ABSTRACT

Results and performances of the QEXAFS double monochromator of the SAMBA beamline (Synchrotron SOLEIL) are presented. The device is capable of speeds of up to 40 Hz, while giving the user the possibility to choose the amplitude of the scan from 0.1° to 4° in a few seconds. The device is composed of two independent units and it is possible to perform scans alternating between two different crystals, literally jumping from low (4 keV) to high (37 keV) energies.

ABSTRACT

Recently, using an animal model of Charcot-Marie-Tooth human disorder, we showed that ascorbic acid (AA) represses PMP22 gene expression by acting on intracellular cAMP concentrations. In this work, we present kinetics data on the inhibitory effect of AA upon adenylate cyclase activity. The data show that this molecule acts as a competitive inhibitor of the enzyme, a finding that opens new pharmacological avenues.

ABSTRACT

INTRODUCTION: In the singing, which requires precise knowledge of the relevant musical code in use, accuracy of intonation plays a central role. Singing in tune requires to perceive pitch precisely and to memorize it before planning and executing the accurate vocal motion, which allows the exact emission of the correct pitch. OBJECTIVES: Our work investigated the role of short term memory of pitch on singing accuracy. MATERIAL AND METHOD: For that purpose, the experimental protocol of Deutsch (1970) was adapted for a perception and a production task. Participants were selected for their singing accuracy and separated into two groups of ten singing in tune and ten out-of-tune. All participants perceived pitch height exactly and were musically non-experts. For the perception and the production tasks, participants had to either compare or reproduce single pitches or two-pitch-sets. For the perception task, participants had to compare either single pitches or two-pitch patterns, all separated by a five seconds delay. For the production task, participants had to reproduce either single pitches or two-pitch patterns after a five seconds delay. The five seconds delay was either filled with intervening numbers, or with intervening tones, or without any disturbing sound. RESULTS: In perception and production task, the presence of intervening tones disturbs deeply the success of the subjects for every trial. Performance of the in-tune singing group is better for all the exercises while the other group had difficulties on single pitches and two-pitch patterns and was more disturbed by the effect of the intervening material. CONCLUSION: The outcome suggests that short term memory of pitch and accuracy of intonation would be closely linked. Further research needs to specify if that would mean that troubles in singing in tune are a consequence of a low-efficient short term memory of pitch, or if that troubles would hold up the right construction of the short term memory of pitch.

ABSTRACT

Time-resolved X-ray absorption spectroscopy (Quick-XANES) has been combined with UV-vis and Raman spectroscopies to study the in situ reduction of Ce4+ to Ce3+ in ethanolic solution with a time resolution of ca. 4-5 s. For this purpose, a cam-driven oscillating double-crystal monochromator with a channel-cut crystal was combined with two spectrometers for UV-vis and Raman spectroscopies in a specialized cell which allows one to fit the optical pathways for all three spectroscopies individually. The results show that high-quality results can be obtained simultaneously, thus giving a detailed insight into the mechanisms of the investigated chemical reaction. The continuous release of nitrate and ethanol ligands from the initial Ce4+ into the solution finally leads to a trivalent cerium species which is only coordinated with water molecules after about 1800 s of reaction time.

ABSTRACT

A new procedure, virofection, designed to stabilize the expression of transfected DNA has been developed. It exploits the capacity of retroviruses to integrate their genome into the chromosomes of host cells. The co-transfection of two plasmids, one carrying the genome of a defective retrovirus vector, the other one encoding all the retroviral proteins, results in a transient production of infectious virus particles. These particles can infect the neighboring cells and this leads to the stable integration of the vector genome. This procedure is time-saving and appears to be quite efficient. When applied to chicken embryonic fibroblasts cultured in vitro, it resulted in the stable expression of the lacZ gene in more than 30% of the cells, and did not induce chronic viremia. Stable lacZ expression was also achieved in chicken embryos in ovo. Virofection appears to be a promising and generally applicable method for implementing stable, safe and efficient gene transfer in vitro and in vivo.
