alternative. The aim of this study was to evaluate phenotypic characteristics and differentiation potential of equine MSCs from synovial fluid (SF) and synovial membrane (SM) of healthy joints (SF-H and SM-H), joints with osteoarthritis (SF-OA and SM-OA) and joints with osteochondritis dissecans (SF-OCD and SM-OCD) to determine the most suitable synovial source for an allogeneic therapy cell bank. METHODS: Expression of the markers CD90, CD105, CD44, and CD34 in SF-H, SM-H, SF-OA, SM-OA, SF-OCD and SM-OCD was verified by flow cytometry, and expression of cytokeratin, vimentin, PGP 9.5, PCNA, lysozyme, nanog, and Oct4 was verified by immunocytochemistry. MSCs were cultured and evaluated for their chondrogenic, osteogenic and adipogenic differentiation potential. Final quantification of extracellular matrix and mineralized matrix was determined using AxioVision software. A tumorigenicity test was conducted in Balb-C(nu/nu) mice to verify the safety of the MSCs from these sources. RESULTS: Cultured cells from SF and SM exhibited fibroblastoid morphology and the ability to adhere to plastic. The time elapsed between primary culture and the third passage was approximately 73 days for SF-H, 89 days for SF-OCD, 60 days for SF-OA, 68 days for SM-H, 57 days for SM-OCD and 54 days for SM-OA. The doubling time for SF-OCD was higher than that for other cells at the first passage (P < 0.05). MSCs from synovial tissues showed positive expression of the markers CD90, CD44, lysozyme, PGP 9.5, PCNA and vimentin and were able to differentiate into chondrogenic (21 days) and osteogenic (21 days) lineages, and, although poorly, into adipogenic lineages (14 days). The areas staining positive for extracellular matrix in the SF-H and SM-H groups were larger than those in the SF-OA and SM-OA groups (P < 0.05). The positive mineralized matrix area in the SF-H group was larger than those in all the other groups (P < 0.05). The studied cells exhibited no tumorigenic effects. CONCLUSIONS: SF and SM are viable sources of equine MSCs. All sources studied provide suitable MSCs for an allogeneic therapy cell bank; nevertheless, MSCs from healthy joints may be preferable for cell banking purposes because they exhibit better chondrogenic differentiation capacity.

The objective of the current article was to compare the quality of the healing process after hysterorrhaphy with catgut and chitosan suture wires via macroscopic, histochemical, and immunohistochemical evaluations. Seven ewes were submitted to a video-assisted laparotomy. A hysterectomy was performed with subsequent catgut and chitosan hysterorrhaphy in both horns. Then, macroscopic evaluation and biopsies of the hysterorrhaphy and control areas were carried out after 30 days. Although postoperative adhesion was observed in five animals of the catgut group, no adhesions were detected in the chitosan one. Additionally, there was a significant difference in increasing collagen type III and connective tissue for the chitosan group when compared to the control and catgut groups. A larger amount of blood vessels was also observed in the control group in comparison with the catgut and chitosan groups. Collagen type I and vascular endothelial growth factor-A (VEGF-A) were significantly different among groups. Thus, chitosan promoted a preventive effect on postoperative adhesion formation in the hysterorrhaphy areas, with increasing collagen type III deposition during tissue mending, which granted an enhanced healing process.

ABSTRACT

O presente estudo descreve a morfologia da trabécula septomarginal no coração de javalis, de modo a oferecer dados sistemáticos que permitam um conhecimento mais detalhado do coração desta espécie. Os fragmentos das trabéculas foram submetidos à técnica histológica convencional, cortados em espessura de 5µm e corados pela Hematoxilina e Eosina e Tricrômico de Masson modificado. Com auxílio de um microscópio de luz AxioscópioZeiss as imagens foram captadas e analisadas através do programa específico de morfometria KS-400 Zeiss. Os dados obtidos foram submetidos ao teste ''U'' de Mann-Whitney (p0,05). A proporção média de tecido muscular estriado cardíaco foi 77,24%±2,31, a de tecido conjuntivo 4,76%±0,59. As miofibras de condução cardíaca apresentaram proporção média de 16,28%±1,71e a vascularização de 6,97%±0,79.


