ABSTRACT

Demulsification and crude oil desorption are usually a necessary step for the treatment of oily sludge in the petroleum industry. In this study a binary mixed bio-surfactant (rhamnolipid / sophorolipid, RL/SL) was used to strengthen the removing oil efficiency for oily sludge by thermal washing method. Surface tension values of the single and the mixed surfactants were carried out to investigate the effect of mixing systems on reducing critical micelle concentrations (CMC) value. The models proposed by Clint, Rubingh and Gibbs et al. had been employed to interpret the formation of mixed micelles and synergism and found out in case of the mass ratios of 4:6 the synergism was the strongest in RL and SL mixed surfactant systems, which was selected as the washing agents to treat the oily sludge produced from Huabei oilfield. Through the optimization of oil washing process parameters, the oil removal rate reached the maximum value (95.66%, residual oil rate 1.98%) at the condition of heating temperature of 45 °C, detergents concentration of 500 mg/L, washing time of 3 h, liquid/solid mass ratio of 1:4, stirring speed of 300 r/min, and washing 4 times. The factors affecting the oil washing effect were analyzed from the composition and performance characteristics of oily sludge samples, washing oil system and washing process parameters. The results showed that low oil content of oily sludge, small specific surface area, strong wetting and solubilization of the oil-washing system all can increase the oil-washing effect and the washing time and temperature had a great influence on the oil-washing effect. Compared with the results of other researchers, the oil washing temperature and the concentration of oil washing agent were significantly lower and high oil removal rate and low residual oil rate were obtained in this study. It was confirmed that thermal oil washing method using RT/SL binary bio-surfactant mixing system was proved to a high-efficiency, low-consumption and wide range of applications technology.

ABSTRACT

A systematic study had been carried out to get insight into the micellar behavior of anionic lipopeptide (LT) and nonionic sophorolipid (SL) in their different mass ratio mixed state using the technique of tensiometry. The models proposed by Clint, Rubingh and Gibbs et al. had been employed to interpret the formation of mixed micelles and found out synergism. The obtained experimental critical micelle concentrations (CMC) were lower than the ideal CMCs, indicating negative deviation from ideal behavior for all multi-component mixed micelles formation. A suited binary bio-surfactant mixing system was selected as the washing agents to treat the oily sludge produced from Huabei oilfield by the thermal bio-surfactant washing method. The results showed that in case of the mass ratios of 8:2 the CMC was dramatically decreased and synergism was the strongest in LT and SL bi mixed surfactant systems. The studied binary mixed bio-surfactant system showed higher washing efficiency for oily sludge than single surfactant system. In addition, the washing power of binary mixed bio-surfactants towards oily sludge was the best at below washing conditions: (a) the concentration of the mixed system (100 mg/L), (b) temperature (55 â„ƒ), (c) ratio of sludge/liquid (1:3), (d) washing time (3 h), and (e) stirring speed (300 rpm). Certainly, the washing abilities of the selected surfactants not only depend on their mixing ratio and washing conditions but also associate with microstructure and mineral components of oily sludge.

ABSTRACT

Nerve growth factor (NGF) is important for peripheral nerve regeneration. However, its short half-life and rapid diffusion in body fluids limit its clinical efficacy. Collagen has favorable biocompatibility and biodegradability, and weak immunogenicity. Because it possesses an NGF binding domain, we cross-linked heparin to collagen tubes to construct nerve guidance conduits for delivering NGF. The conduits were implanted to bridge a facial nerve defect in rats. Histological and functional analyses were performed to assess the effect of the nerve guidance conduit on facial nerve regeneration. Heparin enhanced the binding of NGF to collagen while retaining its bioactivity. Also, the nerve guidance conduit significantly promoted axonal growth and Schwan cell proliferation at 12 weeks after surgery. The nerve regeneration and functional recovery outcomes using the nerve guidance conduit were similar to those of autologous nerve grafting. Therefore, the nerve guidance conduit may promote safer nerve regeneration.

ABSTRACT

B7 homologue 6 (B7-H6), a newly identified member of the B7 costimulatory molecule family, is not only a crucial regulator of NK cell-mediated immune responses through binding to NKp30 but also has clinical implications due to its abnormal expression in human cancers. Here, we show that B7-H6 expression is abnormally upregulated in glioma tissue and that B7-H6 is coexpressed with stem cell marker Sox2. Intriguingly, B7-H6 was rarely detected on the surface of glioma cell lines but was abundantly expressed in glioma stem-like cells (GSLCs) that were derived from the glioma cell lines in vitro. Surprisingly, B7-H6 was the only one that was preferentially expressed in the GSLCs among the B7 family members. Functionally, knockdown of B7-H6 in GSLCs by siRNAs led to the inhibition of cell proliferation, with decrease in the expression of the oncogene Myc as well as inactivation of PI3K/Akt and ERK/MAPK signaling pathways. Moreover, we determined that three genes CBL (Casitas B-Lineage Lymphoma Proto-Oncogene), CCNT1 (Cyclin T1), and RNMT (RNA guanine-7 methyltransferase) were coexpressed with B7-H6 and c-myc in glioma tissue samples from the TCGA database and found, however that only RNMT expression was inhibited by the knockdown of B7-H6 expression in the GSLCs, suggesting the involvement of RNMT in the B7-H6/c-myc axis. Extending this to 293T cells, we observed that knocking out of B7-H6 with CRISPR-Cas9 system also suppressed cell proliferation. Thus, our findings suggest B7-H6 as a potential molecule for glioma stem cell targeted immunotherapy.

ABSTRACT

A preliminary study was conducted to examine the contamination of polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs) in surface sediments collected from the Liaohe River. The contamination levels of PCDD/Fs ranged from 317.3 to 509.3 pg/g dw. According to the calculated WHO-TEQ (Toxic Equivalent Quantity of World Health Organization), the range of PCDD/Fs is 13.87-40.88 pg/g dw. A principal component analysis and a cluster analysis suggested that PCDD/Fs in the sediments were mainly from gasoline/diesel vehicle emissions and solid waste incineration. The study revealed that the establishment of protected areas did not markedly reduced the ecological risk caused by PCDD/Fs in the Liaohe River protected areas sediments. It is necessary to study further the environmental impact of PCDD/Fs on the environment.

ABSTRACT

The aim of this study was to evaluate the toxicity of ammonia nitrogen, metals and organic contaminants in sediment collected from the Liaohe River Protected Areas. The TIE was applied to 16 samples. The zeolite, resin, and coconut charcoal were used to mask toxicity of the three kinds of pollutants, respectively. Then quantitative analyses together with a battery of bioassays were performed to evaluate toxic effects. At last, the spiking tests were used to confirm the major contributors to toxicity. The results of toxicity identification showed the ammonia nitrogen, γ-HCHs, As and Cd may cause toxic hazards to benthic organisms. The significant correlation between the survival and volume ratio of the sediment and overlying water confirmed ammonia nitrogen and Cd were the major toxic pollutants that cause the biological toxicity. We confirmed bioassays combined with masking agent, spiking tests and quantitative analyses were suitable tools for detecting toxicity.
