ABSTRACT

The citrus industry loses a significant amount of mandarin fruits either before or shortly after harvesting due to rind disorder. Different citrus cultivars are impacted by a physiological rind disorder that lowers fruit quality and marketability. Although the primary etiology of this condition is unknown, changes in relative humidity (RH) and rind water status can make it worse. The damage is initiated in the fall, especially following rain. It begins with irregular water-soaked areas that develop into dark-brown, necrotic lesions covering large portions of the fruit's surface. The damage is evident in some citrus types such as Satsuma Owari mandarins and other cultivars. In this study, we attempted to understand and control the occurrence of this kind of rind disorder in Satsuma Owari mandarins growing under California conditions. Our data showed that fruit located in the outer part of the canopy suffer more than fruit in the interior canopy. We were able to reduce this damage in Satsuma Owari mandarins by applying 2,4-dichlorophenoxyacetic acid (2,4-D) at 16 milligrams/Liter (mg/L), gibberellic acid (GA3) at 20 mg/L, or Vapor Gard® at 0.5 percent (v/v) at the color break stage. However, GA3 caused a delay in color development by approximately four weeks. GA3-treated fruit changed their color completely four weeks after the control, and the rind damage was at a very low percentage. Delaying rind senescence could be a good strategy to reduce the damage in mandarin orchards. Data showed that in addition to the benefits of the different treatments on preventing rind disorder at harvest, they have some beneficial effects during storage for four weeks either at 0.5 or 7.5 °C.

ABSTRACT

Citrus fruit's appearance is the primary criterion used to assess its quality for the fresh market, hence the rind's condition is a crucial quality trait. Pre-harvest rind disorder is one of the major physiological problems in mandarins. The disorder occurs right before harvest following rain events in some Mandarin varieties. Despite the economic damage caused by this kind of disorder, very limited information is available about the molecular mechanisms underlying the occurrence of this disorder. In the present study, we evaluated the primary metabolites, antioxidants, and hormones associated with the pre-harvest rind disorder in Mandarins. The study was carried out using ten-year-old 'Owari' Satsuma mandarin trees grafted on 'Carrizo' rootstock and grown in a commercial orchard in San Joaquin Valley, California, USA. Samples were collected from healthy tissue of healthy fruit (HF_HT), healthy tissue of damaged fruit (DF_HT), and damaged tissue of damaged fruit (DF_DT). Damaged fruit (DF_HT and DF_DT) showed lower cellulose concentrations than healthy fruit tissues (HF_HT), however, had similar contents of pectin and hemicellulose. The antioxidant activities showed no significant difference in all paired comparisons between samples as expressed in the malondialdehyde (MDA) content. However, DF_DT had a higher H2O2 content compared to HF_HT, but DF_HT had a similar content to that of HF_HT. Furthermore, peroxidase (POD) and polyphenol oxidase (PPO) activities were increased in DF_DT compared to HF_HT (P = 0.0294) and DF_HT (P = 0.0044), respectively. Targeted metabolomics analysis revealed that a total of 76 metabolites were identified in Satsuma rind tissues, and the relative concentrations of 43 metabolites were significantly different across studied samples. The hormonal analysis showed the involvement of jasmonate O-methyltransferase, jasmonic acid-amido synthetase JAR1-like, and JA-isoleucine may key role in causing the rind disorder in mandarins. In addition, the damaged fruit tissues have a higher level of jasmonic acid (JA), 12-oxo-phytodienoic acid, and JA-isoleucine than undamaged tissue.

ABSTRACT

Botryosphaeria branch canker and dieback of avocado (Persea americana Mill.) has expanded in avocado-growing areas in recent years. Twenty-one avocado groves in the major producing regions of California were surveyed in 2018 and 2019. Monthly inoculations of wounded, green, and lignified branches of 'Hass' and 'Lamb Hass' were conducted. Botryosphaeriaceae were the predominant fungi recovered from cankered tissues collected across the surveyed traditional and high-density orchards and caused symptoms on all six sampled cultivars. These fungi were also recovered in asymptomatic twigs and other organs and thus exist as a potential reservoir for future infections. Molecular analyses of 173 isolates showed that Neofusicoccum luteum had the greatest incidences across sites and cultivars, with 83 and 29% recovered from Hass and Lamb Hass, respectively. Pathogenicity tests on excised (Hass, GEM, and Hass mutants) and attached shoots from potted (Hass) and mature avocado trees (Hass and Lamb Hass) showed that all species were pathogenic on wounded, green, and mature branches of the specified cultivars. Monthly inoculations of wounded, green, and lignified branches of Hass and Lamb Hass showed that both stem types were susceptible throughout the inoculation periods, regardless of the avocado phenological stage. In temperature-dependent growth and infection studies, growth of three points could vary during the growing season. Botryosphaeriaceae grown was higher between 20 and 30°C, but only Lasiodiplodia theobromae significantly grew and caused external lesions at 35°C. Lasiodiplodia theobromae also grew more on perseitol-amended media, all indicating its adaptation to warmer temperatures and capacity in metabolizing the avocado-produced sugar. Overall, this study extended our knowledge of the prevalence, identity, and pathogenicity of Botryosphaeriaceae on avocado cultivars, which will be useful to tailor management strategies.