The goal of this study was to describe the morphology of septomarginaltrabecula in the heart of wild boars to provide systematic data that enable and assist the more detailed knowledge of the heart of this species. The fragments of trabecula were subjected to conventional histological technique, cut to a thickness of 5µm and stained with Hematoxylin and Eosin and modified Masson's trichrome. With the use of digital optical microscope Zeiss Axioscópio took up photomicrographs and the images were analyzed employing image analysis program Zeiss KS-400. The data were subjected to application of Mann-Whitney U test. The mean and standard error of the proportion of connective tissue was 4.76%±0.59, the striated muscle tissue was 77.24%±2.31, the cardiac conduction myofibers16.28%±1.71and the vascularization, 6.97%±0.79.

ABSTRACT

O timo é um órgão linfático primário que desenvolve sua atividade em organismos jovens. Apesar de sua função ser responsável por mecanismos fundamentais na aquisição das defesas e conseqüentes respostas orgânicas, ela ainda não está totalmente esclarecida, nem tampouco as bases morfológicas que respondem por tais funções, como o processo de desenvolvimento e involução do órgão. Objetivou-se analisar e caracterizar os aspectos morfológicos do timo, tais como seu tamanho e volume, e aspectos histológicos do timo em gatos, correlacionando o sexo e o desenvolvimento etário. Doze timos provenientes de fetos de gatos domésticos (Felis domesticus) sem raça definida (SRD), machos e fêmeas, separados em três grupos etários. O timo apresentou-se com uma coloração rosa-pálida e com duas porções, a torácica e a cervical, sendo que cada uma delas possuía um lobo direito e um lobo esquerdo em sua maioria. A porção torácica localizava-se em região de mediastino cranial, entre os pulmões e à base do coração. E a porção cervical estendia-se além das costelas em sentido cranial, estando localizada ventralmente à traqueia. A estrutura celular do timo demonstrou-se organizada com a presença de agregados concêntricos, os chamados corpúsculos tímicos, formados por células epiteliais, sustentada por uma cápsula de tecido conjuntivo de onde partiam septos que ao penetrar no órgão dividia-o em lóbulos. Ocorreram variações significativas quanto à lobação e as dimensões do timo entre indivíduos da mesma faixa etária, e entre sexos diferentes. Os valores relativos ao comprimento, espessura e largura, de maneira geral, apresentaram aumento, em conformidade ao desenvolvimento dos animais, mas com diferenças entre os sexos.


The thymus is a primary lymphatic organ that develops its activity in young organisms. But despite its function is fundamental mechanisms responsible for the acquisition and subsequent body defenses, it is not yet fully understood, neither the morphological basis accounting for such functions as the process of development and the organ involution. The objective was to analyze and characterize the morphology of the thymus, such as its size and volume, and histological aspects of the thymus in cats, correlating sex and age development. Twelve foetus of mongrel domestic cats (Felis domesticus), males and females, divided into three age groups. The thymus presented two parts with a pale pink color, the thoracic and cervical portion; each has a right lobe and a left lobe mostly. The thoracic portion was located in the region of cranial mediastinum, between the lungs and the heart base. And the cervical portion extended beyond the ribs cranially and is located ventral to the trachea. The thymus cellular structure was shown by the presence of organized concentric aggregates, named Hassall's corpuscles, formed by epithelial cells, supported by a connective tissue capsule that penetrated the parenchyma dividing it into lobules. Significant changes with number of lobes and thymus size between individuals of the same age, and between sex. The values for length, thickness and width, in general, showed an increase, according to the development of animals, with differences between sex.

ABSTRACT

The increase of heart collagen fibers in diabetics is a well known fact, but the consequences are not defined. The aim was to quantify the cardiac collagen fibers in normal and diabetic rats treated with vitamin C. We selected 32 Wistar rats, 16 diabetic animals induced endovenously with streptozootocin, and 16 healthy animals, half of them, diabetics and normals, were treated with vitamin C for 90 days. After the experimental proceeding, the hearts were removed and processed accordingly to conventional protocol for optical microscopy and specific staining for collagen. The results showed that the diabetic rats presented increase in the number of cardiac collagen fibers, but the ones treated with vitamin C showed little accumulation of fibers. It could be concluded that treatment with vitamin C is important for the prevention of heart failure in diabetic animals.


