ABSTRACT

Bartonella spp. are intracellular hemotropic bacteria primarily transmitted by arthropod vectors to various mammalian hosts, including humans. In this study, we conducted a survey on wild populations of sigmodontine rodents, Akodon azarae and Oxymycterus rufus, inhabiting the Paraná River delta region. The study involved eight grids organized in a crossed 2 × 2 design, where four of the grids were exposed to cattle while the other four were not, and four grids were located in implanted forest while the remaining four were in natural grasslands. Our objective was to examine whether the occurrence of Bartonella spp. in rodents was associated with silvopastoral activities (cattle raising associated with timber production) conducted in the region. Additionally, we evaluated the associations between Bartonella infection and other environmental and host factors. We present compelling evidence of a significant positive association between Bartonella prevalence and the presence of implanted forests and cattle. Furthermore, we identified the presence of a Bartonella genotype related to the pathogen Bartonella rochalimaea, infecting both A. azarae and Ox. rufus. These findings suggest that anthropogenic land-use changes, particularly the development of silvopastoral practices in the region, may disrupt the dynamics of Bartonella.

ABSTRACT

The haematophagous mite Ornithonyssus sylviarum may cause important economic losses in commercial poultry farms whilst also potentially affecting the health of farm workers. The dynamics of this ectoparasite has been linked to several factors, including wild birds, fomites, farm workers, management of hen houses, and host traits. Along two consecutive years, we carried out systematic sampling at three laying hen farms located in Santa Fe province, Argentina, with the aim of identifying factors that may influence O. sylviarum prevalence and intensity. We found that the density of feathers around the hen vent area and the presence of Menoponidae lice were negatively associated with mite abundance. We also found that the density of hens in the cages was negatively associated with mite prevalence, suggesting a possible dilution effect, whereas prior reports found a positive association with hen density. In addition, summer was the season with minimum mite prevalences and intensities, contrary to previous studies in northern farms where warm weather appeared to prompt an increase in mite populations. Another factor associated with mite intensity was age, but this effect varied depending on the season, which hints that the association between hen's age and mites is complex. Basic epidemiological knowledge on O. sylviarum in poultry farms from South America may aid in a more efficient and integrative approach to its control.

ABSTRACT

The majority of known Philornis species (Diptera: Muscidae) cause subcutaneous myiasis in Neotropical altricial and semi-altricial nestlings. Generating information about these parasites is of relevance as they are a threat for the conservation of some wild bird species and also because they are an excellent model to study the ecology of host-parasite interactions. Although there are records of this parasitism in adult birds, the parasitic larvae are primarily associated with nestlings. However, systematic surveys including active search for larvae in older birds have been very scarce. Here we present results from a survey conducted along 6 years and encompassing a sample of 3029 subadult and adult birds in Santa Fe province, Argentina, where broods are frequently parasitised by the Philornis torquans complex. We found only one dipteran larva parasitising a greater thornbird. This larva was morphologically compatible with Philornis. Given the extremely low estimated prevalence (1/3029 = 0.03%), we conclude that the presence of larvae in subadult and adult birds is an extremely rare event in the studied bird community and that the species belonging to the P. torquans complex present in central Argentina are almost exclusively parasites of nestlings.

ABSTRACT

Bats are among the most diverse, widespread, and abundant mammals. In Argentina, 67 species of bats have been recorded, belonging to 5 families and 29 genera. These high levels of biodiversity are likely to complicate identification at fieldwork, especially between closely related species, where external morphology-based approaches are the only immediate means for a priori species assignment. The use of molecular markers can enhance species identification, and acquires particular relevance in capture-release studies. In this study, we discuss the extent of the use of the mitochondrial cytochrome b gene for species identification, comparing external morphology identification with a molecular phylogenetic classification based on this marker, under the light of current bat systematics. We analyzed 33 samples collected in an eco-epidemiological survey in the province of Santa Fe (Argentina). We further sequenced 27 museum vouchers to test the accuracy of cytochrome b -based phylogenies in taxonomic identification of bats occurring in the Pampean/Chacoan regions of Argentina. The cytochrome b gene was successfully amplified in all Molossid and Vespertilionid species except for Eptesicus, for which we designed a new reverse primer. The resulting Bayesian phylogeny was congruent with current systematics. Cytochrome b proved useful for species-level delimitation in non-conflicting genera (Eumops, Dasypterus, Molossops) and has infrageneric resolution in more complex lineages (Eptesicus, Myotis, Molossus). We discuss four sources of incongruence that may act separately or in combination: 1) molecular processes, 2) biology, 3) limitations in identification, and 4) errors in the current taxonomy. The present study confirms the general applicability of cytochrome b -based phylogenies in eco-epidemiological studies, but its resolution and reliability depend mainly, but not solely, on the level of genetic differentiation within each bat genus.