ABSTRACT

The domestication history of the avocado (Persea americana) remains unclear. We created a reference genome from the Gwen varietal, which is closely related to the economically dominant Hass varietal. Our genome assembly had an N50 of 3.37 megabases, a BUSCO score of 91%, and was scaffolded with a genetic map, producing 12 pseudo-chromosomes with 49,450 genes. We used the Gwen genome as a reference to investigate population genomics, based on a sample of 34 resequenced accessions that represented the 3 botanical groups of P. americana. Our analyses were consistent with 3 separate domestication events; we estimated that the Mexican group diverged from the Lowland (formerly known as "West Indian") and Guatemalan groups >1 million years ago. We also identified putative targets of selective sweeps in domestication events; within the Guatemalan group, putative candidate genes were enriched for fruit development and ripening. We also investigated divergence between heterodichogamous flowering types, providing preliminary evidence for potential candidate genes involved in pollination and floral development.

ABSTRACT

In this work, descriptive analysis (DA) and consumer panels were conducted on "Hass" and "3-29-5" (GEM® ) avocados, grown in southern California. Both panels encompassed at least five time points across the 2019 harvest season. The DA panel identified and evaluated overall richness, creamy, smooth, watery, oily, sweet, bitter, umami, salty, astringent, buttery, nutty, and green. The texture attributes received the highest scores in both "Hass" and "3-29-5." Both varieties increased in richness, creaminess, and oiliness at harvests 5 and 6. The consumer panel found that "3-29-5" showed more changes in its eating experience over the season than "Hass," which agreed with dry weight data collected in a simultaneous analytical study. Correspondence analysis indicated that "Hass" samples had a consistent sensory profile over the harvest season, whereas "3-29-5" changed substantially, becoming more closely associated with a positive eating experience late in the harvest season. This is the first work to characterize avocado flavor over the harvest season using both trained and consumer sensory panels. PRACTICAL APPLICATION: Many aspects of avocado were found to have some impact on flavor, but textural properties were by far the most important in determining how well the fruit was liked. This information will be useful in future taste evaluations of avocado and the ongoing development of new avocado varieties.

ABSTRACT

The research objective was to characterize avocado's aroma-active volatiles and use information about its overall composition, such as lipid profile, to discuss likely biosynthetic origins. To achieve this, two varieties, "Hass" and "3-29-5" (GEM), were evaluated during their commercial harvest period for dry weight, moisture content (freeze-drying), oil content (Soxhlet extraction), fatty acid composition, and aroma profile. Solvent-assisted flavor evaporation and aroma extract dilution analysis were performed on aroma extracts. Oleic acid (>50%) was the prominent fatty acid in the oil of both varieties. The majority of the aroma-active compounds in avocado are lipid-derived. The most notable compounds are 1-octen-3-one (mushroom) with a flavor dilution factor as high as 8192, hexanal (grassy), (Z)-4-decenal, an unknown, and (E,E)-2,4-nonadienal. Over the mid-to-late harvest season, a decline in hexanal and an increase in octanal were observed. In contrast to "Hass", the hexanal content was relatively stable in "3-29-5".