O aumento do conteúdo de fibras colágenas no coração de diabéticos é um fato bastante conhecido, suas conseqüências ainda são objeto de estudo e causam certa controvérsia, portanto este trabalho objetivou estudar a variação na quantidade das fibras de colágeno cardíacas em animais normais e diabéticos tratados pela vitamina C. Para isso foram selecionados 32 ratos Wistar, 16 diabéticos induzidos pela injeção endovenosa de estreptozootocina e 16 normais, sendo metade deles tratados com Vitamina C (diabéticos e normais) por um período de 90 dias. Após período experimental, os corações foram retirados e processados segundo protocolo convencional para microscopia óptica e coloração específica para colágeno. Os resultados mostram que animais diabéticos apresentam maior quantidade de fibras de colágeno cardíacas e que o tratamento com a vitamina C determinou um menor acúmulo na quantidade dessas fibras.

ABSTRACT

O objetivo deste trabalho foi de analisar morfometricamente fragmentos histológicos do baço de animais normais e diabéticos, comparando os resultados encontrados e relacionando-os ao sexo e a suplementação da vitamina C. Foram utilizados 32 ratos Wistar, os quais foram analisados número de vasos, o número de folículos germinativos (polpa branca) e o diâmetro dos vasos de cada animal. As análises histológicas e morfometricas foram feitas em amostras de 5µm de espessura demonstrando que: na quantidade de folículos germinativos, observamos resultados comparando, independente do sexo, animais controles suplementados com vitamina C e controles não suplementados (p£"0,05; F=0,1452); na quantidade de vasos, observamos resultados comparando, fêmeas diabéticas suplementadas pela vitamina C e fêmeas diabéticas não suplementadas (pd"0,05; F=6.8893); e no diâmetro dos vasos, observamos resultados comparando fêmeas, tanto no grupo controle quanto ao grupo diabético, suplementadas pela vitamina C quando comparadas às fêmeas não suplementadas pela vitamina C (p<0,05; U=121.50; Z(U)=2.1234) e (p<0,05; F=4.8134). De um modo geral, a indução de diabetess modifica o diâmetro vascular nas fêmeas e que a administração de vitamina C interfere nos dados métricos relativos ao diâmetro vascular somente nas fêmeas.


The objective of this study was to analyze morphometric histological shatters of the spleen of normal and diabetic animals, comparing the joined results and relating them in the sex and the suplementation of vitamin C. Had been used 32 Wistar rats, which had been analyzed: number of vases; the number of germinative folicules (white pulp); and the diameter of the vases of each animal. Histological and morphometric analyses were applied on 5µm thick samples and showed that: in the amount of germinative folicules, we observe resulted comparing, independent of the sex, animals controls supplemented with vitamin C and controls not supplemented (p£0.05; F=0.1452); in the amount of vases, we observe resulted comparing, of diabetic females supplemented by vitamin C when compared with not supplemented (pd"0.05; F=6.8893); and in the diameter of the vases, we observe resulted comparing females, as much in the group has controlled how much to the diabetic group, supplemented with vitamin C when compared with the females not supplemented with vitamin C (p<0.05; U=121.50; Z(U)=2.1234) and (p<0,05; F=4.8134). In a general way, the induction of diabetes modifies the vascular diameter in the females and that the vitamin C administration only intervenes with relative metric data to the vascular diameter in the females.