Wild birds may be considered a possible source of parasitic mesostigmatid mites for poultry, but only few studies explored this hypothesis. In addition, there is very little information about the parasitic mites present in commercial poultry systems from southern South America. To contribute with data on parasitic mesostigmatid mites at the domestic-wildlife interface, we conducted a study in which samples were systematically collected from laying hens and wild birds (adults and nestlings), for two years at three commercial laying hen farms. The occurrence of mesostigmatid mites were compared among hosts. A proportion of the collected mites were morphologically identified to the species level, finding that host preference varied greatly depending on mite species: laying hens were only parasitized by Ornithonyssus sylviarum, wild bird nestlings were mostly parasitized by Ornithonyssus bursa, and in small proportion, by O. sylviarum, while adult passerines were parasitized by both Ornithonyssus species, and sporadically by Pellonyssus cf. reedi and Dermanyssus cf. triscutatus. In laying hens, there was intra- and inter-annual variability in mite occurrence, but no consistent seasonal pattern, whereas in adult wild birds, mites showed the highest prevalence in spring and the lowest in summer. Not coinciding with this general pattern, the occurrence of O. bursa matched the reproductive activity of wild birds. A phylogenetic analysis based on a fragment of the 16S rRNA gene was carried out for a subsample of the mites collected, showing that the O. sylviarum mites present on adult wild birds and laying hens had the same haplotype (100% identity). Additionally, mites obtained from wild birds morphologically identified as O. bursa presented two distinctive haplotypes (89.8% identity), one phylogenetically related to O. sylviarum and the other to O. monteiroi. These findings show that in central Argentina commercial laying hens are parasitized mainly by O. sylviarum while wild birds are also hosts to other mite species. Adult wild passerines, especially house sparrows, may be a source of O. sylviarum for commercial poultry.

ABSTRACT

The aim of this study was to assess the presence of Rickettsia in soft ticks (Acari: Argasidae) collected from insectivorous bats (Chiroptera) in Santa Fe province, Argentina. First, a subset of ticks were mounted in Hoyer's medium to be determined by morphological characters and then confirmed by sequencing the mitochondrial 16S rRNA gene. Also ticks were processed by PCR assays using primers CS-78 and CS-323, which amplify a fragment of the Rickettsia spp. gltA gene. Positive ticks were subjected to a second PCR round with primers Rr190.70p and Rr190.701n of the spotted fever group rickettsiae ompA gene. A phylogenetic analysis was performed with Maximum-likelihood method, and the best fitting substitution models were determined with the Akaike Information Criterion. Five bats of the species Eptesicus diminutus Osgood, 1915, Eptesicus furinalis (d'Orbigny and Gervais, 1847), Eptesicus spp. (Vespertilionidae), and Molossops temminckii Burmeister, 1854 (Molossidae) were parasitized with Ornithodoros hasei (Schulze, 1935) larvae. One E. diminutus ticks' tested positive to "Candidatus Rickettsia wissemanii", a spotted fever group rickettsiae. The association O. hasei -"Ca. R. wissemanii" detected in this study represents the first evidence of a Rickettsia in Ornithodoros ticks in Argentina and the third report of this association in America. Also, this finding constitutes the first record of "Ca. R. wissemanii" in Argentina. Finally, we found for the first time the insectivorous bats E. diminutus and E. furinalis parasitized with O. hasei larvae. These findings add two new hosts and a new location, the southernmost recorded to date, for O. hasei.

ABSTRACT

Calodium hepaticum (Nematoda; Capillariidae) is a parasitic nematode of mammals with a cosmopolitan distribution. Adults of this nematode can infect the liver of many mammalian species, including humans, but the main hosts are members of the superfamily Muroidea. Among these, Rattus spp. have the highest apparent prevalences reported worldwide. There are only two reports of C. hepaticum infecting Sigmodontinae species (Muroidea: Cricetidae). In this survey, we examined the occurrence of C. hepaticum in two assemblages of Sigmodontinae rodents from Argentina (Santa Fe and Entre Ríos provinces). The diagnosis was made by morphological features, histopathological exam, and molecular characterization of 18S ribosomal RNA gene region. Here, we show that C. hepaticum is a common parasite of Sigmodontinae. We report the infection in six species (all new hosts): Akodon azarae, Calomys callidus, Calomys venustus, Oligoryzomys flavescens, Oligoryzomys nigripes, and Oxymycterus rufus. This is the first report of C. hepaticum in Sigmodontinae rodents from Argentina and the second record in this subfamily for South America. It is also the first confirmation of C. hepaticum infection in Sigmodontinae by molecular diagnosis. The genetic findings and the prevalences observed, together with the existing information on C. hepaticum, lead us to propose that Rattus spp. brought C. hepaticum to the New World exposing Sigmodontinae rodents, which are frequently infected by this parasite. The high prevalence of infection in A. azarae (41.2%) suggests that this host may be playing an important role in C. hepaticum dynamics in the New World.