ABSTRACT

Phytophthora root rot (PRR), caused by Phytophthora cinnamomi, is the most destructive disease of avocado worldwide. In the United States, mefenoxam and phosphonate products are currently the only registered fungicides for managing avocado PRR. Four new Oomycota-specific and two registered fungicides, all with different modes of action, were evaluated. Seventy-one isolates of P. cinnamomi from avocado in California, most of them collected between 2009 to 2017, were tested for their in vitro sensitivity to the six fungicides. Baseline sensitivity ranges and mean values (in parentheses) of effective concentrations to inhibit mycelial growth by 50% (EC50) for the new fungicides ethaboxam, fluopicolide, mandipropamid, and oxathiapiprolin were 0.017 to 0.069 µg/ml (0.035), 0.046 to 0.330 µg/ml (0.133), 0.003 to 0.011 µg/ml (0.005), and 0.0002 to 0.0007 µg/ml (0.0004), respectively. In comparison, the EC50 value range (mean) was 0.023 to 0.138 µg/ml (0.061) for mefenoxam and 12.9 to 361.2 µg/ml (81.5) for potassium phosphite. Greenhouse soil inoculation trials with 8-month-old Zutano seedlings and 10-month-old Dusa and PS.54 clonal rootstocks were conducted to assess the efficacy of these fungicides for managing PRR. Mefenoxam and potassium phosphite were effective treatments; however, oxathiapiprolin, fluopicolide, and mandipropamid were more effective. Ethaboxam was effective in reducing PRR on the rootstocks evaluated. Oxathiapiprolin reduced PRR incidence and pathogen population size in the soil by >90%, and plant shoot growth and root dry weight were significantly increased compared with the control; thus, oxathiapiprolin was one of the best treatments overall. The high activity and performance of these new fungicides supports their registrations on avocado for use in rotation and mixture programs, including with previously registered compounds, to reduce the risk of development and spread of resistance in pathogen populations.

ABSTRACT

Phytophthora cinnamomi, the causal agent of Phytophthora root rot (PRR), is the most destructive disease of avocado worldwide. A previous study identified two genetically distinct clades of A2 mating type avocado isolates in California; however, the phenotypic variation among them was not assessed. This study described the phenotype of a subset of isolates from these groups regarding growth rate, growth temperature, virulence, and fungicide sensitivity. Isolates corresponding to the A2 clade I group exhibited higher mycelial growth rate and sensitivity to higher temperatures than other isolates. Among the fungicides tested, potassium phosphite had the highest 50% effective concentration for mycelial growth inhibition and oxathiapiprolin had the lowest. Mycelial growth rate and potassium phosphite sensitivity phenotypes correlate with specific groups of isolates, suggesting that these traits could be a group characteristic. Moreover, isolates that are more virulent in avocado and less sensitive to potassium phosphite were identified. A detached-leaf P. cinnamomi inoculation method using Nicotiana benthamiana was developed and validated, providing an alternative method for assessing the virulence of a large number of isolates. This information will help avocado PRR management and assist breeding programs for the selection of rootstocks resistant against a more diverse pathogen population.

Pomegranate (Punica granatum L.) is an important fruit in many cultures. The fruit and juice have risen in popularity as it was discovered that pomegranate has relatively high antioxidant activity compared to most other fruits. In this study, six cultivars were utilized to determine consumer acceptance compared to the industry standard, 'Wonderful,' which comprises 90% to 95% of commercial production in the United States. Fruit were sourced from 2 cultivar field trials, one in inland Riverside, California, and one in coastal Ventura County, California. Cultivars selected for the study included 'Eversweet,' 'Green Globe,' 'Haku Botan,' 'Loffani,' 'Phoenicia,' 'Wonderful,' and 'cv. 857,' an heirloom cultivar from Ventura County, CA, U.S.A. Pomegranate arils were subject to sensory evaluation by 87 untrained consumer panelists in late 2016. Panelists were given pomegranate arils and asked to score the samples using a 9-point Hedonic scale for the following fruit quality traits: aril color, sweetness, tartness, seed hardness, bitterness, and overall desirability. There were significant differences among cultivars for all traits assessed by the sensory panelists. There were differences in acceptance among consumers for 'Wonderful' depending on if it was grown on the coast versus inland, and consumers preferred inland- versus coastal-grown 'Wonderful.' 'Wonderful' pomegranate was associated with cultivars that consumers scored low on desirability for bitterness. Cultivars that scored well in overall desirability compared with 'Wonderful' were 'cv. 857,' 'Eversweet,' 'Green Globe,' and 'Phoenicia.' PRACTICAL APPLICATION: Consumer sensory panels are important to determine scientifically which cultivars are desired by the public. These panels allowed for the determination of which pomegranate cultivars are liked or disliked by consumers and why. If the pomegranate growers know the most desirable cultivars for consumers, they are more likely to adopt and plant them, thus potentially increasing the diversity in the marketplace, as has been with apples, peaches, plums, pears, mangoes, strawberries, raspberries, blueberries, and citrus.