ABSTRACT

O nó sinoatrial, por se encontrar topograficamente instalado como componente inicial do sistema de condução, é responsável pela geração dos impulsos nervosos determinantes da contração cardíaca. Estudos relacionados à morfologia do nó, visando conhecer a origem, trajeto e distribuição dos vasos neste tecido são conhecidos, contudo, no que diz respeito a estes aspectos e aos dados quantitativos da irrigação nodal, no que se refere ao comportamento vascular arterial e a densidade vascular arterial desta região, a literatura é escassa. Com este objetivo foram utilizados 30 corações de suínos SRD, sendo 27 injetados com resina vinílica corada, para análise da origem e trajeto da ANSA (artéria do nó sinoatrial) e 3 corações injetados com solução aquosa de carvão coloidal (tinta nanquim) para proceder à análise estereológica. As artérias atriais originaram-se tanto da artéria coronária direita quanto da esquerda, com predominância da primeira (66,66 por cento e 33,33 por cento, respectivamente). Quando originada da coronária direita, a irrigação ocorreu pelo ramo AADAM (artéria atrial direita cranial medial) em 14 casos e pelos ramos AADAI (artéria atrial direita cranial medial) em 2 casos e AADAL (artéria atrial direita cranial lateral) em 2 casos. Em 9 casos (33,33 por cento) originou-se pela artéria coronária esquerda: quatro pelo ramo AASPL (artéria atrial esquerda caudal lateral), dois pelo ramo AASAI (artéria atrial esquerda cranial intermédia) e três pelo ramo AASAM (artéria atrial esquerda cranial medial). Anastomoses interarteriais, com participação dos vasos responsáveis pela irrigação do território do nó sinoatrial foram observadas na maioria dos casos (25 corações). The sinoatrial node, for being topographically installed as the initial component of the conduction system, is responsible for the production of the nervous impulses, which determines the cardiac contraction. There have been made studies related to the node's morphology in order to know the origin, distribution and quantification of the vases in this tissue, however, in spite of the results and quantitative data of the nodal irrigation - arterial vascular conduct and arterial vascular density, at the nodal level - the literature is scarce. With this objective 27 SRD swine hearts, injected with colored resin for macroscopic analysis of the origin and distribution of ANSA (sinoatrial node artery), 3 others injected with watery solution of colloidal coal (dyed nanquim) were used, to mark the route of the vases on the node level for stereological analysis. The atrial arteries originated as well from the right coronary artery as from the left one, with predominance of the first (66.66 percent and 33.33 percent, respectively). When originated from the right coronary artery, there existed the following branches: AADAM (right cranial medial atrial artery) in 14 cases, AADAI (right cranial intermedial atrial artery) in 2 cases, and AADAL (right cranial lateral atrial artery) in 2 cases. In 9 cases (33.33 percent) the following branches originated from the left coronary artery: 4 through branch AASPL (left caudal lateral atrial artery), 2 through branch AASAI (left cranial intermedial atrial artery) and 3 through branch AASAM (left cranial medial atrial artery). ABSTRACT