ABSTRACT

Hosts counteract infections using two distinct defence strategies, resistance (reduction in pathogen fitness) and tolerance (limitation of infection damage). These strategies have been minimally investigated in multi-host systems, where they may vary across host species, entailing consequences both for hosts (virulence) and parasites (transmission). Comprehending the interplay among resistance, tolerance, virulence and parasite success is highly relevant for our understanding of the ecology and evolution of infectious and parasitic diseases. Our work investigated the interaction between an insect parasite and its most common bird host species, focusing on two relevant questions: (i) are defence strategies different between main and alternative hosts and, (ii) what are the consequences (virulence and parasite success) of different defence strategies? We conducted a matched field experiment and longitudinal studies at the host and the parasite levels under natural conditions, using a system comprising Philornis torquans flies and three bird hosts - the main host and two of the most frequently used alternative hosts. We found that main and alternative hosts have contrasting defence strategies, which gave rise in turn to contrasting virulence and parasite success. In the main bird host, minor loss of fitness, no detectable immune response, and high parasite success suggest a strategy of high tolerance and negligible resistance. Alternative hosts, on the contrary, resisted by mounting inflammatory responses, although with very different efficiency, which resulted in highly dissimilar parasite success and virulence. These results show clearly distinct defence strategies between main and alternative hosts in a natural multi-host system. They also highlight the importance of defence strategies in determining virulence and infection dynamics, and hint that defence efficiency is a crucial intervening element in these processes.

Both species of ticks are vectors of Rickettsia parkeri, a spotted fever group rickettsia. Ticks were recovered from tourists in August 2014 and December 2015 at the Federico Wildermuth Foundation (31° 59'S, 61° 24'O), San Martin Department, Santa Fe province. Five adult ticks were morphologically identified as A. tigrinum (3 females and 1 male) and A. triste (1 female). This is the first finding including both Amblyomma maculatum group species, A. triste and A. tigrinum, together in the same locality in Argentina. This finding suggests that this site might have favorable features for the development of both species of R. parkeri vector. Further studies including sampling of a larger number of ticks and detection of R. parkeri DNA are needed to better document the epidemiology of this rickettsia in Santa Fe.

El objetivo del presente trabajo es comunicar el hallazgo de Amblyomma triste por primera vez en la provincia de Santa Fe, Argentina, y sumar una nueva localidad para Amblyomma tigrinum en la misma provincia. Ambas especies de garrapatas son reconocidos vectores de Rickettsia parkeri, agente etiológico de la fiebre manchada. Los especímenes fueron colectados durante el mes de agosto de 2014 y diciembre de 2015 a partir de turistas en la Fundación Federico Wildermuth (31° 59'S, 61° 24'O), departamento de San Martín, provincia de Santa Fe. Se colectaron 5 garrapatas adultas que fueron identificadas morfológicamente como A. tigrinum (3 hembras y 1 macho) y A. triste (1 hembra). El presente hallazgo de A. triste y A. tigrinum representa el primero en incluir a ambas especies del complejo Amblyomma maculatum en una misma localidad y permite alertar sobre la presencia de vectores de R. parkeri en la zona. Nuevos estudios que incluyan el muestreo de un mayor número de garrapatas y la detección de R. parkeri permitirán dilucidar más aspectos sobre la situación de esta rickettsia en Santa Fe.

ABSTRACT

The present study explores associations of different factors (i.e. host parameters, presence of other ectoparasites and [mainly biotic] environmental factors) with burdens of Ixodes loricatus immature stages in one of its main hosts in Argentina, the rodent Akodon azarae. For 2 years, rodents were trapped and sampled monthly at 16 points located in four different sites in the Parana River Delta region. Data were analysed with generalized linear mixed models with a negative binomial response (counts of larvae or nymphs). The independent variables assessed were (a) environmental: trapping year, presence of cattle, type of vegetation, rodent abundance; (b) host parameters: body length, sex, body condition, blood cell counts, natural antibody titers and (c) co-infestation with other ectoparasites. Two-way interaction terms deemed a priori as relevant were also included in the analysis. Most of the associations investigated were found significant, but in general, the direction and magnitude of the associations were context-dependent. An exception was the presence of cattle, which was consistently negatively associated with both larvae and nymphs independently of all other variables considered and had the strongest effect on tick burdens. Mites, fleas and Amblyomma triste were also significantly associated (mostly positively) with larval and nymph burdens, and in many cases, they influenced associations with environmental or host factors. Our findings strongly support that raising cattle may have a substantial impact on the dynamics of I. loricatus and that interactions within the ectoparasite community may be an important-but generally ignored-driver of tick dynamics.

