ABSTRACT

A new nutrient medium has been designed to culture and isolate the plague microbe ChDS-37 on the basis of the pancreatic digest of baker's yeast. The results of laboratory tests of the designed medium, by using 10 plague microbe strains and those of approval during the tactical and special training of a specialized antiepidemic team (SAET), suggest that the medium has some advantage over reference media and creates prerequisites for being incorporated into the mobilization reserve of a SAET.

ABSTRACT

Fragmentation of the pYT plasmid of the plague pathogen by ten restriction endonucleases has been studied. The evidence has been obtained in support of the possible presence of the movable genetic element containing a HindIII site within the plasmid pYT. The gene encoding the I fraction of the plague pathogen has been cloned. The physical map of the pYT plasmid has been constructed with the use of restriction endonucleases BamHI, XhoI, BstEII, SmaI, EcoRI, and HindIII. The fragment of the plasmid DNA coding for the synthesis of the plague pathogen fraction I has been mapped.

ABSTRACT

Various methods for phage multiplication have been tested: the method of cellophane membranes, the method of combined media, the method of forced aeration of a liquid medium, growth at suboptimal temperature and growth at optimal temperature under stationary conditions. The phage in a high concentration (n X 10(10) plaque-forming units per ml) can be obtained by any of these methods with the exception of the last one yielding lower (by 1 or 2 orders) effectiveness of multiplication. The main factors which determine the titer of the phage in lysate are concentration of live cells in the suspension and temperature of growth.

ABSTRACT

Electron microscopy methods were used to determine the main parameters of the structural organization of cholera "C" phage. The particles of the phage were found to consist of a capsid of icosahedral shape and a thin noncontractile process. The capsid with the triangulation number 7 consists of 72 morphological subunits and contains DNA with the molecular weight of (68.16 +/- 1.63) x 10(6) daltons. The process of the phage consists of 216 morphological subunits and ends with a peculiar adsorption apparatus including a distal filament and three fibrils with knobs at the ends.