The relation between hyperglycemia and diabetic neuropathy has already been demonstrated in some studies. Among the theories proposed for its etiology the oxidative stress stands out. The performance of nitric oxide as a link between the metabolic and vascular neuropathogenic factors that triggers the diabetic neuropathy has already been put forward. This study aimed to assess the quantification and measurements of the cell body profile area (CBPA) of NADPH-diaphorase reactive (NADPH-dp) myenteric neurons of the jejunum of diabetic rats (induced by streptozotocin) supplemented with Ascorbic Acid (AA). These changes in the myenteric neurons seem to be related to the gastrointestinal disturbances observed in diabetes mellitus (DM). Twenty male Wistar rats (Rattus norvegicus) were distributed in 4 groups (n=5): controls (C), control supplemented (CS), diabetic (D), and diabetic suplemented (DS). DM was induced by estreptozotocin (50mg/kg body wt). One week after the induction and confirmation of the DM (glycemia exam), animals of the groups CS and DS received 50mg of AA three times a week by gavage. After 90 days of experiment, the animals were anesthetized with lethal thiopental dose (40mg/kg) and the collected jejunum processed for the histochemistry NADPH-diaphorase technique. Whole-mount preparations were obtained for quantitative and morphometric analysis of the myenteric neurons. A quantity of jejunum neurons in the Group D (96±7.5) was not different (P>0.05) from Group DS (116±8.08), C (92±9.7), and CS (81±5.4), but in Group DS the quantity was higher (P<0.05) than in Group C and CS. The CBPA of neurons from Group D (189.50±2.68µm²) and DS (195.92±3.75µm²) were lower (P<0.05) than from Group C (225.13±4.37µm²) and CS (210.23±3.15µm²). A relação entre hiperglicemia e neuropatia diabética foi demonstrada em várias pesquisas. Entre as teorias propostas para sua etiologia destaca-se o estresse oxidativo. O papel do óxido nítrico como elo entre os fatores neuropatogênicos metabólico e vascular que ativam a neuropatia diabética tem sido ressaltado. Este estudo objetivou avaliar a quantificação e a morfometria da área do perfil do corpo celular (CBPA) de neurônios mioentéricos NADPH-diaforase reativos (NADPH-dp) do jejuno de ratos diabéticos e suplementados com Ácido Ascórbico (AA), uma vez que alterações nos neurônios mioentéricos parecem estar relacionadas aos distúrbios gastrointestinais observados no diabetes mellitus (DM). Vinte ratos machos da linhagem Wistar (Rattus norvergicus) foram distribuídos em 4 grupos (n=5): controle (C), controle suplementado (CS), diabético (D) e diabético suplementado (DS). O DM foi induzido através de injeção de estreptozootocina (50mg/kg de peso corporal). Uma semana depois da indução e confirmação do DM (glicemia), animais dos grupos CS e DS receberam, via gavagem, 50mg de AA três vezes por semana. Após 90 dias de período experimental, os animais foram anestesiados com dose letal de thiopental intravenosa (40mg/kg) e o jejuno foi retirado e processado para a técnica histoquímica da NADPH-diaforase. Preparados de membrana foram obtidos para análises quantitativa e morfométrica dos neurônios mioentéricos. A quantidade de neurônios do jejuno do Grupo D (96±7,5) não diferiu (P>0,05) dos Grupos DS (116±8,08), C (92±9,7) e CS (81±5,4), mas no Grupo DS o número de neurônios foi superior (P<0,05) aos Grupos C e CS. A CBPA de neurônios do Grupo D (189,50±2,68µm²) e DS (195,92±3,75µm²) foi menor (P<0,05) do que a dos Grupos C (225,13±4,37µm²) e CS (210,23±3,15µm²). ABSTRACT

O timo é um órgão de grande importância imunológica durante a vida fetal e o período neonatal. Ele é o precursor da linfopoiese e apresenta alta atividade linfopoiética, sendo também o maior órgão linfóide durante a vida intrauterina e até o nascimento. Neste estudo, os aspectoso morfológicos do desenvolvimento prenatal de timos de cães foram descritos em fetos de diferentes idades e sexos. Vinte e quatro fetos de cães domésticos, sem raça definida, machos e fêmeas, foram divididos em 4 grupos etários. O timo apresentou-se composto por dois lobos unidos por um tecido de conexão, com coloração rósea, localizados no espaço mediastinal cranial. A porção cranial estendeu-se pouco além do primeiro par de costelas. As células que formavam o parênquima do timo estavam bem organizadas. Agrega dos concêntricos chamados Corpúsculos de Hassal foram observados, envolvidos por uma delgada cápsula de tecido conjuntivo que é espessada por tecido adiposo. O tamanho dos vasos aumentou gradativamente com a idade dos fetos, sendo maior nos animais próximos ao nascimento. Os timos das fêmeas apresentaram maior volume (Vref) e dimensões de tamanho que nos machos.


The thymus is an organ with great immunological relevance during foetus life and neonatal period, It is the predecessor of lymphopoiesis and shows high lymphopoietic activity, being also the largest lymphoid organ during intrauterine life and at birth. In this study, the morphological aspects of the prenatal development of dog thymus were described in foetus of different ages and sexes. Twenty-four foetus of mongrel domestic dogs, males and females, were divided into four age groups. The thymus presented two lobes linked by a rose-colored tissue connection that was localized in the cranial mediastinal space. The cranial portion protrudes slightly beyond the first pair of ribs. The cells that formed the thymus parenchyma were well organized. Concentric aggregates named Hassal corpuscles were observed, involved by a thin capsule of connective tissue thickened by adipose tissue. The vessels size gradually increased with the foetus age, being larger in animals that were closer to birth. The volume (Vref) and size parameters of female thymus were larger than in males.