Here we offer a multivariable analysis that explores associations of different factors (i.e., environmental, host parameters, presence of other ectoparasites) with the interaction of Amblyomma triste immature stages and one of its main hosts in Argentina, the rodent Akodon azarae. Monthly and for two years, we captured and sampled rodents at 16 points located at 4 different sites in the Parana River Delta region. The analyses were conducted with Generalized Linear Mixed Models with a negative binomial response (counts of larvae or nymphs). The independent variables assessed were: (a) environmental: trapping year, season, presence of cattle; type of vegetation (natural grassland or implanted forest); rodent abundance; (b) host parameters: body length; sex; body condition; blood cell counts; natural antibody titres; and (c) co-infestation with other ectoparasites: other stage of A. triste; Ixodes loricatus; lice; mites; and fleas. Two-way interaction terms deemed a priori as relevant were also included in the analysis. Larvae were affected by all environmental variables assessed and by the presence of other ectoparasites (lice, fleas and other tick species). Host factors significantly associated with larval count were sex and levels of natural antibodies. Nymphs were associated with season, presence of cattle, body condition, body length and with burdens of I. loricatus. In most cases, the direction and magnitude of the associations were context-dependent (many interaction terms were significant). The findings of greater significance and implications of our study are two. Firstly, as burdens of A. triste larvae and nymphs were greater where cattle were present, and larval tick burdens were higher in implanted forests, silvopastoral practices developing in the region may affect the population dynamics of A. triste, and consequently the eco-epidemiology of Rickettsia parkeri. Secondly, strong associations and numerous interactions with other ectoparasites suggest that co-infestations may be more important for tick dynamics than has so far been appreciated.

In the present study, we report the first in vitro isolation and infection intensity of Rickettsia parkeri in Amblyomma triste ticks from Argentina. No genetic differences in the molecular targets evaluated were found between R. parkeri isolates from Argentina and those R. parkeri isolates reported in Uruguay and Brazil, both obtained from A. triste. Only a minor difference was observed when compared to R. parkeri isolated from Amblyomma maculatum from United States. Moreover, the prevalence of infection by R. parkeri in ticks collected from the vegetation in the Paraná Delta was high (20.4%). Interestingly, the distribution of R. parkeri infection intensity observed in A. triste ticks was distinctly bimodal, with approximately 60% of the infected ticks presenting high rickettsial loads (3.8×10(5)-4.5×10(7) ompA copies/tick) and the remainder with low rickettsial levels (5.6×10(1)-6.5×10(3) ompA copies/tick). This bimodality in R. parkeri infection intensity in ticks could determine differences in the severity of the disease, but also be important for the infection dynamics of this pathogen. Further research exploring the distribution of rickettsial infection levels in ticks, as well as its determinants and implications, is warranted.

ABSTRACT

Information on autochthonous ticks and their hosts is scarce in South America, especially in Argentina. To contribute to tick knowledge in the region, 2094 ticks were collected from the vegetation, humans, domestic and wild animals at a host-and-tick rich area of northern Argentina during six field trips conducted in 1999 (January and August), 2000 (March and November), and 2001 (March and June). The ticks were identified as Amblyomma cajennense (Fabricius), Amblyomma coelebs Neumann, Amblyomma sp., Haemaphysalis juxtakochi Cooley, H. leporispalustris (Packard), Ixodes pararicinus Keirans & Clifford, I. loricatus Neumann, I. longiscutatum Boero and Ixodes sp. Small mammals were mainly parasitized by immature stages of Ixodes; humans and domestic animals, predominantly by Amblyomma spp., and birds, mainly by nymphs and larvae of Haemaphysalis spp.

Com o objetivo de contribuir para o conhecimento dos carrapatos na região, 2094 carrapatos foram coletados da vegetação, de humanos e de animais domésticos e selvagens numa área no norte da Argentina rica em carrapatos e hospedeiros, durante seis viagens de campo conduzidas em 1999 (janeiro e agosto), 2000 (março e novembro) e 2001 (março e junho). Os carrapatos foram identificados como Amblyomma cajennense (Fabricius), A. coelebs Neumann, Amblyomma sp., Haemaphysalis juxtakochi Cooley, H. leporispalustris (Packard), Ixodes pararicinus Keirans & Clifford, I. loricatus Neumann, I. longiscutatum Boero e Ixodes sp. Pequenos mamíferos foram principalmente parasitados por estágios imaturos de Ixodes; humanos e animais domésticos, predominantemente por Amblyomma spp., e pássaros, principalmente por ninfas e larvas de Haemaphysalis spp.
