ABSTRACT

Uterine fibrosis is an acquired disorder leading to menstrual irregularities, implantation impairment, and abortion. Mesenchymal stromal cells (MSCs) have antifibrotic properties through chemokine secretion. MSC-conditioned media (MSC-CM) contain paracrine components-exosomes-with a great potential for repairing damaged tissue or preventing fibrosis. The main goal of this study was to evaluate the preventive effects of bone marrow-derived MSC-CM (BM-MSC-CM) on uterine fibrosis after uterine curettage in rabbits. This study included 12 female rabbits (24 uterine horns in total). Excised uteri of each of the 12 female rabbits were randomly divided into four groups of intact negative control, curettage positive control, BM-MSC injection, and BM-MSC-CM injection in the way that two corresponding uteri from a rabbit were allocated to different groups. The MSC-CM were collected from cultivated BM-MSCs 48 hours after having been washed three times and replaced in serum-free media. Through a surgical approach, the caudal parts of the uteri were submitted to traumatic endometrial curettage, except for the intact negative uteri. After suturing the uterine walls, BM-MSCs or BM-MSC-CM were injected in the curettage site. Endometrial regeneration was histologically evaluated 30 days after treatment. Based on the evaluation of histomorphometric indices, curettage with or without preventive injections increased the growth of endometrial layers. However, the amount of fibrotic tissue in the CM and the BM-MSC injection groups was the same as the normal control groups, and all were less than the curettage group. A single injection of CM of MSCs after 30 days prevented the fibrotic tissue formation induced by curettage in endometrial layers of rabbits. Injecting BM-MSC-CM immediately after curettage prevented and reduced the uterine fibrosis similar to BM-MSCs in a rabbit model.

ABSTRACT

INTRODUCTION: The purpose of this study was to compare canal transportation and the centering ability of Reciproc (VDW, Munich, Germany), WaveOne (Dentsply Maillefer, Ballaigues, Switzerland), and EdgeFile (EdgeEndo, Albuquerque, NM) rotary systems using cone-beam computed tomographic imaging. METHODS: Ninety mesiobuccal mandibular first molar uncalcified canals with at least a 19-mm length, a canal curvature of 15°-30° (the Schneider method), and a mature apex were selected. Canals were randomly divided into 3 groups of 30 teeth, and canal preparation with the Reciproc, WaveOne, and EdgeFile systems was performed according to the manufacturers' instructions. Cone-beam computed tomographic images were taken before and after instrumentation in the same position. Apical transportation was calculated in the distances of 2, 3, and 4 mm from the apex. Kruskal-Wallis and Mann-Whitney U tests were used to statistically analyze the data. RESULTS: The mean canal transportation was significantly lower with EdgeFile (P < .001) followed by the WaveOne rotary system. Moreover, the centering ability of the EdgeFile system was higher than that of the WaveOne and Reciproc systems. CONCLUSIONS: The EdgeFile rotary system showed the lowest transportation in both the mesiodistal and buccolingual directions and the highest centering ability. The Reciproc system showed the highest transportation and the lowest centering ability.

ABSTRACT

A yeast culture grown in a nutrient-rich medium initially containing 2% glucose is not limited in calorie supply. When yeast cells cultured in this medium consume glucose, they undergo cell cycle arrest at a checkpoint in late G1 and differentiate into quiescent and non-quiescent cell populations. Studies of such differentiation have provided insights into mechanisms of yeast chronological aging under conditions of excessive calorie intake. Caloric restriction is an aging-delaying dietary intervention. Here, we assessed how caloric restriction influences the differentiation of chronologically aging yeast cultures into quiescent and non-quiescent cells, and how it affects their properties. We found that caloric restriction extends yeast chronological lifespan via a mechanism linking cellular aging to cell cycle regulation, maintenance of quiescence, entry into a non-quiescent state and survival in this state. Our findings suggest that caloric restriction delays yeast chronological aging by causing specific changes in the following: 1) a checkpoint in G1 for cell cycle arrest and entry into a quiescent state; 2) a growth phase in which high-density quiescent cells are committed to become low-density quiescent cells; 3) the differentiation of low-density quiescent cells into low-density non-quiescent cells; and 4) the conversion of high-density quiescent cells into high-density non-quiescent cells.

ABSTRACT

Composed of trillions of individual microbes, the human gut microbiota has adapted to the uniquely diverse environments found in the human intestine. Quickly responding to the variances in the ingested food, the microbiota interacts with the host via reciprocal biochemical signaling to coordinate the exchange of nutrients and proper immune function. Host and microbiota function as a unit which guards its balance against invasion by potential pathogens and which undergoes natural selection. Disturbance of the microbiota composition, or dysbiosis, is often associated with human disease, indicating that, while there seems to be no unique optimal composition of the gut microbiota, a balanced community is crucial for human health. Emerging knowledge of the ecology of the microbiota-host synergy will have an impact on how we implement antibiotic treatment in therapeutics and prophylaxis and how we will consider alternative strategies of global remodeling of the microbiota such as fecal transplants. Here we examine the microbiota-human host relationship from the perspective of the microbial community dynamics.

ABSTRACT

INTRODUCTION: The aim of this study was to assess the radiographic technical quality of root canal therapy performed by fifth year students of Dental School of Hamadan University of Medical Sciences from 2015 to 2016. METHODS AND MATERIALS: Four hundred and seventy records of root canal therapies were evaluated. Records with graphies taken as initial, master apical file (MAF), master apical cone (MAC) and final radiographs were included in the study and records of patient younger than 16 years and older than 68 years were excluded from further investigations. Lastly, 432 teeth were selected. Obturation length, canal tapering, quality and density of filling material were the variables investigated in the present study. Two independent investigators examined the radiographies using a magnifying lens (×2) and x-ray viewer. Data were analyzed using chi-square test. RESULTS: The technical quality of root filling performed by undergraduate dental students was classified as acceptable in 10.4% of cases. Moreover, 70.8% of teeth had adequate filling, 17.1% were underfilled and 12% were overfilled. The three groups were significantly different in terms of working length and taper quality. One hundred ninety four (44.9%) records had adequate taper and 109 (25%) records had adequate density. There was a significant association between teeth location and the length of obturation so that the probability of a successful treatment was higher in maxillary teeth. Furthermore, the rate of a proper length of obturation was higher among incisors than that of premolars and molars. CONCLUSION: The technical quality of root canal therapy performed by dental students in Hamadan University of medical sciences is not as acceptable as it should be. One of the most important factors in this regard is a high student/professor ratio.
